  • Acoustic comparison of voice use in solo and choir singing.

    • An experiment was carried out in which eight bass/baritone singers were recorded while singing in both choral and solo modes. Together with their own voice, they heard the sound of the rest of the choir and a piano accompaniment, respectively. The recordings were analyzed in several ways, including computation of long-time-average spectra for each passage, analysis of the sound levels in the frequency ranges corresponding to the fundamental and the "singer's formant," and a comparison of the sung levels with the levels heard by the singers. Matching pairs of vowels in the two modes were inverse filtered to determine the voice source spectra and formant frequencies for comparison. Differences in both phonation and articulation between the two modes were observed. Subjects generally sang with more power in the singer's formant region in the solo mode and with more power in the fundamental region in the choral mode. Most singers used a reduced frequency distance between the third and fifth formants for increasing the power in the singer's formant range, while the difference in the fundamental was mostly a voice source effect. In a choral singing mode, subjects usually adjusted their voice levels to the levels they heard from the other singers, whereas in a solo singing mode the level sung depended much less on the level of an accompaniment.

  • Efficient production of native, biologically active human cystatin C by Escherichia coli

    • A cDNA encoding the mature human cysteine proteinase inhibitor cystatin C was fused to the coding sequence for the Escherichia coli outer membrane protein A signal peptide, and the recombinant gene was expressed in E. coli under the control of the λ PR promoter, an optimized Shine-Dalgarno sequence and the λ cI 857 repressor. When induced at 42°C, such cells expressed large amounts of recombinant cystatin C. The recombinant protein was isolated in high yield and characterized. All physicochemical properties investigated, including the positions of disulfide bonds, indicated that the E. coli derived cystatin C was identical to cystatin C isolated from human biological fluids, except that the proline residue in position three was not hydroxylated. The recombinant protein displayed full biological activity against papain, cathepsin B and dipeptidyl peptidase I.

    • Recombinant cystatin C producing clones were isolated from a human placenta λgt11 cDNA library. The cDNA insert of one of the clones, containing 777 base pairs, encodes the complete mature cystatin C (120 amino acids) and a hydrophobic leader sequence of 26 amino acids, indicating an extracellular function of the inhibitor. The deduced protein sequence confirms the protein sequence of cystatin C isolated from human urine, but differs in one position from the sequence of the cystatin C fragment deposited as amyloid in hereditary cerebral hemorrhage with amyloidosis.

  • Radioimmunoassay of beta-microseminoprotein, a prostatic-secreted protein present in sera of both men and women

    • We describe a simple radioimmunoassay of beta-microseminoprotein, one of the three most abundant secretory proteins of the prostate gland. The detection limit of the assay is 1 microgram/L, and its precision, expressed as the total coefficient of variation, is less than 10% for values between 10 and 150 micrograms/L. Using this assay, we found that beta-microseminoprotein immunoreactivity was present in sera from both sexes at about the same concentration. The protein detected had the same molecular size on gel chromatography as the protein isolated from seminal plasma, and dilution curves for the sera paralleled that for the pure protein. The findings suggest that beta-microseminoprotein is present in serum of healthy subjects of both sexes and that it originates in tissue other than the prostate gland. The range of the serum concentration was 0-10.6 micrograms/L (median 4.1) for 51 healthy adult women and 1.1-14.7 micrograms/L (median 6.2) for 35 healthy adult men not older than 40 years. In males with prostatic cancer the concentration in serum was highly variable and often greatly increased. The concentration of beta-microseminoprotein was correlated with that of creatinine in serum, suggesting that the protein is eliminated--at least partly--from the circulation by glomerular filtration. Little of the protein was present in the urine of women. In urine from men the concentration was high and variable, probably because of local contribution from the prostate gland to the urethral urine.

    • We study three problems:PROBLEM A. Let P(D) be a matrix of partial differential operators with constant coefficient and U an open, convex and bounded set in Rn. Consider the system P(D)u = f where f lies in a Sobolev space on Rn and satisfies the compatibility conditions in U. We give conditions for the existence of a solution u which is, roughly speaking, more regular than f by the degree of P(D) in the (global) Sobolev space meaning.PROBLEM B. Let P(D) and U be as above and let K be a compact convex subset of U. Let u be a function in a quasi-analytic class on U\ K such that P(D)u = 0 there. We give conditions on P(D) which imply that every such u may by continued to U as a solution of the considered homogeneous system and in the same quasi-analytic class. The conditions are also necessary.PROBLEM C. Let u be a (ultra-) distribution with compact support on Rn. The condition for u to act surjectively on a class of (ultra-distributions is usually expressed as a condition on the decrease of the Fourier transform of u. We localize this condition for the Gevrey distributions. We also prove that some measures of Cantor type, in particular the tenary measure, do not act surjectively on the usual distributions on R1.

  • Light-in-flight recording. 5: Theory of slowing down the faster-than-light motion of the light shutter

    • Light-in-flight recording by holography uses a picosecond pulse for the reference beam, which like a sheet of light intersects the hologram plate and produces a sensitivity area that with a speed faster than light moves over the plate like a light shutter. If, however, the front of the reference pulse by diffraction in a grating is tilted relative to its direction of motion, the velocity of the light shutter can be slowed down resulting in increased recording time. The practical result using a reflection grating was a true recording that corresponded to a time compression of two to one. To minimize distortions of the recorded pulse shape we studied intersections that are identical for apparent (ellipsoidal) and true (spheroidal) wavefronts.
