- More than 30 patients with multiple sclerosis (MS) and Parkinson's disease (PD) have been reported so far. Theories on the co-occurrence of MS and PD range from coincidental to causal. There has been only one report of MS in young onset PD in a patient heterozygous for Parkin mutation. We report a patient with MS who developed signs typical for PD and was found to be heterozygous mutation carrier in the gene for glucocerebrosidase (GBA1), a well-known risk factor for PD.

- A new class of recessive Arabidopsis mutants, designated cristal (cii) has been isolated which display several abnormalities reminiscent of hyperhydric symptoms. These characteristics include translucent and wrinkled cotyledons and leaves, abnormal chloroplast organization, a reduced amount of chlorophyll, a reduced dry weight and a decreased number of palisade cells in the leaves accompanied by an increase of intercellular space, and therefore give a vitreous appearance to the aerial part. The phenotype is also dependent on the culture medium water potential. The cril gene was mapped on chromosome 4 close to the DHS1 marker.

Increased auxin efflux in the IAA-overproducing sur1 mutant of Arabidopsis thaliana: A mechanism for reducing auxin levels?

- With the aim of investigating the mechanisms that maintain auxin homeostasis in plants, we have monitored the net uptake and metabolism of exogenously supplied indole-3-acetic acid (IAA) and naphthalene-1-acetic acid (NAA) in seedlings of wild type and the IAA-overproducing mutant sur1 of Arabidopsis thaliana. Tritiated IAA and NAA entered the seedling tissues within minutes and were mostly accumulated as metabolites, probably amino acid and sugar conjugates, The mutant seedlings were marked by a strong increase of [H-3]IAA metabolism and a reduction of the accumulation levels of both free [H-3]IAA and [H-3]NAA. The same characteristics were observed in wild-type seedlings grown on 5 mu M picloram. We measured [H-3]NAA uptake in the presence of high concentrations of unlabeled NAA or the auxin efflux carrier inhibitor naphthylphthalamic acid (NPA). This abolished the difference in free [H-3]NAA accumulation between the mutant or picloram-treated seedlings and wild-type seedlings. These data indicated that active auxin efflux carriers were present in Arabidopsis seedling tissues. Picloram-treated seedlings and seedlings of the IAA-overproducing mutant sur1 displayed increased auxin efflux carrier activity as well as elevated conjugation of IAA. There is previous evidence to suggest that conjugation is a means to remove excess IAA in plant cells. Here, we discuss the possibility of efflux constituting an additional mechanism for regulating free IAA levels in the face of an excess auxin supply.

- A new auxin homeostasis gene in Arabidopsis called SUR2 has been identified. This gene, mapped to the bottom of chromosome 4, is defined by two recessive nuclear mutants designated superroot2 (sur2), which display several abnormalities reminiscent of auxin effects. A number of these characteristics are similar to the phenotype of the previously described auxin-overproducing mutant superroot1 (sur1); however, several lines of evidences reveal that the SUR2 gene defines a new key point in the regulation of endogenous auxin concentrations. The phenotype of the sur1 sur2 double mutant is additive. Analysis by gas chromatography coupled to mass spectrometry indicated increased levels of free indole-3-acetic acid correlated with a decreased level of bound auxin in the sur2 mutant. These results suggest that SUR2 may be involved in the control of auxin conjugation.

- Members of the tissue kallikrein-related peptidase (KLK) family not only regulate several important physiological functions, but aberrant expression has also been associated with various malignancies. Clinically, KLKs have been suggested as promising biomarkers for diagnosis and prognosis in many types of cancer. As of yet, expression of KLKs and their role in skin cancers are, however, poorly addressed. Malignant melanoma is an aggressive disease associated with poor prognosis. Hence, diagnostic biomarkers to monitor melanoma progression are needed. Herein, we demonstrate that although mRNA of several KLKs are aberrantly expressed in melanoma cell lines, only the KLK7 protein is highly secreted in vitro. In line with these findings, ectopic expression of KLK7 in human melanomas and its absence in benign nevi were demonstrated by immunohistochemistry in vivo. Interestingly, overexpression of KLK7 induced a significant reduction in melanoma cell proliferation and colony formation. Moreover, KLK7 overexpression triggered an increase in cell motility and invasion associated with decreased expression of E-cadherin and an upregulation of MCAM/CD146. Our results demonstrate, for the first time, that aberrant KLK7 expression leads to a switch from proliferative to invasive phenotype, suggesting a potential role of KLK7 in melanoma progression. Thus, we hypothesize that KLK7 may represent a potential biomarker for melanoma progression.

- Changes in urban design and traffic policy, demography, climate and associated adaptation, mitigation measures and environmental policies are likely to modify both outdoor and indoor air quality and therefore public health. The project aims to improve our understanding of future exposure situations and their impact on health, from an interdisciplinary approach. This will be achieved by using various state-of-the-art atmospheric models, measurements, epidemiological studies and reviews. To assess population full exposure, an integrated view accounting both for indoor and outdoor air pollution as well as for population time activity data will be developed. New dose-response functions will be estimated between health outcome, air pollution and temperature in order to better estimate the effects on the foetus and young children. Ultimately, scenarios of future urban climate and air quality will be simulated, combining future exposure scenarios, population scenarios and exposure-response functions to describe the effects of different trends and relevant policies on relative risk and burden of illness attributed to urban pollutants and their interactions with extreme temperatures. Also the mitigation strategies that can be used to reduce urbanization and climate change effects on the local urban meteorology and air quality will be assessed. With applications in several large European cities, the project will study the impact of several alternative adaptation scenarios on urban air quality and human health to a mid-century horizon (2030-2060) accounting for the effects of a changing urban climate. Scenario-based health impact assessments will combine exposure information from climate models, emission scenarios, policy evaluation studies and concentration calculations with exposure-response functions from epidemiological studies of vulnerable groups within the project and previously published functions for mortality and hospital admissions. The effects of socioeconomic and demographic trends will be discussed, the predicted health impacts and benefits associated with different interventions and policies and other urban changes will be described.

- We address a general system of complex Monge-Ampère equations on Fano horosymmetric manifolds and give necessary and sufficient conditions for existence of solutions in terms of combinatorial data of the manifold. This gives new results about Mabuchi metrics, twisted Kähler-Einstein metrics and coupled Kähler-Ricci solitons and provides a unified approach to many previous results on canonical metrics on Kähler manifolds.

SYZ and optimal transport atability of Weyl polytopes

- We prove optimal transport stability (in the sense of Andreasson and the second author) for reflexive Weyl polytopes: reflexive polytopes which are convex hulls of an orbit of a Weyl group. When the reflexive Weyl polytope is Delzant, it follows from work of Li, Andreasson, Hultgren, Jonsson, Mazzon, McCleerey, that the weak metric SYZ conjecture holds for the Dwork family in the corresponding toric Fano manifold. In particular, we show that the weak metric SYZ conjecture holds for centrally symmetric smooth Fano toric manifolds.
