  • Elevated fecal peptidase D at onset of colitis in Galphai2(-/-) mice, a mouse model of IBD

    • Background The identification of novel fecal biomarkers in inflammatory bowel disease (IBD) is hampered by the complexity of the human fecal proteome. On the other hand, in experimental mouse models there is probably less variation. We investigated the fecal protein content in mice to identify possible biomarkers and pathogenic mechanisms. Fecal samples were collected at onset of inflammation in Galphai2(-/-) mice, a well-described spontaneous model of chronic colitis, and from healthy littermates. The fecal proteome was analyzed by two-dimensional electrophoresis and quantitative mass spectrometry and results were then validated in a new cohort of mice. As a potential top marker of disease, peptidase D was found at a higher ratio in Galphai24mouse feces relative to controls (fold change 27; p = 0.019). Other proteins found to be enriched in Gai2(-/-) mice were mainly pancreatic proteases, and proteins from plasma and blood cells. A tendency of increased calprotectin, subunit S100-A8, was also observed (fold change 21; p = 0.058). Proteases are potential activators of inflammation in the gastrointestinal tract through their interaction with the proteinase-activated receptor 2 (PAR2). Accordingly, the level of PAR2 was found to be elevated in both the colon and the pancreas of Galphai24- mice at different stages of disease. These findings identify peptidase D, an ubiquitously expressed intracellular peptidase, as a potential novel marker of colitis. The elevated levels of fecal proteases may be involved in the pathogenesis of colitis and contribute to the clinical phenotype, possibly by activation of intestinal PAR2.

  • Gender dependent importance of IRAK-1 in dextran sulfate sodium induced colitis

    • Toll-like receptor (TLR) signaling is important for the induction of pro-inflammatory cytokines and interferon (IFN)-inducible genes in response to bacterial and viral challenge. Interleukin-1 receptor-associated kinase-1 (IRAK-1) is a signaling kinase situated downstream of the adapter protein myeloid differentiation factor 88 (MyD88) in the TLR intracellular signaling cascade and is required for normal signal transduction through this pathway. We investigated the importance of IRAK-1 in intestinal inflammation by using the dextran sulfate sodium (DSS)-colitis model. We show that IRAK-1 deficient mice are protected against systemic signs of inflammation, i.e., weight loss and spleen enlargement compared to wild-type controls irrespective of gender. However, IRAK-1(-/y) males but not IRAK-1(-/-) females display significant protection against colitis and thymic atrophy compared to wild-type mice. Our results indicate a gender specific effect of IRAK-1 in the DSS-induced colitis, an interesting finding since the Irak-1 gene is located on the X-chromosome and several inflammatory diseases have a gender dependent incidence.

  • Value conflicts and non-compliance: Attitudes to whistleblowing in Swedish organisations

Purpose: The purpose of the study is to explore potential value conflicts between information security work and whistleblowing activities by analysing attitudes to whistleblowing among white-collar workers in Swedish organisations. Design/methodology/approach: The study is conducted using survey data among (n = 674) Swedish white-collar workers. Statistical analyses are conducted to explore variations in acceptance of whistleblowing and analyse the relationship between acceptance for whistleblowing and information security attitudes and behaviours. Findings: The study finds strong support for whistleblowing in both public and private spheres, and by both private and public sector employees. The study also finds stronger acceptance for intra-organisational whistleblowing, while support for external whistleblowing is low. Finally, the study shows that the whistleblowing activities might be perceived as coming in conflict with information security work, even as the support for including whistleblowing functions in information security practices is high. Research limitations/implications: With a focus on one country, the study is limited in terms of empirical scope. It is also limited by a relatively small number of respondents and survey items relating to whistleblowing, which in turn affects its explanatory value. However, the study does provide unique new insight into a specific form of “non-compliance”, i.e. whistleblowing, which merits further investigation. Originality/value: Few studies exist that combine insights from the fields of whistleblowing and information security research. Thus, this study provides a basis for further investigation into attitudes and behaviours linked to whistleblowing in public and private organisations, as well as attendant value conflicts related to information security management and practice.

  • Clientelism and ethnic divisions in African countries

    • This article investigates the commonly assumed link between ethnic divisions and clientelism in African politics by examining the role of contextual ethnic divisions and specific ethnic affiliations in shaping attitudes towards clientelism. The empirical findings, drawing on quantitative data for 38,293 survey respondents across 25 African countries suggest important country heterogeneity, but also highlight some regularities. In particular, the ethnic composition of the population in the area of residence, rather than the individual's own ethnic affiliation, is important in shaping support for clientelism. Individuals living in a region where the majority of the population is the president's co-ethnics tend to be more supportive of clientelism, regardless of their own ethnic affiliation. The wide coverage of the results makes them especially interesting; while a number of studies suggest ethnically based targeted transfers in specific sectors or settings, this study explores the links between different forms of ethnic divisions and support for clientelism in a large multi-country African sample.

    • Background--Atrial fibrillation (AF) ablation improves patient-reported outcomes, irrespective of mode of intermittent rhythm monitoring. We evaluated the use of an AF-specific and a generic patient-reported outcomes instrument during continuous rhythm monitoring 2 years after AF ablation. Methods and Results--Fifty-four patients completed the generic 36-Item Short-Form Health Survey and the AF-specific AF6 questionnaires before and 6, 12, and 24 months after AF ablation. All patients underwent continuous ECG monitoring via an implantable loop recorder. The generic patient-reported outcomes scores were compared with those of a Swedish age- and sexmatched population. After ablation, both summary scores reached normative levels at 24 months, while role-physical and vitality remained lower than norms. Responders to ablation (AF burden < 0.5%) reached the norms in all individual 36-Item Short-Form Health Survey domains, while nonresponders (AF burden > 0.5%) reached norms only in social functioning and mental component summary. All AF6 items and the sum score showed moderate to large improvement in both responders and nonresponders, although responders showed significantly greater improvement in all items except item 1 from before to 24 months after ablation. Higher AF burden was independently associated with poorer physical component summary and AF6 sum score. Conclusions--The AF-specific AF6 questionnaire was more sensitive to changes related to AF burden than the generic 36-Item Short-Form Health Survey. Patients improved as documented by both instruments, but a higher AF burden after ablation was associated with poorer AF-specific patient-reported outcomes and poorer generic physical but not mental health. Our results support the use of an AF-specific instrument, alone or in combination with a generic instrument, to assess the effect of ablation. © 2018 The Authors.

  • Winter carbon dioxide effluxes from Arctic ecosystems: An overview and comparison of methodologies

    • The winter CO2 efflux from subnivean environments is an important component of annual C budgets in arctic ecosystems and consequently makes prediction and estimations of winter processes as well as incorporations of these processes into existing models important. Several methods have been used for estimating winter CO2 effluxes, involving different assumptions about the snow pack, all aiming to quantify CO2 production. Here, four different methods are compared and discussed: (1) measurements with a chamber on the snow surface, Fsnow; (2) chamber measurements directly on the soil, Fsoil, after snow removal; (3) diffusion measurements, F2-point, within the snow pack; and (4) a trace gas technique, FSF6, with multiple gas sampling within the snow pack. According to measurements collected from shallow and deep snow cover in High-Arctic Svalbard and Sub-Arctic Sweden during the winter of 2007-2008, the four methods differ by up to two orders of magnitude in their estimates of total winter emissions. The highest mean winter CO2 effluxes, 7.7-216.8 mg CO2 m-2 h-1, were observed using Fsoil and lowest values, 0.8-12.6 mg CO2 m-2 h-1, using FSF6. The Fsnow and F2-point methods were both within the lower range, 2.1-15.1 mg CO2 m-2 h-1 and 6.8-11.2 mg CO2 m-2 h-1, respectively. These differences are considered to be a result of contrasting methods, but also because the assumptions within the methods are not the same when quantifying CO2 production and effluxes to the atmosphere. Since snow can act as a barrier to CO2, Fsoil is assumed to measure soil production, whereas FSF6, Fsnow and F2-point are considered better approaches for quantifying exchange processes between the soil, snow, and the atmosphere. This study indicates that estimates of winter CO2 emissions may vary more as a result of the method used than due to the actual variation in soil CO2 production or release. This is a major concern, especially when CO2 efflux data are used in climate models or in carbon budget calculations, thus highlighting the need for further development and validation of accurate and appropriate techniques.

    • The aim of this study was to explore the oral health in Swedish individuals with the diagnosis of homozygote cystathionine beta synthase-deficient homocystinuria (HC), a rare disorder of amino acid metabolism affecting connective tissue, in which the phenotypic abnormalities include dislocation of the optic lens, skeletal abnormalities, thromboembolic events, and sometimes mental retardation. Further aims were to evaluate the oral findings against previous oral observations in a medical case report, such as high narrow palate, mandibular prognathia, crowding and early eruption of teeth. Every hospital in Sweden was contacted, with the inquiry of patients with diagnosis of HC,which resulted in 14 individuals participating in oral clinical examination. The oral findings evaluated against previous medical case reports showed to be partly in accordance with previous observations. Dental health showed to be compromised in a majority of cases. Together with the fact that methionine restriction (low-protein diet) is involved in the treatment of the condition and might result in a diet high in sugars, this points out the role of regular dental checkups and preventive oral care for individuals suffering from HC. In addition, short dental roots were a finding not previously reported in the literature. All the studied cases had central maxillary incisors with short roots, when compared to reference values used.

    • OBJECTIVES: In the light microscope compare the amount of bone saucerization for non-threaded cylindrical and threaded implant designs in retrieved samples from patients. MATERIAL AND METHOD: Consecutively received retrieved oral implants from 117 patients, whereof 85 non-threaded cylindrical and 85 Branemark implants, have been included in the study. For 75 non-threaded cylindrical and 46 Branemark implants was the entire implant length available for calculation. Undecalcified ground sections were investigated in the light microscope with calculation of percentage of implant length coronal to the first bone-implant contact and percentage of bone to implant contact. RESULTS: Mean value for implant length coronal to first bone-implant contact was 65%, standard error of the mean (SEM) 3 (range 0-100%), for non-threaded cylindrical implants and 43%, SEM 6 (range 0-100%) for Branemark implants. Mean values of bone contact along the entire implant length was 23%, SEM 2 (range 0-65%), for the non-threaded cylindrical implants and 33%, SEM 5 (range 0-93%) for the Branemark implants. CONCLUSION: Within the limitations of this retrospective, retrieval study non-threaded cylindrical implants demonstrated a greater bone saucerization when evaluated in the light microscope.

    • The aim of this report was to describe the bone tissue response to Branemark oral implants retrieved from patients. The material consisted of consecutively received Branemark threaded oral implants and related patient data provided by clinicians. The implant samples were processed into undecalcified sections for evaluation under the light microscope. The analysis demonstrated a lower percentage of bone-to-implant contact for the unloaded implants as compared to the loaded implants. When the threads were divided into four different regions, the loaded implants had a lower percentage of bone-contacting length at the thread top as compared to the other three regions.
