    • Inflammation and impaired mitochondrial oxidative phosphorylation are considered key players in the development of several metabolic disorders, including diabetes. We have previously shown inflammation and mitochondrial dysfunction in the hypothalamus of an animal model for anorexia, the anx/ anx mouse. Moreover, increased incidence of eating disorders, e.g., anorexia nervosa, has been observed in diabetic individuals. In the present investigation we evaluated whether impaired mitochondrial phosphorylation and inflammation also occur in endocrine pancreas of anorectic mice, and if glucose homeostasis is disturbed. We show that anx/ anx mice exhibit marked glucose intolerance associated with reduced insulin release following an intraperitoneal injection of glucose. In contrast, insulin release from isolated anx/ anx islets is increased after stimulation with glucose or KCl. In isolated anx/ anx islets there is a strong downregulation of the mitochondrial complex I (CI) assembly factor, NADH dehydrogenase (ubiquinone) 1α subcomplex, assembly factor 1 ( Ndufaf1), and a reduced CI activity. In addition, we show elevated concentrations of free fatty acids (FFAs) in anx/ anx serum and increased macrophage infiltration (indicative of inflammation) in anx/ anx islets. However, isolated islets from anx/ anx mice cultured in the absence of FFAs do not exhibit increased inflammation. We conclude that the phenotype of the endocrine pancreas of the anx/ anx mouse is characterized by increased levels of circulating FFAs, as well as inflammation, which can inhibit insulin secretion in vivo. The anx/ anx mouse may represent a useful tool for studying molecular mechanisms underlying the association between diabetes and eating disorders.

    • We present first results on heteroepitaxy of InP on silicon on insulator (SOI). We also demonstrate InP nanopillar fabrication by means of selective epitaxy. Selective epitaxy is also exploited to fabricate advanced photonic integrated devices for Optical Code Division Multiplex Access (OCDMA) networking applications.

    • During the meetings of the theory and modelling working group, within the MEL-ARI (Microelectronics Advanced Research Initiative) and NID-FET (Nanotechnology Information Devices-Future and Emerging Technologies) initiatives of the European Commission, we have been discussing the current status and the future perspectives of nanoscale device modelling. The outcome of such a discussion is summarized in the present paper, outlining the major challenges for the future, such as the integration of nonequilibrium phenomena and of molecular-scale properties. We believe that modelling has a growing importance in the development of nanoelectronic devices and must therefore make a move from physics to engineering, providing valid design tools, with quantitative predictive capabilities.

  • Contamination and correlation with toxicity of sediment samples from the Skiagerrak and Kattegat

    • The pollution state in the Skagerrak and Kattegat was investigated by determination of pollutant concentrations and toxicity of sediment samples from 11 stations in the area. A comparison was made with the sediment from a reference site near the Faroe Islands. Polycyclic aromatic hydrocarbons (PAH) and organochlorines were determined in whole sediment and heavy metals and ammonia were analysed in filtered pore water. Toxicity was bioassayed in whole sediment with Nitocra spinipes and Daphnia magna, in pore water with Mytilus edulis larvae and in solvent extracts from sediment with tests measuring etoxyresorufin-O-deethylase (EROD) activity in Oncorhyncus mykiss and rate of denitrification. Sites close to Goteborg and in an area from the Oslo fjord to the Norwegian Trench were most polluted. Sediment from the Faroe Islands was least polluted and also least toxic. Multivariate statistical analysis indicates that the different tests were sensitive to different kinds of pollutants. Effects on mussel larvae correlated strongest with the occurrence of ammonia, manganese, cadmium and PAHs, Nitocra with alpha-hexachlorocyclohexane (HCH) and p,p'-DDD, Daphnia with arsenic and gamma-HCH, fish EROD activity with benzo[ghi]perylene and unknown compounds associated with organic carbon, and denitrification with chlordanes, dieldrin and a few PAHs. The results indicate that sampling sites close to Goteborg are so polluted that harmful effects on the ecosystem probably occur.
