- In 80 patients with polycythaemia vera (PV) a total of 108 venous blood samples were obtained and analysed for EDTA-plasma erythropoietin (EPO) concentration. At the time of study 21 of the PV patients were newly diagnosed and had prior to blood sampling neither received phlebotomy treatment nor therapy with myelosuppressive agents; these subjects had a mean plasma EPO concentration of 0.5+/-0.9 IU/L. Thirty-seven patients treated with phlebotomy only had a mean plasma EPO concentration of 2.5+/-2.9 IU/L. The mean plasma EPO concentrations for 26 patients treated with hydroxyurea, 13 patients treated with radiophosphorous and 11 patients given a combination of myelosuppressive agents were 8.9+/-8.0, 10.9+/-12.6 and 7.2+/-7.4 IU/L, respectively. Untreated patients and patients on phlebotomy only had significantly lower values for plasma EPO than patients on therapy with myelosuppressive drugs. This finding persisted also after a correction for differences in haemoglobin levels had been introduced. Thereby, the present results would suggest a difference in the EPO feedback system in untreated and phlebotomised PV patients compared to PV patients treated with myelosuppressive agents.

Selection of prey size and prey species by 1-group cod Gadus morhua: effects of satiation level and prey handling times

- We performed laboratory experiments to investigate feeding behaviour of 1-group Atlantic cod Gadus morhua and compared the results with stomach content records from wild 1-group cod feeding within a shallow bay on the west coast of Sweden. On the basis of the field observations, 3 prey species were investigated: juvenile shore crabs Carcinus maenas, brown shrimps Crangon crangon and 0-group plaice Pleuronectes platessa. When fed single species meals in the laboratory, satiation level was mostly independent of prey size, but cod consumed more plaice than shrimps and more shrimps than crabs. Once satiated on crabs, cod ate plaice if they were subsequently offered, but the reverse was not true. The time taken to ingest crabs was independent of the prey:cod (P:C) length ratio, cod length or stomach fullness, whereas plaice and shrimp ingestion times increased with P:C length ratio, and shrimp times also increased with stomach fullness. Consequently, the profitability of crabs increased up until the maximum edible size, whereas shrimp and plaice profitability peaked at P:C length ratios lower than the maximum edible sizes. For a given prey species, size selection in the field correlated closely with the size-dependent profitability relationships. Species selection had no apparent dependence upon handling-time profitability(, species-dependent satiation level or gastric evacuation rate. Additional factors of probable importance include prey evasiveness, prey abundance, habitat patchiness and abiotic factors such as prevailing light conditions.

- We prove a posteriori error estimates for a finite element method for steady-state, energy dependent, Fokker-Planck and Fermi pencil beam equations in two space dimensions and with a {\sl forward-peaked scattering} (i.e., with velocities varying within the right unit semi-circle). Our estimates are based on a {\sl transversal symmetry assumption}, together with a strong stability estimate for an associated dual problem combined with the Galerkin orthogonality of the finite element method.

- The Herpes simplex virus type I origin binding protein (OBP) is a sequence-specific DNA-binding protein and a dimeric DNA helicase encoded by the UL9 gene. It is required for the activation of the viral origin of DNA replication oriS. Here we demonstrate that the linear double-stranded form of oriS can be converted by heat treatment to a stable novel conformation referred to as oriS*. Studies using S1 nuclease suggest that oriS* consists of a central hairpin with an AT-rich sequence in the loop. Single-stranded oligonucleotides corresponding to the upper strand of oriS can adopt the same structure. OBP forms a stable complex with oriS*. We have identified structural features of oriS* recognized by OBP. The central oriS palindrome as well as sequences at the 5' side of the oriS palindrome were required for complex formation. Importantly, we found that mutations that have been shown to reduce oriS-dependent DNA replication also reduce the formation of the OBP-oriS* complex. We suggest that oriS* serves as an intermediate in the initiation of DNA replication providing the initiator protein with structural information for a selective and efficient assembly of the viral replication machinery.

- The role of two important tumour suppressor genes, p16 and p53, was evaluated in cartilaginous tumour tissues. Genomic DNA from 22 chondrosarcomas, 5 benign chondroid tumours, 1 sample of reactive proliferative cartilage and 2 samples of normal cartilage were analysed using polymerase chain reaction, single strand conformational polymorphism, DNA sequencing and methylation-specific polymerase chain reaction. The p16 gene was found to be partly methylated in 5 high-grade chondrosarcomas and homozygously deleted in 1 chondrosarcoma. Moreover, a polymorphism was detected in 3 malignant tumours, but not in benign tumours or normal cartilage. Analysis of the p53 gene revealed an unchanged structure in all samples. These findings show a role for p16, but not p53, in chondrosarcoma.

Price adjustments by a gasoline retail chain

- We use daily data to examine price responses in the Swedish gasoline market to changes in the Rotterdam spot price, exchange rates and taxes. The distribution of price adjustments by a leading retail chain, for the period January 1980 to December 1996, is symmetric with no small adjustments. An error correction model shows that, in the short run, prices gradually move towards the long-run equilibrium in response to cost shocks. There is some evidence that, also in the short run, prices are stickier downwards than upwards. Prices respond more rapidly to exchange rate movements than to the spot market price. Our analysis emphasizes that to fully understand price adjustments it is necessary to examine data sets where the sample frequency at least matches that of price adjustments.
