    • An important part of a society’s resilience is how prepared it is to cope with the changing conditions during the alpha and omega phases according to resilience theory. Lars Thofelt, an academic from the mid Sweden region, early recognized this need for students to develop skills needed for a societal change, and devoted his life to pedagogy suitable for this. The main outcome of his achievement was the interdisciplinary university program in Ecotechnics/Ecotechnology (Ekoteknik in Swedish), at Mid Sweden University. Ecology, economy and technology in cooperation for sustainable development were the original approach, and still are.Thofelt’s ideas had a main focus of helping students develop their inherent capabilities of solving problems and overcome obstacles. After Thofelt’s 12 years at the program his ideas were carried on by former colleagues and students, and the teaching further developed with a mix of the Thofelt tradition and other experiences brought in by new employees. This paper describes this interdisciplinary teaching approach with special focus on development of resilience capacity in students.It was concluded that 1) a key element to develop resilience skills in students is to push them to a self-propelled learning behavior rather than traditional teaching, 2) not too easily provide the students with answers will develop their problem solving skills, 3) doing-before-reading teaching is more time consuming but seem to give deeper knowledge, 4) interdisciplinary teaching will in the long run benefit from having the interdisciplinary team within the department, rather than as a conglomerate of several departments.

  • Subcrestal placement of dental implants with an internal conical connection of 0.5 mm versus 1.5 mm: Outcome of a multicentre randomised controlled trial 1 year after loading

    • Purpose: To evaluate whether there are some clinical benefits by placing single dental implants either 0.5 or 1.5 mm subcrestally in healed bone crests. Materials and methods: Sixty partially edentulous patients requiring two single implant-supported crowns had both sites randomly allocated either to 0.5 mm or 1.5 mm subcrestal implant placement according to a split-mouth design at six centres. Implants were submerged in aesthetic areas or nonsubmerged in non-aesthetic areas for 3 months. Provisional acrylic crowns were delivered and were replaced after 2 months by definitive metal-ceramic crowns. Patients were followed to 1 year after loading. Outcome measures were: crown and implant failures; complications; aesthetics assessed using the pink esthetic score (PES); peri-implant marginal bone level changes; and patient preference, recorded by blinded assessors. Results: One patient dropped out. One patient lost both implants to infection at impression taking. Three complications affected three patients of the 0.5 mm group and two complications affected two patients of the 1.5 mm subcrestally placed implants. One patient had complications at both implants. There were no statistically significant differences for complications between group (difference of proportion = 0.02; 95% Cl-0.06 to 0.09; P (McNemar test) = 1.000). At delivery of definitive crowns, 2 months after loading, the mean aesthetic score was 11.22 +/- 1.91 and 11.12 +/- 1.59 for the 0.5 and 1.5 mm group, respectively. At 1 year after loading, the mean aesthetic score was 12.09 +/- 1.66 and 12.10 +/- 1.52 for the 0.5 and 1.5 mm group, respectively. There were no statistically significant differences between the two groups at 2 months (P (paired t test) = 0.626) or at 1 year (P (paired t test) = 0.920). One year after loading, patients of the 0.5 mm lost on average 0.21 +/- 0.51 mm and those of the 1.5 mm group 0.11 +/- 0.36 mm, the difference being not statistically significant (difference = 0.10; 95% Cl-0.01 to 0.20; P (paired t test) = 0.078). Patients did not prefer any depth of the implant placement over the other. There were no differences in outcomes between centres. Conclusions: No statistical or clinical differences were noticed when placing implants 0.5 mm or 1.5 mm subcrestally therefore clinicians can do as they prefer.

  • Synthesis and characterization of zeolite films and membranes

    • In this work, a seeding technique was used to synthesize films and membranes of FAU, LTA and MFI type zeolites. In the first part, hydrothermal growth was performed without organic template molecules, which resulted in template-free zeolite films and membranes. The samples were characterized by Scanning Electron Microscopy, X-ray Powder Diffraction and permeation measurements with gaseous probe molecules. Thin films of FAU-type zeolite were prepared on polished single crystals. The thickness and morphology of the films could be controlled by varying the synthesis conditions. Preparation of LTA-type membranes was also attempted. However, the membranes cracked during drying at temperatures above room temperature. Template free MFI membranes with higher quality could be prepared. These membranes had a maximum separation factor alpha of 17.8 at 220°C for a n-butane/i-butane mixture. Cracks formed at temperatures higher than 250°C. Crack formation in zeolite membranes at high temperatures has also been reported by several other groups. Since no model for the crack formation process has been established in the literature, the second part of this work was devoted to study crack formation and to develop a model. Relatively thick (ca 1800 nm) alpha-alumina supported MFI films, prepared using organic template molecules (TPA+), were selected for the study since data on crack formation in the form of SEM images and permeation measurements for these membranes had been obtained in earlier work by the group. These membranes were further studied by in-situ High Temperature X-ray Powder Diffraction experiments in the present work. In addition, MFI powder and a blank alpha-alumina support were also investigated. Data were collected with the aid of a Synchrotron radiation facility as well as with a conventional laboratory instrument for the temperature cycle 25-500-25°C. The Rietveld method was used to determine the unit cell parameters of MFI and alpha-alumina as well as the TPA+ occupancy of MFI. The out-of- plane strain (i.e. strain in the direction perpendicular to the film surface) in the film and the support was calculated. In addition, the microstructure of the support was investigated by pattern decomposition and Williamson-Hall plots. In agreement with previous reports in the literature, it was found that the TPA-MFI structure contracts as a consequence of template removal and possibly also a structure intrinsic mechanism and the alpha-alumina support expands. Hence, a large thermal expansion mismatch occurs in the membranes during heating. An overall out- of-plane compressive strain was observed for the MFI film during heating, which indicates an in-plane tensile stress (i.e. in the direction parallel to the film surface) in the film. This result was explained by the larger expansion of the support, compared to the film. The alpha-alumina support was also found to be under an overall out-of-plane compressive strain at non-ambient temperatures, presumably due to zeolite in the pores of the support. The microstrain for the MFI coated alpha-alumina support increased during heating, and remained during cooling, which indicate the formation of structural defects in the support. Based on these results and results from earlier work, a model for crack formation was proposed: In the thick films (ca 1800 nm) studied in the present work, the crystals are well intergrown. During heating, the MFI crystals contracts and the alpha- alumina support expands. Consequently, a thermal stress develops in the composite which eventually leads to formation of cracks. In addition, part of the stress is also released via formation of structural defects in the alpha-alumina support. In thinner films (ca 500 nm), the crystals are less well intergrown and the thermal expansion mismatch between the crystals in the film and the support leads to opening of grain boundaries in the film rather than cracks.

    • Recently, activating mutations of the full length ALK receptor, with two hot spots at positions F1174 and R1275, have been characterized in sporadic cases of neuroblastoma. Here, we report similar basal patterns of ALK phosphorylation between the neuroblastoma IMR-32 cell line, which expresses only the wild-type receptor (ALK(WT)), and the SH-SY5Y cell line, which exhibits a heterozygous ALK F1174L mutation and expresses both ALK(WT) and ALK(F1174L) receptors. We demonstrate that this lack of detectable increased phosphorylation in SH-SY5Y cells is a result of intracellular retention and proteasomal degradation of the mutated receptor. As a consequence, in SH-SY5Y cells, plasma membrane appears strongly enriched for ALK(WT) whereas both ALK(WT) and ALK(F1174L) were present in intracellular compartments. We further explored ALK receptor trafficking by investigating the effect of agonist and antagonist mAb (monoclonal antibodies) on ALK internalization and down-regulation, either in SH-SY5Y cells or in cells expressing only ALK(WT). We observe that treatment with agonist mAbs resulted in ALK internalization and lysosomal targeting for receptor degradation. In contrast, antagonist mAb induced ALK internalization and recycling to the plasma membrane. Importantly, we correlate this differential trafficking of ALK in response to mAb with the recruitment of the ubiquitin ligase Cbl and ALK ubiquitylation only after agonist stimulation. This study provides novel insights into the mechanisms regulating ALK trafficking and degradation, showing that various ALK receptor pools are regulated by proteasome or lysosome pathways according to their intracellular localization.
