  • Delivering needle-free insulin using AERx (R) iDMS (Insulin Diabetes Management System) technology

    • Background and Methods: Inhaled insulin has recently emerged as an alternative to subcutaneous insulin administration. One inhaled insulin device, AERx (R) (a registered trademark of Aradigm Corp., Hayward, CA, or its affiliates in the United States and other countries) insulin diabetes management system (iDMS) (Novo Nordisk A/S, Bagsvoerd, Denmark), uses a unique liquid human insulin strip to deliver an aerosol of insulin to the lungs. AERx iDMS enables 1-unit increment dosing, and the device ensures that the insulin dose is released at the optimal point of inhalation for delivery to the lungs. Results: Compared with subcutaneous human insulin, the pharmacokinetic and pharmacodynamic profile of inhaled insulin with AERx iDMS is similar, but with a more rapid onset of action. Data from these pharmacokinetic studies have also demonstrated that inhaled insulin dosing with AERx iDMS is as consistent and reproducible as subcutaneous human insulin. Conclusions: In individuals with diabetes prandial inhaled insulin with AERx iDMS is as effective and well tolerated as subcutaneous prandial human insulin or insulin aspart in terms of glycemic control and overall hypoglycemia. No major safety concerns have been raised with respect to pulmonary function tests. Other clinical studies using AERx iDMS in special populations, such as smokers, people with asthma, or people suffering from upper respiratory tract infections, have provided important information regarding the use of inhaled insulin in these circumstances. Overall, pulmonary insulin delivery with the AERx iDMS device appears to be a promising safe and efficacious alternative to subcutaneous insulin injections.

  • Genome survey sequencing of the wine spoilage yeast Dekkera (Brettanomyces) bruxellensis

    • The hemiascomycete yeast Dekkera bruxellensis, also known as Brettanomyces bruxellensis, is a major cause of wine spoilage worldwide. Wines infected with D. bruxellensis develop distinctive, unpleasant aromas due to volatile phenols produced by this species, which is highly ethanol tolerant and facultatively anaerobic. Despite its importance, however, D. bruxellensis has been poorly genetically characterized until now. We performed genome survey sequencing of a wine strain of D. bruxellensis to obtain 0.4x coverage of the genome. We identified approximately 3,000 genes, whose products averaged 49% amino acid identity to their Saccharomyces cerevisiae orthologs, with similar intron contents. Maximum likelihood phylogenetic analyses suggest that the relationship between D. brurellensis, S. cerevisiae, and Candida albicans is close to a trichotomy. The estimated rate of chromosomal rearrangement in D. bruxellensis is slower than that calculated for C albicans, while its rate of amino acid evolution is somewhat higher. The proteome of D. bruxellensis is enriched for transporters and genes involved in nitrogen and lipid metabolism, among other functions, which may reflect adaptations to its low-nutrient, high-ethanol niche. We also identified an adenyl deaminase gene that has high similarity to a gene in bacteria of the Burkholderia cepacia species complex and appears to be the result of horizontal gene transfer. These data provide a resource for further analyses of the population genetics and evolution of D. bruxellensis and of the genetic bases of its physiological capabilities.

  • Domain-wall contribution to magnetoresistance of a ferromagnetic (Ga,Mn)As layer

    • Low-temperature charge-carrier transport in simple magnetoresistive nanodevices, consisted of narrow constrictions of submicron width in the epitaxial layer of a ferromagnetic (Ga,Mn)As semiconductor, has been investigated and correlated with magnetic properties of the layer. The devices containing constrictions; revealed abrupt jumps of a reduced resistance that appeared when the sweeping magnetic field crossed the regions of the coercive field of the layer magnetization. In contrast, the non-constricted reference device displayed abrupt jumps of an enhanced resistance at the same values of magnetic field. We interpret the both features, whose positions on the magnetic-field scale reflect the hysteresis loop of magnetization, as manifestation of domain wall contribution to the (Ga,Mn)As layer resistance. The negative contribution of a domain wall to the resistance in the constricted device results most likely from the suppression of the weak localization effects by a domain wall located at the constriction.

  • Linking agricultural policies to population trends of Swedish farmland birds in different agricultural regions

    • 1. The widespread declines of farmland birds have generally been linked to agricultural intensification. We tested the hypotheses that (i) changes in agricultural policy, through its effects on agricultural intensification and (ii) regional differences in agricultural intensification affect temporal and spatial population trends of farmland birds in Sweden. 2. We analysed regional bird population trends (1976–2003) for seven common farmland bird species: the migratory lapwing Vanellus vanellus, skylark Alauda arvensis, starling Sturnus vulgaris and linnet Carduelis cannabina and the resident tree sparrow Passer montanus, house sparrow P. domesticus and yellowhammer Emberiza citrinella. We identified three periods of agricultural policy in Sweden between 1976 and 2003: the intensification period (i.e. 1976–87; promoting increased production), the set-aside period (1987–95; promoting extensification of farming) and the Common Agricultural Policy (CAP) period (1995–2003; promoting increased production). Population trends were compared between three types of Swedish farmlands: open plains (intensive farming with a marked intensification), mosaic farmlands (i.e. farmland-dominated forest mosaics, less intensive farming, but show moderate intensification) and forest regions (i.e. forest-dominated farmlands with low intensity farming and extensification/abandonment). 3. The four migrants displayed clear significant trend switches between the policy periods, with declines in the ‘intensification period’ and the ‘CAP period’ and less negative or even positive population trends in the ‘set-aside period’. The population trends of the three resident species showed no clear pattern in relation to agricultural policy periods. 4. All species except tree sparrow displayed significantly different population trends between farmland regions. Four species (lapwing, skylark, linnet and house sparrow) declined most in the open plains and the forest regions, whereas two species (starling and yellowhammer) declined most in the mosaic farmlands. 5. Synthesis and applications. Large-scale changes in agriculture policy have a strong potential to change the present poor state of farmland biodiversity as shown by the generally positive population trends in the ‘set-aside period’. It also suggests extensification to be beneficial to farmland birds. However, in regions of low profitability and an already ongoing extensification, a further extensification will lead to loss of both farmland habitat and bird diversity. In such regions mixed farming needs to be retained and hence should be supported.

  • Comfort and Safety as Perceived by Wheelchair-Seated Bus Passengers

    • Wheelchair-seated passengers on European and Swedish urban transit buses can travel rearwards, resting against a bulkhead, without the use of tiedown equipment. However, users sometimes disapprove of being the only passengers who have to travel rearwards, and also unrestrained. In order to investigate comfort and safety as perceived by wheelchair-seated bus passengers riding either forwards or rearwards, an experimental field study was carried out. Participating subjects were 31 wheelchair users and a group of 44 ambulant passengers. Each subject made two consecutive, identical 15-minute trips, randomly beginning either forwards or backwards, and interviews were conducted after each trip. During the trips, three-axis bus motion was recorded. The concept of categorization was used as a theoretical background and basis for discussion of the comfort and safety constructs. The results indicated that while the majority preferred the forward-facing position regarding comfort issues, a minority felt that the level of perceived safety was better. Wheelchair-seated passengers were more positive towards the rearward position than the ambulant group.
