    • The objective of this study was to predict voluntary intake by cattle of grass silages of different fermentation quality from in vitro gas production data. Further, biological concepts of intake related gas data were examined by cluster analysis to silage chemical composition. Samples from 24 grass silages were characterized from their gas profiles. Observed silage dry matter intake was modelled by partial least squares regression analysis. Cumulative gas volumes and rates of gas production during early incubation, asymptotic gas volume as well as discrete lag time, and cumulative gas volumes at 48 and 72 h were the best descriptors of the variation in silage intake. The discrete lag time had a strong negative influence on intake and made up clusters with chemical variables indicating secondary fermentation. In vitro gas production data did not improve silage dry matter intake predictions compared with current models based on chemical analysis of silage fermentation quality.

    • Many scientific applications deal with data from a multitude of different sources, e.g., measurements, imaging and simulations. Each source provides an additional perspective on the phenomenon of interest, but also comes with specific limitations, e.g. regarding accuracy, spatial and temporal availability. Effectively combining and analyzing such multimodal and partially incomplete data of limited accuracy in an integrated way is challenging. In this work, we outline an approach for an integrated analysis and visualization of the atmospheric impact of volcano eruptions. The data sets comprise observation and imaging data from satellites as well as results from numerical particle simulations. To analyze the clouds from the volcano eruption in the spatiotemporal domain we apply topological methods. We show that topology-related extremal structures of the data support clustering and comparison. We further discuss the robustness of those methods with respect to different properties of the data and different parameter setups. Finally we outline open challenges for the effective integrated visualization using topological methods.

  • Odin observation of water in the IRAS 16293-2422 low-mass star forming region

    • We present the Odin satellite submillimeter measurement of the ground state transition of ortho-water at 557 GHz around the low-mass protostar IRAS16293-2422. The three following regions have been probed: the on-source position, and the two regions of interaction between the molecular outflows and the dense ambient cloud, i .e. on the SiO peak E1, and near the deuterated peak (cf Lis et al.(2002)). The three spectra reveal broad emission, with a narrow and deep absorption feature at the velocity of the quiescent gas. The water line profiles and intensities have been compared and combined with the CO(3- 2) and HCO+ lines from Lis et al.(2002) after convolution over the Odin beam. The contribution from the central envelope on the water line emission has been estimated with the infall model of Shoier et al. 2002. We show that the water emission is dominated by the outflows, particularly in the red wing of the lines. Using a c-shock modelling (Cabrit et al. 2005), we derive an abundance of X(H2O) 1 - 3 × 10-5 of H2, and shock velocities of 12 - 15 km/s. We also show that the absorption feature is due to the cold outer halo and corresponds to a lower limit of a few 10-9 on the H2O abundance.

  • Herders of Indian and European Cattle Share Their Predominant Allele for Lactase Persistence

    • Milk consumption and lactose digestion after weaning are exclusively human traits made possible by the continued production of the enzyme lactase in adulthood. Multiple independent mutations in a 100-bp region-part of an enhancer-approximately 14-kb upstream of the LCT gene are associated with this trait in Europeans and pastoralists from Saudi Arabia and Africa. However, a single mutation of purported western Eurasian origin accounts for much of observed lactase persistence outside Africa. Given the high levels of present-day milk consumption in India, together with archaeological and genetic evidence for the independent domestication of cattle in the Indus valley roughly 7,000 years ago, we sought to determine whether lactase persistence has evolved independently in the subcontinent. Here, we present the results of the first comprehensive survey of the LCT enhancer region in south Asia. Having genotyped 2,284 DNA samples from across the Indian subcontinent, we find that the previously described west Eurasian -13910 C>T mutation accounts for nearly all the genetic variation we observed in the 400- to 700-bp LCT regulatory region that we sequenced. Geography is a significant predictor of -13910*T allele frequency, and consistent with other genomic loci, its distribution in India follows a general northwest to southeast declining pattern, although frequencies among certain neighboring populations vary substantially. We confirm that the mutation is identical by descent to the European allele and is associated with the same >1 Mb extended haplotype in both populations.

    • Identification and evaluation of factors important for thermostability in proteins is a growing research field with many industrial applications. This study investigates the effects of introducing a novel disulfide bond and engineered electrostatic interactions with respect to the thermostability of holo azurin from Pseudomonas aeruginosa. Four mutants were selected on the basis of rational design and novel temperature-dependent atomic displacement factors from crystal data collected at elevated temperatures. The atomic displacement parameters describe the molecular movement at higher temperatures. The thermostability was evaluated by optical spectroscopy as well as by differential scanning calorimetry. Although azurin has a high inherent stability, the introduction of a novel disulfide bond connecting a flexible loop with small alpha-helix (D62C/K74C copper-containing mutant), increased the T(m) by 3.7 degrees C compared with the holo protein. Furthermore, three mutants were designed to introduce electrostatic interactions, K24R, D23E/K128R, and D23E/K128R/K24R. Mutant K24R stabilizes loops between two separate beta-strands and D23E/K128R was selected to stabilize the C-terminus of azurin. Furthermore, D23E/K128R/K24R was selected to reflect the combination of the electrostatic interactions in D23E/K128R and K24R. The mutants involving electrostatic interactions had a minor effect on the thermostability. The crystal structures of the copper-containing mutants D62C/K74C and K24R have been determined to 1.5 and 1.8 A resolution. In addition the crystal structure of the zinc-loaded mutant D62C/K74C has also been completed to 1.8 A resolution. These structures support the selected design and provide valuable information for evaluating effects of the modifications on the thermostability of holo azurin.
