    • The introduction of this paper provides an overview of infants' prediction skills of action goals, as well as their ability to predict perceptual acoustic information. Prediction skills' neurological correlates in general are discussed. A central hypothesis under investigation is that there are commonalities between the development of speech and manipulation. The current research is focused on the communication mode investigating infants' ability to associate images of familiar objects with auditory-stimuli presented both as whole words in intact form and as disrupted (partly noise-replaced) spoken words. The looking behaviour of the infants was measured with the Tobii eye-tracking device. The results suggest that 11 to 16 month-old infants recognize the target object when the word referring to it was intact, i.e. when the name of the object was presented in its entirety. However, the infants did not seem to recognize the target object when the word referring to it was partially masked so that only its initial phonetic information was presented. These results indicate that young infants are sensitive to phonetic information of the words and may need more extensive linguistic experience in order to derive full lexical forms from partially masked words. The paper concludes with suggestions for future demonstrations of anticipation of speech.

  • Hemochromatosis (HFE) gene mutations and risk of gastric cancer in the European Prospective Investigation into Cancer and Nutrition (EPIC) study

    • Hereditary hemochromatosis (HH) is a strong risk factor for hepatocellular cancer, and mutations in the HFE gene associated with HH and iron overload may be related to other tumors, but no studies have been reported for gastric cancer (GC). A nested case-control study was conducted within the European Prospective Investigation into Cancer and Nutrition (EPIC), including 365 incident gastric adenocarcinoma and 1284 controls matched by center, sex, age and date of blood collection. Genotype analysis was performed for two functional polymorphisms (C282Y/rs1800562 and H63D/rs1799945) and seven tagSNPs of the HFE genomic region. Association with all gastric adenocarcinoma, and according to anatomical localization and histological subtype, was assessed by means of the odds ratio (OR) and 95% confidence interval (CI) estimated by unconditional logistic regression adjusted for the matching variables. We observed a significant association for H63D with OR (per rare allele) of 1.32 (CI = 1.03-1.69). In subgroup analyses, the association was stronger for non-cardia anatomical subsite (OR = 1.60, CI = 1.16-2.21) and intestinal histological subtype (OR = 1.82, CI = 1.27-2.62). Among intestinal cases, two tagSNPs (rs1572982 and rs6918586) also showed a significant association that disappeared after adjustment for H63D. No association with tumors located in the cardia or with diffuse subtype was found for any of the nine SNPs analyzed. Our results suggest that H63D variant in HFE gene seems to be associated with GC risk of the non-cardia region and intestinal type, possibly due to its association with iron overload although a role for other mechanisms cannot be entirely ruled out.

  • Impact of Cigarette Smoking on Cancer Risk in the European Prospective Investigation into Cancer and Nutrition Study

    • Purpose Our aim was to assess the impact of cigarette smoking on the risk of the tumors classified by the International Agency for Research on Cancer as causally associated with smoking, referred to as tobacco-related cancers (TRC). Methods The study population included 441,211 participants (133,018 men and 308,193 women) from the European Prospective Investigation Into Cancer and Nutrition. We investigated 14,563 participants who developed a TRC during an average follow-up of 11 years. The impact of smoking cigarettes on cancer risk was assessed by the population attributable fraction (AF(p)), calculated using the adjusted hazard ratios and 95% CI for current and former smokers, plus either the prevalence of smoking among cancer cases or estimates from surveys in representative samples of the population in each country. Results The proportion of all TRC attributable to cigarette smoking was 34.9% (95% CI, 32.5 to 37.4) using the smoking prevalence among cases and 36.2% (95% CI, 33.7 to 38.6) using the smoking prevalence from the population. The AF(p) were above 80% for cancers of the lung and larynx, between 20% and 50% for most respiratory and digestive cancers and tumors from the lower urinary tract, and below 20% for the remaining TRC. Conclusion Using data on cancer incidence for 2008 and our AF(p) estimates, about 270,000 new cancer diagnoses per year can be considered attributable to cigarette smoking in the eight European countries with available data for both men and women (Italy, Spain, United Kingdom, the Netherlands, Greece, Germany, Sweden, Denmark).

  • Structure evolution of spin-coated phase separated EC/HPC films

    • Porous phase-separated films made of ethylcellulose (EC) and hydroxypropylcellulose (HPC) are commonly used for controlled drug release. The structure of these thin films is controlling the drug transport from the core to the surrounding liquid in the stomach or intestine. However, detailed understanding of the structure evolution is lacking. In this work, we use spin-coating, a widely applied technique for making thin uniform polymer films, to mimic the industrial manufacturing process of fluidized bed spraying. The aim of this work is to understand the structure evolution and phase separation kinetics of single layer and multi-layer spin-coated EC/HPC films. The structure evolution is characterized using confocal laser scanning microscopy (CLSM) and image analysis. The influence of spin-coating parameters and EC:HPC ratio on the final phase-separated structure and the film thickness was determined. Varying spin speed and EC:HPC ratio gave us precise control over the characteristic length scale and thickness of the film. The results show that the phase separation occurs through spinodal decomposition and that the characteristic length scale increases with decreasing spin speed and with increasing HPC ratio. The thickness of the spin-coated film decreases with increasing spin speed. Furthermore, optimized spin-coating parameters were selected to study the kinetics of phase separation in situ, in particular the coarsening mechanisms and the time dependence of the domain’s growth as a function of EC:HPC ratio. We identified two main coarsening mechanisms: interfacial tension driven hydrodynamic growth for the bicontinuous structure and diffusion driven coalescence for the discontinuous structures. In addition, we obtained information on the wetting, the shrinkage, and the evaporation process by looking at a film cross section, which allowed an estimation of the binodal of the phase diagram. The findings from this work give a good understanding of the mechanisms responsible for the morphology development and open the road towards tailoring thin EC/HPC film structures for controlled drug release.

    • Porous phase-separated ethylcellulose/hydroxypropylcellulose (EC/HPC) films are used to control drug transport from pharmaceutical pellets. The drug transport rate is determined by the structure of the porous films that are formed as water-soluble HPC leaches out. However, a detailed understanding of the evolution of the phase-separated structure in the films is lacking. In this work, we have investigated EC/HPC films produced by spin-coating, mimicking the industrial fluidized bed spraying. The aim was to investigate film structure evolution and coarsening kinetics during solvent evaporation. The structure evolution was characterized using confocal laser scanning microscopy and image analysis. The effect of the EC:HPC ratio (15 to 85 wt% HPC) on the structure evolution was determined. Bicontinuous structures were found for 30 to 40 wt% HPC. The growth of the characteristic length scale followed a power law, L(t) ∼ t(n), with n ∼ 1 for bicontinuous structures, and n ∼ 0.45-0.75 for discontinuous structures. The characteristic length scale after kinetic trapping ranged between 3.0 and 6.0 μm for bicontinuous and between 0.6 and 1.6 μm for discontinuous structures. Two main coarsening mechanisms could be identified: interfacial tension-driven hydrodynamic growth for bicontinuous structures and diffusion-driven coalescence for discontinuous structures. The 2D in-plane interface curvature analysis showed that the mean curvature decreased as a function of time for bicontinuous structures, confirming that interfacial tension is driving the growth. The findings of this work provide a good understanding of the mechanisms responsible for morphology development and open for further tailoring of thin EC/HPC film structures for controlled drug release. © 2022 The Royal Society of Chemistry
