  • Cortical activity in precision- versus power-grip tasks: an fMRI study

    • Most manual grips can be divided in precision and power grips on the basis of phylogenetic and functional considerations. We used functional magnetic resonance imaging to compare human brain activity during force production by the right hand when subjects used a precision grip and a power grip. During the precision-grip task, subjects applied fine grip forces between the tips of the index finger and the thumb. During the power-grip task, subjects squeezed a cylindrical object using all digits in a palmar opposition grasp. The activity recorded in the primary sensory and motor cortex contralateral to the operating hand was higher when the power grip was applied than when subjects applied force with a precision grip. In contrast, the activity in the ipsilateral ventral premotor area, the rostral cingulate motor area, and at several locations in the posterior parietal and prefrontal cortices was stronger while making the precision grip than during the power grip. The power grip was associated predominately with contralateral left-sided activity, whereas the precision-grip task involved extensive activations in both hemispheres. Thus our findings indicate that in addition to the primary motor cortex, premotor and parietal areas are important for control of fingertip forces during precision grip. Moreover, the ipsilateral hemisphere appears to be strongly engaged in the control of precision-grip tasks performed with the right hand.

  • Evidence for the involvement of the posterior parietal cortex in coordination of fingertip forces for grasp stability in manipulation

    • Grasp stability during object manipulation is achieved by the grip forces applied normal to the grasped surfaces increasing and decreasing in phase with increases and decreases of destabilizing load forces applied tangential to the grasped surfaces. This force coordination requires that the CNS anticipates the grip forces that match the requirements imposed by the self-generated load forces. Here, we use functional MRI (fMRI) to study neural correlates of the grip-load force coordination in a grip-load force task in which six healthy humans attempted to lift an immovable test object held between the tips of the right index finger and thumb. The recorded brain activity was compared with the brain activity obtained in two control tasks in which the same pair of digits generated forces with similar time courses and magnitudes; i.e., a grip force task where the subjects only pinched the object and did not apply load forces, and a load force task, in which the subjects applied vertical forces to the object without generating grip forces. Thus neither the load force task nor the grip force task involved coordinated grip-load forces, but together they involved the same grip force and load force output. We found that the grip-load force task was specifically associated with activation of a section of the right intraparietal cortex, which is the first evidence for involvement of the posterior parietal cortex in the sensorimotor control of coordinated grip and load forces in manipulation. We suggest that this area might represents a node in the network of cortical and subcortical regions that implement anticipatory control of fingertip forces for grasp stability.

  • COMPANION-Towards Co-operative Platoon Management of Heavy-Duty Vehicles

    • The objective of the EU project COMPANION is to develop co-operative mobility technologies for supervised vehicle platooning, in order to improve fuel efficiency and safety for goods transport. The potential social and environmental benefits inducted by heavy-duty vehicle platoons have been largely proven. However, until now, the creation, coordination, and operation of such platoons have been mostly neglected. In addition, the regulation and standardization of coordinated platooning, together with its acceptance by the end-users and the society need further attention and research. In this paper we give an overview over the project and present the architecture of the off-board and onboard platforms of the COMPANION cooperative platoon management system. Furthermore, the consortium reports on the first results of the human factors for platooning, legislative analysis of platooning aspects, clustering and optimization of platooning plans and prediction of congestion due to planned special events. Finally, we present the method of validation of the system via simulation and trials.

  • Trends in municipal and comprehensive planning (1980-2012) from an Ecosystem Services Perspective in Southern Sweden

    • In Sweden a unique project supported by the Swedish Environmental Protection Agency is developed in close collaboration with seven coastal municipalities in Skåne. These municipalities face a range of different environmental challenges, from municipalities along the eastern coast line to municipalities in the western Öresund region. Research questions and approaches are built on case studies and scenarios to assess local municipality planning processes. The objective is to study the premises of implementing the use of the Ecosystem Service (ES) approach in municipal planning and decision making. This study presents a project component that focuses on the analysis of the comprehensive plans in the city of Malmö from 1980 to 2012. Interviews with staff from municipal departments were done as a complement to the collection of data from the comprehensive plans. The results show current trends in municipal and comprehensive planning from an ES perspective, i.e., the use of ES as a tool for sustainable development. The results further show that the ES concept was explicitly first found in the comprehensive plan from 2012. Implicitly, approaches for ecosystem services management and related planning changed from “land use”, “green areas” and “green space” (1980) towards “recreation” (1990) and “biological life cycle” and “biodiversity” (2000). This change indicates that the process in local municipality planning has shifted towards a more holistic view of the importance of ecosystems for a sustainable society and healthy life, which is currently further advanced via the ES concept.

  • MRI staging using gadodiamide for soft-tissue tumors of the head and neck region. Results from a phase II trial and a 5-year clinical follow-up

    • In order to document the safety, tolerability and efficacy of gadodiamide outside CNS, an open, non-drug comparative study was performed in patients with tumors of the head and neck region. Fifty adult patients were included and 48 patients received the contrast medium. The examinations were performed on a 1.5 T imager using transverse, non-enhanced T1- and PD-/T2-weighted conventional spin-echo sequences, followed by a contrast-enhanced transverse T1-weighted sequence. Post-contrast images provided more diagnostic information compared to unenhanced images in 33 of 48 patients (69%). This information was of significant help in four and of moderate help in 14 cases. Post-contrast images compared to non-enhanced T1-weighted showed improvement in lesion delineation for 29 of the 43 patients where a lesion was observed. Only in two patients was the diagnostic information lower post-contrast. A comparison between all pre-contrast images versus contrast medium enhanced showed post-contrast images to give more diagnostic information in 14 and less in nine patients. No patient experienced discomfort in relation to gadodiamide injection. Only one adverse event occurred which was described as thirst, being of moderate intensity. The 5-year clinical outcome was analyzed and compared with the pre-operative staging. The case-books of all patients were reviewed and in 44 patients all information could be found. Of those, 18 were still alive, one with active disease (AAD) and 17 with no evidence of disease (NED). Two of those four patients, where information was incomplete, showed NED and two had died. This trial showed that contrast-enhancement using gadodiamide for evaluation of soft tissue tumors in the head and neck region was safe and provided statistically significant more diagnostic information compared with unenhanced images. MRI, when compared with palpation/inspection, changed tumor staging in approximately 30% of all cases.

    • One approach to investigate gene function, by silencing the activity of certain proteins, is the usage of double stranded decoy oligodeoxynucleotides (ds decoy ODNs). Decoy, in this sense, is ds ODNs bearing the consensus binding sequence for a DNA-binding protein. This can be used in clinical settings to attenuate the effect of overexpressed transcription factors in tumor cells. We here choose to target the oncogenic protein Myc. Since oligonucleotides are poorly internalized to cells, a cell-penetrating peptide, TP10, was coupled to the Myc decoy, using two different strategies. Either TP10 was simply mixed with ds decoy ODNs forming complexes through non-covalent electrostatic interactions, or by having a nona-nucleotide overhang in one of the decoy strands, and adding a complementary PNA sequence coupled to an NLS sequence and TP10, which could hybridize to the Myc decoy.By using these strategies, uptake was significantly enhanced, especially with the co-incubation approach. Interestingly, various endocytosis inhibitors had no effect on the uptake pattern, suggesting that uptake of these complexes is not mediated via endocytosis. Finally, a decreased proliferative capacity was observed when treating the neuroblastoma cell line N2a with TP10–PNA conjugate hybridized to Myc decoy compared to naked Myc decoy and untreated cells. A dose-dependent decrease in proliferation was also observed in MCF-7 cells, when using both strategies. These results suggest an alternative way to efficiently deliver ds ODNs into cells using the cell-penetrating peptide TP10 and prevent tumor growth by targeting the oncogenic protein Myc.
