  • Prognostic value of nuclear FBI-1 in patients with rectal cancer with or without preoperative radiotherapy

    • Factor that binds to the inducer of short transcripts of the human immunodeficiency virus-1 (FBI-1) represents as a crucial gene regulator in colorectal cancer; however, the correlation between FBI-1 and preoperative radiotherapy (RT) in rectal cancer (RC) has not yet been reported. The aim was to detect FBI-1 expression in patients with RC with or without RT, by immunohistochemistry and quantitative polymerase chain reaction, and to analyze its association with clinicopathological features and response to RT. The results from immunohistochemistry analysis (n=139) and reverse transcription-quantitative polymerase chain reaction (n=55) demonstrated that FBI-1 was overexpressed in patients with RC, whether they had received preoperative RT or not. Subsequently, the association between FBI-1 expression, and the clinicopathological features and response to RT in patients with RC was analyzed. Cytoplasmic FBI-1 was upregulated in non-RT (n=77) and RT (n=62) groups (17.7 vs. 74.0%, P<0.001; 41.1 vs. 69.4%, P=0.002, respectively) of patients with RC compared with normal mucosa. However, nuclear FBI-1 was downregulated (75.8 vs. 22.1%, P<0.001; 83.9 vs. 35.5%, P<0.001, respectively) in both groups. RT had no significant effect on FBI-1 expression in RC tissues. Furthermore, nuclear FBI-1 was positively associated with tumor-node-metastasis stage and distant recurrence (P=0.003 and P=0.010, respectively). In patients with stage I, II or III RC, higher nuclear FBI-1 expression was associated with poorer disease-free survival [hazard ratio (HR)=1.934, 95% confidence interval (CI): 1.055-3.579, P=0.033] and overall survival (HR=2.174, 95% CI: 1.102-4.290, P=0.025), independently of sex, age, growth pattern, differentiation and RT. In addition, FBI-1 was positively correlated with numerous biological factors, including p73 [Spearman's correlation coefficient (rs)=0.332, P=0.007], lysyl oxidase (rs=0.234, P=0.043), Wrap53 (rs=-0.425, P=0.0002) and peroxisome proliferator-activated receptor δ (rs=-0.294, P=0.026). In conclusion, the present study demonstrated that nuclear FBI-1 was an independent prognostic factor in patients with RC and correlated with numerous biological factors, which indicated that it may have multiple roles in RC.

    • A portable rescue device and a method for locating, by means of a first rescue device set in a search mode, a second rescue device set in a distress mode. In the method, a distress signal carrying a device identification is received from said second rescue device. A first bearing and a second bearing to the second rescue device are obtained. The first and second bearings are taken from a first and a second position, respectively. A distance between these positions is determined. A current distance and a current bearing to the second rescue device are determined on basis of the first and second bearings and the distance. The current bearing and the current distance are communicated to a user of the first rescue device. The portable rescue device is used for performing the method and for that purpose it includes a first communication unit for distress signal transmission and reception; a compass; a processor; a user interface; and a mode switch for switching between a search mode and a distress signal mode. The first communication device has an antenna structure that provides directional capability.

  • Teacher professional learning in response to contemporary challenges in early childhood education and care

    • The purpose of this doctoral thesis is to generate new knowledge, through the empirical study, of how an Early Childhood Education and Care (ECEC) work team collectively learn about two contemporary challenges in ECEC, by means of a theoretical framework. More specifically, how they learn about the relationship between teaching and play, particularly in relation to children’s digital experiences in play. The thesis is based on a combined research and development project where a work team in preschool were introduced to the theoretical framework Playresponsive early childhood education and care (PRECEC) which has implications for how to understand teaching and play activities in preschool. A sociocultural perspective functions as the theoretical framework of the thesis and the data consists of ten audio and Zoom-recorded focus group conversations which have been analyzed through Sociocultural Discourse Analysis. The thesis contains three empirical studies where the findings show how reading research articles and watching video-recordings play a central part when the participants appropriate theoretical concepts. The findings also show how time and dialogue based on scientific knowledge matters for the learning process when the work team change their reasoning about the teacher’s role in play. In addition, the findings illustrate how PRECEC, for the participants, mediate an understanding of play and teaching as responsive activities requiring mutual (digital) experiences. This mediating process is discussed as potentially enabling teacher agency. Overall, the findings contribute with new knowledge regarding how an ECEC work team learn about contemporary challenges by means of PRECEC and are therefore discussed in relation to implications for what becomes important when organizing for professional development in ECEC settings.

    • Inverse oxide/metal model systems are frequently used to investigate catalytic structure-function relationships at an atomic level. By means of a novel atomic layer deposition process, growth of single-site Fe1Ox on a Pt(111) single crystal surface was achieved, as confirmed by scanning tunneling microscopy (STM). The redox properties of the catalyst were characterized by synchrotron radiation based ambient pressure X-ray photoelectron spectroscopy (AP-XPS). After calcination treatment at 373 K in 1 mbar O2 the chemical state of the catalyst was determined as Fe3+. Reduction in 1 mbar H2 at 373 K demonstrates a facile reduction to Fe2+ and complete hydroxylation at significantly lower temperatures than what has been reported for iron oxide nanoparticles. At reaction conditions relevant for preferential oxidation of CO in H2 (PROX), the catalyst exhibits a Fe3+ state (ferric hydroxide) at 298 K while re-oxidation of iron oxide clusters does not occur under the same condition. CO oxidation proceeds on the single-site Fe1(OH)3 through a mechanism including the loss of hydroxyl groups in the temperature range of 373 to 473 K, but no reaction is observed on iron oxide clusters. The results highlight the high flexibility of the single iron atom catalyst in switching oxidation states, not observed for iron oxide nanoparticles under similar reaction conditions, which may indicate a higher intrinsic activity of such single interfacial sites than the conventional metal-oxide interfaces. In summary, our findings of the redox properties on inverse single-site iron oxide model catalyst may provide new insights into applied Fe-Pt catalysis. [Figure not available: see fulltext.]
