  • Upscaling Northern Peatland CO2 Fluxes Using Satellite Remote Sensing Data

    • Peatlands play an important role in the global carbon cycle as they contain a large soil carbon stock. However, current climate change could potentially shift peatlands from being carbon sinks to carbon sources. Remote sensing methods provide an opportunity to monitor carbon dioxide (CO2) exchange in peatland ecosystems at large scales under these changing conditions. In this study, we developed empirical models of the CO2 balance (net ecosystem exchange, NEE), gross primary production (GPP), and ecosystem respiration (ER) that could be used for upscaling CO2 fluxes with remotely sensed data. Two to three years of eddy covariance (EC) data from five peatlands in Sweden and Finland were compared to modelled NEE, GPP and ER based on vegetation indices from 10 m resolution Sentinel-2 MSI and land surface temperature from 1 km resolution MODIS data. To ensure a precise match between the EC data and the Sentinel-2 observations, a footprint model was applied to derive footprint-weighted daily means of the vegetation indices. Average model parameters for all sites were acquired with a leave-one-out-cross-validation procedure. Both the GPP and the ER models gave high agreement with the EC-derived fluxes (R-2 = 0.70 and 0.56, NRMSE = 14% and 15%, respectively). The performance of the NEE model was weaker (average R-2 = 0.36 and NRMSE = 13%). Our findings demonstrate that using optical and thermal satellite sensor data is a feasible method for upscaling the GPP and ER of northern boreal peatlands, although further studies are needed to investigate the sources of the unexplained spatial and temporal variation of the CO2 fluxes.

  • Mammalian nest predation induces small-scale nest site switching in territorial tawny owl (Strix aluco) females

    • Nest predation is a major factor affecting fitness in birds. Individuals are expected to respond to nest predation by selecting safe nesting sites and by moving away from risky sites. Thereby, perceived risk or experience of predation should lead to shifts in nest site selection. Experimental studies on behavioural and life-history consequences of nest predation have traditionally manipulated the risk of predation and studied the immediate consequences thereof. Fewer studies have however analysed the behavioural consequences of perceived predation risk to future breeding events and we know little about how sedentary territorial species respond to nest predation. We experimentally manipulated tawny owl (Strix aluco) breeding nest site choice by providing an additional alternative nest box within the territory, nearby the original nesting sites. The new nest box was provided either after a successful reproductive event (control group), or following a failed reproductive event caused by a nest predator (i.e. pine marten Martes martes, predated group). We show that tawny owls generally switched to the alternative nest site in the current breeding season when the nest was predated in the previous year, whereas they used the same nest after a successful breeding. We found no effects of previous predation experience on the probability to breed nor on clutch size. We conclude that small scale movement within the territory are used by tawny owls to minimize predation risk and that the owls use information on past predation events and nest failure to optimize their breeding decision in the following season.

    • The Swedish Transport Administration is responsible for long-term planning of the transport system, as well as for building and maintaining public roads and railways. The work of the Transport Administration involves large-scale changes in the landscape and affects a wide range of heritage sites. In road and railway projects, cultural heritage is handled in the same way as other environmental issues and there is a system for identifying and coordinating different environmental requirements during planning and building. The Transport Administration has also developed a method for identifying, evaluating and analyzing cultural heritage values that are affected by various planned infrastructure measures, called Cultural Heritage Analysis. This analysis is used as one of several planning documentations and the results are used in other planning and decision documents, such as e.g. design programs and environmental impact assessments. The methods used by the Transport Administration enables a broad approach to cultural heritage issues and makes it possible to work widely with several different types of cultural heritages, regardless of whether they are protected by law or not.

  • Experimentally determined swelling pressures and geochemical interactions of compacted Wyoming bentonite with highly alkaline solutions

    • The estimated quantity of cement for construction and sealing purposes is around 9E5 kg in the planned Swedish KBS3 repository for nuclear waste. The highly alkaline cement pore fluid (pH > 12) may affect other components in the repository, and especially the bentonite buffer is of concern. In this study, we simulated possible interactions between cement and bentonite by contacting highly compacted bentonite with high molar hydroxide solutions in a series of laboratory experiments. Wyoming bentonite (MX-80) and purified homo-ionic Na- and Ca-montmorillonite were used for tests with 0.1, 0.3 and 1.0 M NaOH and saturated Ca(OH)2 solutions. Pressure cells with permeable filters were loaded with compacted discs of bentonite at the proposed buffer density (2000 kg/m(3) at full water saturation). A hydroxide solution was circulated on one side of the cell and an isotonic chloride solution on the other during a minimum of 45 days. Swelling pressure and solution pH were monitored during the tests and the change in the solution composition and bentonite mineralogy were determined after completed tests. No effect on swelling pressure was observed in tests with 0.1 M NaOH (pH 12.9) or saturated Ca(OH)2 solutions (pH 12.4) and the mineralogical/chemical changes of the clay were minimal. The bentonite swelling pressure was significantly reduced in the tests with 0.3 (pH 13.3) and 1.0 M (pH 13.8) NaOH solutions. The reduction seems to be due to an instant osmotic effect, and to a continuous dissolution of silica minerals, resulting in mass loss and, consequently, a decrease in density. At these high pH, the release of silica was dominating and the CEC of the clay increased by 20-25%. The structural formula of the smectite and X-ray diffraction tests for nonexpandability (Greene-Kelly test) provided strong evidence that the dissolution of montmorillonite proceeds incongruently through an initial step of beidellitization. The calculated rate of silica release from montmorillonite is 1.6E-9 g g(-1) clay s(-1) for 1.0 M and 5E-10 g g-1 clay s-1 for 0.3 M NaOH solutions. The Si release rate is, however, not a straightforward measure of the montmorillonite dissolution rate due to the non-stoichiometric dissolution. Upon contact between bentonite and NaCl solutions, ion-equilibrium is established between the external solution and the exchangeable cations of the clay. A similar initial pressure response on exposure of bentonite to NaOH solutions indicates that such equilibrium may establish also with an external NaOH solution. If so, the OH-concentration of the clay pore water will be lower than that of the external solution, which would explain that dissolution rates in our experiments with highly compacted bentonite are lower than those reported for batch experiments with hydroxide solutions of the same concentration.     • Metabolism of MPTP (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) to the neurotoxin MPP+ in the brain causes permanent Parkinson's disease-like symptoms by destroying dopaminergic neurons in the pars compacta of the substantia nigra in humans and non-human primates. However, the complete molecular pathology underlying MPTP-induced parkinsonism remains poorly understood. We used dual polarity matrix-assisted laser desorption/ionization mass spectrometry imaging to thoroughly image numerous glycerophospholipids and sphingolipids in coronal brain tissue sections of MPTP-lesioned and control non-human primate brains (Macaca mulatta). The results revealed specific distributions of several sulfatide lipid molecules based on chain-length, number of double bonds, and importantly, hydroxylation stage. More specifically, certain long-chain hydroxylated sulfatides with polyunsaturated chains in the molecular structure were depleted within motor-related brain regions in the MPTP-lesioned animals, e.g., external and internal segments of globus pallidus and substantia nigra pars reticulata. In contrast, certain long-chain non-hydroxylated sulfatides were found to be elevated within the same brain regions. These findings demonstrate region-specific dysregulation of sulfatide metabolism within the MPTP-lesioned macaque brain. The depletion of long-chain hydroxylated sulfatides in the MPTP-induced pathology indicates oxidative stress and oligodendrocyte/myelin damage within the pathologically relevant brain regions. Hence, the presented findings improve our current understanding of the molecular pathology of MPTP-induced parkinsonism within primate brains, and provide a basis for further research regarding the role of dysregulated sulfatide metabolism in PD.

    • A strategy to determine effective diffusion coefficients of proteins in chromatographic gels is presented in this article. An experimental methodology based on frontal liquid chromatography was combined with a numerical methodology based on a mathematical model describing the chromatographic process including the extra-column dispersion, the dispersion due to the packed bed, the external mass transfer from the bulk phase to the stationary phase, and the diffusive transport within the stationary phase. The methodology has several advantages compared to previously reported methods to determine diffusion coefficients in that no other equipment than an HPLC is required, any class of stationary phases can be investigated as long as the experiments are performed under non-binding conditions, and no modification, e.g., moulding of slabs or membranes, to the stationary phase is required. To show the applicability of the methodology, the effective diffusion coefficients of lysozyme, bovine serum albumin, and immunoglobulin gamma in Sepharose (TM) CL-4B were determined and shown to be comparable with those determined with other methods.   • Vulnerability of Glioblastoma Cells to Catastrophic Vacuolization and Death Induced by a Small Molecule

    • Glioblastoma multiforme (GBM) is the most aggressive form of brain cancer with marginal life expectancy. Based on the assumption that GBM cells gain functions not necessarily involved in the cancerous process, patient-derived glioblastoma cells (GCs) were screened to identify cellular processes amenable for development of targeted treatments. The quinine-derivative NSC13316 reliably and selectively compromised viability. Synthetic chemical expansion reveals delicate structure-activity relationship and analogs with increased potency, termed Vacquinols. Vacquinols stimulate death by membrane ruffling, cell rounding, massive macropinocytic vacuole accumulation, ATP depletion, and cytoplasmic membrane rupture of GCs. The MAP kinase MKK4, identified by a shRNA screen, represents a critical signaling node. Vacquinol-1 displays excellent in vivo pharmacokinetics and brain exposure, attenuates disease progression, and prolongs survival in a GBM animal model. These results identify a vulnerability to massive vacuolization that can be targeted by small molecules and point to the possible exploitation of this process in the design of anticancer therapies.
