  • Lrig1 is a cell-intrinsic modulator of hippocampal dendrite complexity and BDNF signaling

    • Even though many extracellular factors have been identified as promoters of general dendritic growth and branching, little is known about the cell-intrinsic modulators that allow neurons to sculpt distinctive patterns of dendrite arborization. Here, we identify Lrig1, a nervous system-enriched LRR protein, as a key physiological regulator of dendrite complexity of hippocampal pyramidal neurons. Lrig1-deficient mice display morphological changes in proximal dendrite arborization and defects in social interaction. Specifically, knockdown of Lrig1 enhances both primary dendrite formation and proximal dendritic branching of hippocampal neurons, two phenotypes that resemble the effect of BDNF on these neurons. In addition, we show that Lrig1 physically interacts with TrkB and attenuates BDNF signaling. Gain and loss of function assays indicate that Lrig1 restricts BDNF-induced dendrite morphology. Together, our findings reveal a novel and essential role of Lrig1 in regulating morphogenic events that shape the hippocampal circuits and establish that the assembly of TrkB with Lrig1 represents a key mechanism for understanding how specific neuronal populations expand the repertoire of responses to BDNF during brain development.

  • Alcohol consumption, cardiac biomarkers, and risk of atrial fibrillation and adverse outcomes

    • AIMS: There is inconsistent evidence on the relation of alcohol intake with incident atrial fibrillation (AF), in particular at lower doses. We assessed the association between alcohol consumption, biomarkers, and incident AF across the spectrum of alcohol intake in European cohorts.METHODS AND RESULTS: In a community-based pooled cohort, we followed 107 845 individuals for the association between alcohol consumption, including types of alcohol and drinking patterns, and incident AF. We collected information on classical cardiovascular risk factors and incident heart failure (HF) and measured the biomarkers N-terminal pro-B-type natriuretic peptide and high-sensitivity troponin I. The median age of individuals was 47.8 years, 48.3% were men. The median alcohol consumption was 3 g/day. N = 5854 individuals developed AF (median follow-up time: 13.9 years). In a sex- and cohort-stratified Cox regression analysis alcohol consumption was non-linearly and positively associated with incident AF. The hazard ratio for one drink (12 g) per day was 1.16, 95% CI 1.11-1.22, P < 0.001. Associations were similar across types of alcohol. In contrast, alcohol consumption at lower doses was associated with reduced risk of incident HF. The association between alcohol consumption and incident AF was neither fully explained by cardiac biomarker concentrations nor by the occurrence of HF.CONCLUSIONS: In contrast to other cardiovascular diseases such as HF, even modest habitual alcohol intake of 1.2 drinks/day was associated with an increased risk of AF, which needs to be considered in AF prevention.

  • Children's Participation in Child Protection Practice in Ghana : Practitioners' Recommendations for Practice

    • The United Nations Convention on the Rights of the Child (UNCRC) includes provisions to ensure that children and young people participate in decisions affecting their lives. Ghana ratified the convention in 1990 making a commitment to review its child protection policies and legislation in compliance with provisions in the UNCRC. Yet, national policies and legislation do not include practical guidelines to promote children's participation in the child protection process. Thus, this qualitative study presents findings from in-depth interviews with 15 child protection practitioners on their views about some practical guidelines to promote children's participation in child protection. Data from the interviews were subjected to constructivist grounded theory analysis. The study findings revealed the age of the child, separate room for children, creating a friendly environment and education as some important factors for practitioners to consider in promoting participatory practices for children. Child protection policies and legislation in Ghana should include these suggestions to ensure that children's views are heard in the child protection process. To realize the overarching goal of achieving active child participation in child protection, further research may focus on the views of parents and children on how to develop culturally relevant strategies to promote child participation.

  • What makes kinship caregivers unprepared for children in their care? : Perspectives and experiences from kinship care alumni in Ghana

    • Evidence from international literature suggests that children in kinship care have more positive experiences than those receiving care in institutions. Kinship care for children is highly used in Ghana as an alternative care option mostly because of the belief that it is important to keep children within their families to continue family relationships, culture and to cement family bonds. However, research in Ghana shows that kinship caregivers may not be prepared to provide care and protection for children in need of parental care. As a result of this, the safety and wellbeing of these children could be at risk. This study reports on a qualitative investigation involving 15 young kinship care alumni in Ghana to explore what kinship caregivers' unpreparedness means and what causes them to be unprepared. Data from the in-depth interviews were analysed following the constructivist grounded theory approach. The study revealed that kinship caregivers' unpreparedness makes it difficult for children's needs to be met. Poverty, unemployment and cultural and religious beliefs emerged as causing caregivers' unpreparedness. Implications of the findings for improving the safety and wellbeing of children in kinship care are discussed.

  • Monuments and their functions in urban public space

    • The aim of the article is to present and discuss functions of public monuments in relation to different dimensions of geographical space. The authors discuss public spaces, monuments, public art, based on a mixed-method approach and an analysis of scientific literature as the main research method. The theoretical discussion is supplemented with the results of Internet searches and an analysis of media supplements, and specific examples are given, including some from Poland. An additional method was autoethnography, which involved an analysis of cultural phenomena based on the authors' experience. The analysis revealed that monuments were part of public art and thus enriched public spaces in cities. They fulfilled different important functions: artistic, symbolic, commemorative, political, social, religious, marketing, and mixed. Additionally, the monuments reflected the contemporary transformation of ideas and social orders and therefore also reflected contemporary urban debates. They were products of social relations, powers, ideas, identities, and the collective memory reflected in the urban spatial structure of cities. The authors conclude that the examples presented in the demonstrate that monuments perform various functions in urban public spaces. From a spatial perspective, the role of monuments depends on their different impacts on people's perceptions and interpretations of space.

  • D-Alanine-Controlled Transient Intestinal Mono-Colonization with Non-Laboratory-Adapted Commensal E. coli Strain HS

    • Soon after birth the mammalian gut microbiota forms a permanent and collectively highly resilient consortium. There is currently no robust method for re-deriving an already microbially colonized individual again-germ-free. We previously developed the in vivo growth-incompetent E. coli K-12 strain HA107 that is auxotrophic for the peptidoglycan components D-alanine (D-Ala) and meso-diaminopimelic acid (Dap) and can be used to transiently associate germ-free animals with live bacteria, without permanent loss of germ-free status. Here we describe the translation of this experimental model from the laboratory-adapted E. coli K-12 prototype to the better gut-adapted commensal strain E. coli HS. In this genetic background it was necessary to complete the D-Ala auxotrophy phenotype by additional knockout of the hypothetical third alanine racemase metC. Cells of the resulting fully auxotrophic strain assembled a peptidoglycan cell wall of normal composition, as long as provided with D-Ala and Dap in the medium, but could not proliferate a single time after D-Ala/Dap removal. Yet, unsupplemented bacteria remained active and were able to complete their cell cycle with fully sustained motility until immediately before autolytic death. Also in vivo, the transiently colonizing bacteria retained their ability to stimulate a live-bacteria-specific intestinal Immunoglobulin (Ig) A response. Full D-Ala auxotrophy enabled rapid recovery to again-germ-free status. E. coli HS has emerged from human studies and genomic analyses as a paradigm of benign intestinal commensal E. coli strains. Its reversibly colonizing derivative may provide a versatile research tool for mucosal bacterial conditioning or compound delivery without permanent colonization.

  • Diving below the surface : A framework for arctic health research to support thriving communities

    • Aims: Historically, health research in the Arctic has focused on documenting ill-health using a narrow set of deficit-oriented epidemiologic indicators (i.e., prevalence of disease and mortality rates). While useful, this type of research does not adequately capture the breadth and complexities of community health and well-being, and fails to highlight solutions. A community’s context, strengths, and continued expressions of well-being need to guide inquiries, inform processes, and contextualize recommendations. In this paper, we present a conceptual framework developed to address the aforementioned concerns and inform community-led health and social research in the Arctic.Methods: The proposed framework is informed by our collective collaborations with circumpolar communities, and syntheses of individual and group research undertaken throughout the Circumpolar North. Our framework encourages investigation into the contextual factors that promote circumpolar communities to thrive.Results: Our framework centers on the visual imagery of an iceberg. There is a need to dive deeper than superficial indicators of health to examine individual, family, social, cultural, historical, linguistic, and environmental contexts that support communities in the Circumpolar North to thrive. A participatory community-based approach in conjunction with ongoing epidemiologic research is necessary in order to effectively support health and wellness.Conclusions: The iceberg framework is a way to conceptualize circumpolar health research and encourage investigators to both monitor epidemiologic indicators and also dive below the surface using participatory methodology to investigate contextual factors that support thriving communities.

  • From Resilient to Thriving : Policy Recommendations to Support Health and Well-being in the Arctic

    • In 2018 – 19, eight Indigenous and non-Indigenous individuals from Canada, Greenland/Denmark, Sweden, and Alaska/United States came together to address research questions relevant to Arctic nations’ shared challenges and opportunities. Our work incorporated critical, community-based perspectives on Arctic health and well-being and promoted strengths-based approaches developed in partnership with Arctic communities. In this article we describe the group’s 16 action-oriented policy recommendations to support health and well-being in the Arctic in four thematic areas: 1) acknowledge and integrate Indigenous rights and knowledges, 2) implement meaningful action to address Indigenous determinants of health, 3) expand health-oriented monitoring and assessment programs, and 4) implement community-led, critical research approaches that focus on partnerships, reciprocity, adherence to ethical guidelines, and funding community-based research. Our recommendations are actionable guidelines for policy and research aimed at reducing inequities, supporting Indigenous expertise and existing knowledge, and promoting thriving communities in the Arctic.

  • Structural Deformation of Sm@C88under High Pressure

    • We have studied the structural transformation of Sm@C88 under pressure up to 18 GPa by infraredspectroscopy combined with theoretical simulations. The infrared-active vibrational modes of Sm@C88 at ambient conditions have been assigned for the first time. Pressure-induced blue and red shiftsof the corresponding vibrational modes indicate an anisotropic deformation of the carbon cage uponcompression. We propose that the carbon cage changes from ellipsoidal to approximately sphericalaround 7 GPa. A smaller deformation of the carbon bonds in the area close to the Sm atom in thecage suggests that the trapped Sm atom plays a role in minimizing the compression of the adjacentbonds. Pressure induced a significant reduction of the band gap of the crystal. The HOMO-LUMOgap of the Sm@C88 molecule decreases remarkably at 7 GPa as the carbon cage is deformed. Also,compression enhances intermolecular interactions and causes a widening of the energy bands. Botheffects decrease the band gap of the sample. The carbon cage deforms significantly above 7 GPa,from spherical to a peanut-like shape and collapses at 18 GPa.

  • High pressure and high temperature induced polymerization of doped C60 materials

    • Several metastable doped C60 polymers are synthesized under high pressure and high temperature (1.5GPa, 573K and 2GPa, 700K, respectively), using C60/ferrocene (Fc, Fe(C5H5)2), C60/Ni(OEP) and C60/AgNO3 as starting materials. Raman and IR spectroscopy are used to study the polymerization of these samples after HPHT treatment. It is found that the polymerization degree is always lower than that of pure C60 treated at same conditions, which is attributed to the space limitation by the dopants. We also find that even at same conditions, the three doped materials form different polymeric phases of the doped materials. This is attributed to the unique initial lattice structures and the different degrees of spatial confinement provided by the dopants.
