  • Seve terranes of the Kebnekaise Mts., Swedish Caledonides, and their amalgamation, accretion and affinity

    • A major allochthon of the Scandinavian Caledonides, the Seve belt has traditionally been considered to be derived from the rifted margin and continent-ocean transition (COT) of Baltica. However, geochronological results obtained from its inferred northern equivalent, the Kalak Nappe Complex (KNC), have been taken to indicate an exotic affinity of this complex and of also Seve terranes, an interpretation adopted in recent palaeogeographic models. In the Kebnekaise Mts., the COT is represented by the Kebnekaise terrane composed of amphibolitized dykes of gabbro and dolerite of depleted magma source and rare felsic and ultramafic rocks. Coronitic dolerite and gabbro with abundant rutile suggest high pressures before or during amalgamation with the underlying Mårma terrane, composed of quartzofeldspathic gneisses intruded by mafic and granitic rocks, the latter including a previously dated c. 845 Ma-old granite. The granite mingled with mildly alkaline dolerites chemically similar to transitional basalts of continental rifts. Following emplacement at shallow (andalusite stability) crustal levels, the igneous complex and host rocks underwent extensive deformation, metamorphism within the sillimanite-kyanite stability field and local migmatization. The Kebnekaise and Mårma terranes amalgamated in early Ordovician as indicated by the U-Pb age of 487±7 Ma obtained from titanite fabrics of deformed granite in thrust vicinity, and by 40Ar-39Ar results. The pressure increase in both terranes suggests that amalgamation occurred during initial subduction and imbrication. Correlation of the Mårma terrane with the KNC is discussed. Results obtained in this study give no reason to ascribe an exotic affinity to the Seve terranes of the Kebnekaise Mts.

  • A Distributed Passive Measurement Infrastructure

    • In this paper we describe a distributed passive measurement infrastructure. Its goals are to reduce the cost and configuration effort per measurement. The infrastructure is scalable with regards to link speeds and measurement locations. A prototype is currently deployed at our university and a demo is online at http://inga.its.bth.se/projects/dpmi. The infrastructure differentiates between measurements and the analysis of measurements, this way the actual measurement equipment can focus on the practical issues of packet measurements. By using a modular approach the infrastructure can handle many different capturing devices. The infrastructure can also deal with the security and privacy aspects that might arise during measurements.

    • The synthesis and characterization of all diastereomers of a linear symmetrically fused tris-Troger's base analogue are described. The diastereomers are unambiguously assigned as syn-anti 1a, anti-anti 1b, and syn-syn 1c isomers, by using X-ray diffraction analysis and NMR spectroscopy. For the first time, the anti-anti and the syn-syn diastereomers of a linear symmetrically fused tris-Troger's base analogue have been synthesized. Molecules 1a and 1c are new cleft compounds and analysis of compound 1a in the solid state shows inclusion of one molecule of CH2Cl2 in the larger aromatic cleft, whereas in isomer 1c disordered solvent molecules are trapped in the extended aromatic cleft. Furthermore, in the solid state, isomer 1c forms infinite open channels along one of the crystallographic axes and perpendicular to this axis there are infinitely extending "wedged-ravines". Importantly, each of the diastereomers 1a-c is resistant to inversion at the stereogenic nitrogen atoms under strongly and weakly acidic conditions in the range from room temperature (RT) to 95 degrees C. This observed configurational stability at the stereogenic nitrogens of 1a-c is unique for analogues of Troger's base in general to date. Finally, the ratio of cleft compounds 1a and 1c significantly increased relative to cavity compound 1b when ammonium chloride was used as an additive in the Troger's base condensation to 1a-c suggesting a templating effect of the ammonium ion.

  • A Spatiotemporal Organ-Wide Gene Expression and Cell Atlas of the Developing Human Heart

    • The process of cardiac morphogenesis in humans is incompletely understood. Its full characterization requires a deep exploration of the organ-wide orchestration of gene expression with a single-cell spatial resolution. Here, we present a molecular approach that reveals the comprehensive transcriptional landscape of cell types populating the embryonic heart at three developmental stages and that maps cell-type-specific gene expression to specific anatomical domains. Spatial transcriptomics identified unique gene profiles that correspond to distinct anatomical regions in each developmental stage. Human embryonic cardiac cell types identified by single-cell RNA sequencing confirmed and enriched the spatial annotation of embryonic cardiac gene expression. In situ sequencing was then used to refine these results and create a spatial subcellular map for the three developmental phases. Finally, we generated a publicly available web resource of the human developing heart to facilitate future studies on human cardiogenesis.

  • Treatment with the soluble guanylate cyclase activator BAY 60-2770 restores in vitro bladder contractile responses in a rat model of chronic prostatitis

    • Aim: Examine how innate bladder contractility and corresponding receptor expression was affected by chronic prostatitis (CP) and how treatment with the soluble guanylate cyclase (sGC) activator BAY 60-2770 influenced this. Methods: To create a functional model for CP, 24 male Sprague-Dawley rats were intraprostatically injected with either zymosan or saline, serving as control. After a recovery period, the rats were treated with either BAY 60-2770 or dimethyl sulfoxide (DMSO; vehicle) on days 8–20. Urine samples were collected for measurement of ATP. On day 21, the bladder was excised and contractile responses to electrical field stimulation (EFS), methacholine, ATP and nitric oxide (NO) were examined in an in vitro organ bath. Subsequently, the expression of purinergic (P2X1 & P2X3) and muscarinic (M3) receptors as well as sGC was examined immunohistochemically. Results: Induction of CP led to significantly attenuated purinergic bladder contractions, which were normalized by treatment with BAY 60-2770. Induction of CP did not alter the contractile bladder responses to EFS, methacholine or NO. However, treatment with BAY 60-2770 led to significantly increased contractile bladder responses to EFS and MeCh and significantly enhanced relaxatory nitrergic responses. There were no significant differences between the groups regarding purinergic or cholinergic receptor expression, however treatment with BAY 60-2770 led to attenuated expression of sGC in the urothelium. Conclusion: Taken together, these findings indicate that drugs targeting the nitric oxide/cyclic guanosine monophosphate (NO/cGMP) pathway may be a promising option to restore alterations in bladder contractility that arise due to CP.

  • Efficient improvement of energy efficiency in small and medium- sized Swedish firms

    • This is a dissertation about efficient implementation of energy efficiency measures in small and medium-sized Swedish firms. The aim is to investigate the potential for economically efficient implementation of energy efficiency improvement measures in small and medium-sized firms. The thesis contains five papers that analyse different aspects that have been put forth in policy documents and academic debate as  methods to improve energy efficiency in non-energy intensive sectors.By reading policy documents, interviewing representatives of small and medium- sized firms and energy auditors as well as analysing data from the Swedish energy audit program, different aspects of energy management practices, energy services and energy audits are considered. The thesis is the product of an interdisciplinary context but economic theory is at the foundation of the analysis and has helped formulate questions and hypotheses that have been tested and explored with the data.The results show that while the potential for improving energy efficiency in the small and medium- sized sector in Sweden is large there are challenges to realizing it in each individual firm. There is potential for improving energy efficiency in the sector and not just for investments in new technology but also for adjusting existing machinery and changing behaviour, but costs for investigating the potential and implementing the measures are large relative to the improvement potential in each company. Energy management practices in this sector are lacking and energy services will only be demanded if reduction in production cost is estimated larger than transaction costs of the service. The Swedish energy audit program has led to the implementation of energy efficiency improvements in the participating firms but compared to other policy instruments it has been a less cost-efficient way to improve energy efficiency in Sweden.

    • Mito-nuclear insertions, or NUMTs, relate to genetic material of mitochondrial origin that have been transferred to the nuclear DNA molecule. The increasing amounts of genomic data currently being produced presents an opportunity to investigate this type of patterns in genome evolution of non-model organisms. Identifying NUMTs across a range of closely related taxa allows one to generalize patterns of insertion and maintenance in autosomes, which is ultimately relevant to the understanding of genome biology and evolution. Here we collected existing pairwise genome-mitogenome data of the order Strigiformes, a group that includes all the nocturnal bird predators. We identified NUMTs by applying percent similarity thresholds after blasting mitochondrial genomes against nuclear genome assemblies. We identified NUMTs in all genomes with numbers ranging from 4 in Bubo bubo to 24 in Ciccaba nigrolineata. Statistical analyses revealed NUMT size to negatively correlate with NUMT's sequence similarity to with original mtDNA region. Lastly, characterizing these nuclear insertions of mitochondrial origin in a comparative genomics framework produced variable phylogenetic patterns, suggesting in some cases that insertions might pre-date speciation events within Strigiformes.

    • Feathers comprise a series of evolutionary innovations but also harbour colour, a key biological trait known to co-vary with life history or complex traits. Those relationships are particularly true in melanin-based pigmentation species due to known pleiotropic effects of the melanocortin pathway - originating from melanin-associated phenotypes. Here, we explore the molecular basis of melanin colouration and expected co-variation at the molecular level in the melanin-based, colour polymorphic system of the tawny owl (Strix aluco). An extensive body of literature has revealed that grey and brown tawny owl colour morphs differ in a series of life history and behavioural traits. Thus, it is plausible to expect co-variation also at molecular level between colour morphs. To investigate this possibility, we assembled the first draft genome of the species against which we mapped ddRADseq reads from 220 grey and 150 brown morphs - representing 10 years of pedigree data from a population in Southern Finland - and explored genome-wide associations with colour phenotype. Our results revealed putative molecular signatures of cold adaptation strongly associated with the grey phenotype, namely, a non-synonymous substitution in MCHR1, plus 2 substitutions in non-coding regions of FTCD and FAM135A whose genotype combinations obtained a predictive power of up to 100% (predicting grey colour). These suggest a molecular basis of cold environment adaptations predicted to be grey-morph specific. Our results potentially reveal part of the molecular machinery of melanin-associated phenotypes and provide novel insights towards understanding the functional genomics of colour polymorphism in melanin-based pigmented species.

  • The Effects of Wind Power on Human Interests : A Synthesis

    • PrefaceThere is a great need for knowledge concerning the impacts of wind power on humans, landscapes, the marine environment, birds, bats and other mammals.Previous studies of these environmental impacts have lacked an overall view of the effects. This has led to deficiencies in the processes surrounding the establishment of new wind farms. Vindval is a knowledge programme undertaken as a collaboration between the Swedish Energy Agency and the Swedish Environmental Protection Agency. Its aim is to gather and communicate scientific knowledge about the impacts of wind power on people and the natural environment. The programme continues until 2013.Vindval comprises some 30 individual research projects, together with four synthesis projects. Syntheses are prepared by experts, who compile and assess overall research results and experience regarding the effects of wind power in four different areas – humans, birds/bats, marine life and terrestrial mammals.The results of this research and synthesis work will provide a basis for environmental impact assessments and for the planning and permitting processes associated with wind power installations. Vindval requires high standards in the review and approval of research proposals, in order to ensure high-quality reports.
