Exploring the Interplay of Lipids and Membrane Proteins

- The interplay between lipids and membrane proteins is known to affect membrane protein topology and thus have significant effect (control) on their functions. In this PhD thesis, the influence of lipids on the membrane protein function was studied using three different membrane protein models.A monotopic membrane protein, monoglucosyldiacylglyecerol synthase (MGS) from Acholeplasma laidlawii is known to induce intracellular vesicles when expressed in Escherichia coli. The mechanism leading to this unusual phenomenon was investigated by various biochemical and biophysical techniques. The results indicated a doubling of lipid synthesis in the cell, which was triggered by the selective binding of MGS to anionic lipids. Multivariate data analysis revealed a good correlation with MGS production. Furthermore, preferential anionic lipid sequestering by MGS was shown to induce a different fatty acid modeling of E. coli membranes. The roles of specific lipid binding and the probable mechanism leading to intracellular vesicle formation were also investigated.As a second model, a MGS homolog from Synechocystis sp. PCC6803 was selected. MgdA is an integral membrane protein with multiple transmembrane helices and a unique membrane topology. The influence of different type of lipids on MgdA activity was tested with different membrane fractions of Synechocystis. Results indicated a very distinct profile compared to Acholeplasma laidlawii MGS. SQDG, an anionic lipid was found to be the species of the membrane that increased the MgdA activity 7-fold whereas two other lipids (PG and PE) had only minor effects on MgdA. Additionally, a working model of MgdA for the biosynthesis and flow of sugar lipids between Synechocystis membranes was proposed.The last model system was another integral membrane protein with a distinct structure but also a different function. The envelope stress sensor, CpxA and its interaction with E. coli membranes were studied. CpxA autophosphorylation activity was found to be positively regulated by phosphatidylethanolamine and negatively by anionic lipids. In contrast, phosphorylation of CpxR by CpxA revealed to be increased with PG but inhibited by CL. Non-bilayer lipids had a negative impact on CpxA phosphotransfer activity.Taken together, these studies provide a better understanding of the significance of the interplay of lipids and model membrane proteins discussed here.

Parallellpublicering av konferensartiklar : principer, tillämpningar och tillgänglighet i svenska publiceringsdatabaser

- Projektets huvudsyfte har varit att undersöka kvalitetsgranskade konferensartiklars tillgänglighet samt konferensers OA- och publiceringspolicies för att på så sätt klarlägga olika aspekter vad det gäller parallellpublicering i svenska publiceringsdatabaser. För ändamålet har projektdeltagarna undersökt ett urval konferenser och konferensartiklar via sina publiceringsdatabaser under ett specifikt tidsintervall och beskrivit dem utifrån ett gemensamt schema.Undersökningen har visat att publiceringsdatabaserna fyller en viktig funktion för att synliggöra konferenspublikationer. Vi har funnit att minst ca. 50 % av konferensartiklarna kan publiceras som post-prints i våra publiceringsdatabaser. Därför är det av stor vikt att fortsätta arbetet med att utnyttja de lokala publiceringsdatabaserna som publiceringskanal och att inte bara använda dem som ett verktyg för parallellpublicering och primärpublicerat material, som avhandlingar och rapporter, utan även som en källa framförallt för konferensartiklar ”utan hem”, som vi nu vet utgör en stor del av vårt undersökningsmaterial (ca. 15 %).Mellan 20-25 % av de undersökta artiklarna fanns i något slags öppet OA-arkiv; ca. hälften av dessa fanns i någon av projektdeltagarnas egen publiceringsdatabas. Detta talar för att den egna publiceringsdatabasen är en viktig faktor för öppen tillgänglighet.Ca. 10% av konferenserna i studien hade en uttrycklig OA-policy eller gav uttryck för en sådan genom att öppet tillgängliggöra konferensartiklar på sina konferenssajter. Ett stort problem när det gäller parallellpublicering av konferensartiklar är just bristen på information om OA-policy.Konferenspubliceringslandskapet är komplicerat och parallellpubliceringen av dokument härifrån är mycket tidskrävande. Vi skulle framför allt önska en liknande policy-resurs som SHERPA/RoMEO för konferenser. Men för närvarande finns inget annat alternativ än att lusläsa konferensernas upphovsrättsinformation till författarna och av denna försöka dra slutsatser om ev. parallellpublicering.För att underlätta framtida hantering och klassificering av konferensartiklar i svenska publicerings-databaser, framför allt med tanke på ev. framtida nationella bibliometriska analyser via SwePub, föreslås ett antal rekommendationer.

- We present an ISOCAM-CVF map of the ρ Oph A region, covering 3′ × 3′. For each 6 arcsec2 pixel, we extract the spectrum from 5–15 μm. We determine the fluxes of the main PAH features by fitting Lorentzian profiles to the spectrum. The peaks of the various PAH components correspond well with the known positions of the PDRs in this vicinity. The spectrum in several pixels exhibits strong rotational lines of molecular hydrogen which can be used to derive the physical properties of the cloud. The H2 emission traces the hot gas of the bipolar CO outflow from VLA1623.

- More inclusive forms of governance are increasingly advocated that allow for stakeholder participation and joint capacity building. Platforms for dialogue and the exchange of knowledge are seen as one tool to integrate different knowledge systems such as science and local knowledge. In this study, we examine a Swedish Co-management Initiative of fisheries using an integrated version of Plummer and Fitzgibbon’s ‘Adaptive Co-Management’ (2004) and Senecah’s ‘Trinity of Voice’ (2004) frameworks in analysing the participation process, communication, and learning outcomes. The results show that participating actors were successful in developing trust and enhancing learning, starting with a conflict situation and diverging interests. Attention to ‘access’ and ‘standing’ as part of participation, and skilled facilitation, were key issues in achieving these results. The article provides insights with regard to the use of established frameworks, here applied to an empirical case, enhance our understanding of learning in conflicted contexts, and helps practitioners in designing and institutionalizing learning processes and platforms in other contexts.

- Surprisingly high amounts of H2O have recently been reported in the circumstellar envelope around the M-type AGB star W Hya. However, substantial uncertainties remain, as the required radiative transfer modelling is difficult due to high optical depths, sub-thermal excitation and the sensitivity to the combined radiation field from the central star and dust grains.
