  • Pheromone of the elm bark beetle Scolytus laevis (Coleoptera Scolytidae) : stereoisomers of 4-methyl-3-heptanol reduce interspecific competition

    • Stereoisomers of 4-methyl-3-heptanol (MH) are pheromone components of several Scolytus bark beetles. The elm bark beetle Scolytus laevis (Coleoptera: Scolytidae) has in previous studies been caught in traps baited with commercial MH containing all four stereoisomers, but the lure has been considered a weak attractant. In this study, we addressed the question whether stereospecific responses by S. laevis to stereoisomers of MH might contribute to its niche separation from other sympatric Scolytus species. Using GC-MS, we analyzed extracts of hindguts and abdomens from male and female S. laevis and the sympatric S. triarmatus. We also tested all four MH-stereoisomers individually and in combinations in the field to determine their role for S. laevis. All four stereoisomers were synthesized via a boronic ester method with 1,2-dicyclohexylethanediol as chiral director. In addition, the (3S,4R)-stereoisomer of MH was prepared through enantioselective, lipase-mediated transesterification of a mixture of the four stereoisomers of MH. Females of both species contained small amounts of syn-MH, and males contained trace amounts of anti-MH. The anti stereoisomer (3R,4S)-MH was attractive to male and female S. laevis, whereas the syn stereoisomer (3S,4S)-MH acted as an inhibitor or deterrent and reduced the catch when added to the attractive isomer. The syn isomer is the main aggregation pheromone component of the larger and sympatric S. scolytus and possibly also of S. triarmatus. The avoidance response of S. laevis to the (3S,4S)-stereoisomer may reduce interspecific competition for host trees.

  • Sprouting capacity from intact root systems of Cirsium arvense and Sonchus arvensis decrease in autumn

    • Perennial weeds are often controlled by mechanical means, which aim at stimulating axillary and adventitious buds to sprout. This happens when the apical dominance of the main shoot is removed by defoliation or when the underground system is fragmented. By repeating the measures, the result is a depletion of storage compounds, which weakens the plants and reduces their capacity to grow and reproduce. However, timing is critical. Earlier research has indicated that emergence from fragments of Sonchus arvensis cease during a period in autumn, while the seasonal pattern of sprouting in Cirsium arvense appears to be inconsistent. We studied the emergence pattern of defoliated plants with undisturbed root systems, from late summer to early spring. Potted plants grown outdoors were exhumed at regular intervals, put under forcing conditions for 4weeks, after which shoots above and below soil level were counted and weighed together with the remaining root systems. In both species, the number and weight of emerged shoots decreased during a period in the autumn. In C.arvense, underground shoots were constantly produced during the same period, while fewer underground shoots were present in S.arvensis. For the latter species, apical dominance does not fully explain the effect; thus, endodormancy might be involved. Root weight increased until withering and did not explain the lack of emergence. Our results suggest an impaired sprouting capacity of undisturbed root systems of C.arvense and S.arvensis during SeptemberOctober, which has implications for the timing and method of control of these species.

    • A key tree species for the forest industry in Europe is Norway spruce [Picea abies (L.) Karst.]. One of its major diseases is stem and butt rot caused by Heterobasidion parviporum (Fr.) Niemelä & Korhonen, which causes extensive revenue losses every year. In this study, we investigated the parallel induction of Norway spruce genes presumably associated with salicylic acid- and jasmonic acid/ethylene-mediated signalling pathways previously observed in response to H. parviporum. Relative gene expression levels in bark samples of genes involved in the salicylic acid- and jasmonic acid/ethylene-mediated signalling pathways after wounding and inoculation with either the saprotrophic biocontrol fungus Phlebiopsis gigantea or with H. parviporum were analysed with quantitative PCR at the site of the wound and at two distal locations from the wound/inoculation site to evaluate their roles in the induced defence response to H. parviporum in Norway spruce. Treatment of Norway spruce seedlings with methylsalicylate, methyljasmonate and inhibitors of the jasmonic acid/ethylene signalling pathway, as well as the Phenylalanine ammonia lyase inhibitor 2-aminoindan-2-phosphonic acid were conducted to determine the responsiveness of genes characteristic of the different pathways to different hormonal stimuli. The data suggest that jasmonic acid-mediated signalling plays a central role in the induction of the genes analysed in this study irrespective of their responsiveness to salicylic acid. This may suggest that jasmonic acid-mediated signalling is the prioritized module in the Norway spruce defence signalling network against H. parviporum and that there seems to be no immediate antagonism between the modules in this interaction.

    • Norway spruce [Picea abies (L.) Karst.] is one of the economically most important conifer species in Europe. The major pathogen on Norway spruce is Heterobasidion parviporum (Fr.) Niemelä & Korhonen. To achieve a better understanding of Norway spruce's defence mechanisms, transcriptional responses in bark to H. parviporum infection were compared with the response to wounding using cDNA-amplified fragment length polymorphism. The majority of the recovered transcript-derived fragments (TDFs) showed a similar expression pattern for infection and wounding treatment, although inoculated samples showed an enhanced reaction. Genes related to systemic acquired resistance, e.g., PR1, accumulated after H. parviporum infection. Simultaneously, several transcripts involved in various aspects of jasmonic acid (JA)- and ethylene (ET)-mediated signalling accumulated. Genes involved in the ubiquitin/proteasome system were also regulated. Expression patterns have been confirmed by quantitative polymerase chain reaction. The expression patterns of the isolated TDFs suggest that infection with H. parviporum in Norway spruce induces a broad defence, with many similarities to non-specific defence responses in angiosperms. The parallel induction of salicylic acid- and JA/ET-mediated pathways implies spatially separated responses in different cell layers, with and without hyphal contact. A set of TDFs were analysed in an independent experiment with unrelated material treated with wounding or with inoculation with H. parviporum or Phlebiopsis gigantea, verifying the original observations and underlining the non-specific defence responses. In addition, our data suggest that rerouting of carbon in secondary metabolism is an integral part of Norway spruce induced defence. We report the sequences of three 3-deoxy-d-arabino-heptulosonate 7-phosphate synthase genes (PaDAHP1, PaDAHP2 and PaDAHP3) and their relative expression in response to wounding and infection with H. parviporum and P. gigantea. The results clearly indicate differential regulation of the three DAHPs in the induced defence responses in Norway spruce. This study gives insights into the central mechanisms in the induced defences in Norway spruce.

    • Heterobasidion parviporum (Fr.) Niemelä & Korhonen and Heterobasidion annosum (Fr.) Bref. sensu lato are some of the major forest pathogens in the northern hemisphere causing root and butt rot to conifers. The relative susceptibility to H. parviporum was investigated in a full-sib family of Norway spruce [Picea abies (L.) Karst.] by inoculating a set of 252 cloned progenies from a controlled cross. Four ramets of each progeny were used and the 2-year-old rooted cuttings were incubated for 6 weeks under greenhouse conditions. The condition of the cuttings was assessed visually and all the plants were in excellent vigour with no mortality recorded during the experiment. To score the relative susceptibility, lesion length in the inner bark and fungal growth in the sapwood were measured. Among the progenies, significant differences were found for fungal growth in the sapwood (p<0.0005). There was no significant difference for lesion length; however, there was a significant positive correlation between fungal growth and lesion length. The broad-sense heritability was 0.11 for fungal growth. This shows that the genetic component for susceptibility to H. parviporum can be detected even within a full-sib family of Norway spruce and that there is a potential for mapping quantitative trait loci for this trait in Norway spruce.

    • Background: In lakes that have become eutrophic due to sewage discharges or nutrient runoff from land, problems such as algal blooms and oxygen deficiency often persist even when nutrient supplies have been reduced. One reason is that phosphorus stored in the sediments can exchange with the water. There are indications that the high abundance of phytoplankton, turbid water and lack of submerged vegetation seen in many eutrophic lakes may represent a semi-stable state. For that reason, a shift back to more natural clear-water conditions could be difficult to achieve. In some cases, though, temporary mitigation of eutrophication-related problems has been accomplished through biomanipulation: stocks of zooplanktivorous fish have been reduced by intensive fishing, leading to increased populations of phytoplankton-feeding zooplankton. Moreover, reduction of benthivorous fish may result in lower phosphorus fluxes from the sediments. An alternative to reducing the dominance of planktivores and benthivores by fishing is to stock lakes with piscivorous fish. These two approaches have often been used in combination. The implementation of the EU Water Framework Directive has recently led to more stringent demands for measures against eutrophication, and a systematic review could clarify whether biomanipulation is efficient as a measure of that kind. Methods: The review will examine primary field studies of how large-scale biomanipulation has affected water quality and community structure in eutrophic lakes or reservoirs in temperate regions. Such studies can be based on comparison between conditions before and after manipulation, on comparison between treated and nontreated water bodies, or both. Relevant outcomes include Secchi depth, concentrations of oxygen, nutrients, suspended solids and chlorophyll, abundance and composition of phytoplankton, zooplankton and fish, and coverage of submerged macrophytes.

    • The aim of this study was to ascertain whether there is a relationship between plant species richness and plant-available N, P and water in an environment subject to little anthropogenic disturbance. To accomplish this we studied the vegetation in matorral shrub-lands in northern Patagonia, Argentina. Due to the variation in slope, precipitation and aspect between the sites water status was determined using the C-12/C-13 fraction, delta C-13, to investigate whether this was a confounding factor. The numbers of herb, shrub, liana and tree species were determined at 20 sites along an estimated precipitation gradient. Leaf P and N content and the delta C-13 of Berberis buxifolia were determined, as well as the soil P and N content at the different sites. A negative correlation was found between species richness and Berberis buxifolia foliar P concentration (52% of the species richness variation was accounted for), and a positive correlation was found between plant species richness and Berberis buxifolia foliar N: P ratios (54% of the species richness variation was accounted for). The relationship between species richness and foliar P was seen when all layers of vegetation were included (trees, lianas, shrubs and herbs). Foliar N showed no correlation with species richness, while soil extractable NH4 showed a weak positive correlation with the number of shrub layer species (lianas, shrubs and trees). The species richness of the shrub layer increased with decreasing values of delta C-13. Low soil P availability thus affects local species richness in the matorral shrub-lands of Patagonia in Argentina although the growth of vegetation in the area has been shown to be limited by N. We suggest that low P levels increase plant species richness because low soil P concentration is associated with a high P partitioning and high potential for niche separation.

  • Climate change and the probability of wind damage in two Swedish forests

    • We simulated how possible changes in wind and ground-frost climate and state of the forest due to changes in the future climate may affect the probability of exceeding critical wind speeds expected to cause wind damage within one northern and one southern study area in Sweden, respectively. The topography of the study areas was relatively gentle and the forests were dominated by Norway spruce (Picea abies (L.) Karst.) and Scots pine (Pinus sylvestris L.). Using estimated changes in the net primary production (NPP) due to climate change and assuming a relative change in the site productivity equal to a relative change in NPP, we simulated possible future states of the forest under gradual adjustment of the site index in response to climate change using the model The Forest Time Machine. Global climate change scenarios based on two emission scenarios and one general circulation model were downscaled to the regional level. The modified WINDA model was used to calculate the sensitivity of the forest to wind and the probability of wind damage for individual forest stands for the periods 2011–2041 and 2071–2100 and for a control period 1961–1990. This was done while taking into account effects on stability of the forest from expected changes in the occurrence of ground frost. Increasing sensitivity of the forest to wind was indicated for both study areas when adhering to recommended management rules of today. Adding also a changed wind climate further increased the probability of wind damage. Calculated probabilities of wind damage were generally higher in the southern study area than in the northern one and were explained by differences in wind climate and the state of the forests, for example with respect to tree species composition. The indicated increase in sensitivity of the forest to wind under the current management regime, and possibly increasing windiness, motivate further analysis of the effects of different management options on the probability of wind damage and what modifications of Swedish forest management are possibly warranted.
