    • Counterparts to two vasoactive peptides previously isolated from fibrin(ogen) degraded by plasmin (EC 3.4.21.7) were synthesized by the solid phase procedure. The synthetic undecapeptide (Ser-Gln-Leu-Gln-Lys-Val-Pro-Pro-Glu-Trp-Lys) was isolated in a homogeneous state by chromatography on Sephadex G-25 and DEAE-Sepharose CL-6B and the pentapeptide (Ala-Arg-Pro-Ala-Lys) by chromatography on BioGel P-6 and column zone electrophoresis. The effect of these two peptides and of fifteen analogs to the pentapeptide on microvascular permeability in rat skin was investigated. The two synthetic counterparts were as potent as the natural peptides. With respect to the analogs, the influence of different functional groups was first studied. This was followed by attempts to minimize the active structure, induce or relieve rigidity of the peptide back-bone or otherwise accomplish modifications by a change in chirality at critical positions. Our results show that the tetrapeptide Arg-Pro-Ala-Lys has the same effect on microvascular permeability as the pentapeptide in the assay system used. Basic amino acids at both ends, as well as a proline residue adjacent to the N-terminal amino acid appear important for full or essentially full activity. On the other hand, substitution of the Ala at position 4 with several other amino acids did not result in a significant loss in biological potency.

    • Separation of feed mixtures and its effect on the production traits of growing-finishing pigs was studied in three experiments. In Experiments I and II the effects of different types of equipment to prevent separation in a silo were studied. Experiment III dealt with separation along a feed transport chain. Experiments I and II clearly demonstrated an increase in feed quality variation by filling a silo without any arrangement to reduce separation. The last feed to be extracted from a silo filled in this manner showed an increasing amount of small particles and contents of crude fibre and ash, a decrease in the bulk density and also in the digestibility of nutrients and in the performance of the pigs. Filling a silo equipped with chutes by an auger or pneumatically through a cyclone did not create separation problems, and consequently there were no differences between these treatments in the production experiments. Separation measured by particle size, bulk density or chemical composition was not noticeable along the feed transport chain (Experiment III). There was no effect on performance of the pigs of their location along the chain. © 1982.

    • Angle resolved UV photoemission spectra obtained from Cu(111)-(√3 × √3)R30° Xe and Cu(111)-(√3 × √3)R30° CO samples are compared with spectra obtained from the clean substrate. Interest is focussed on the adsorbate induced changes in the Cu3d band range of initial energies. From the similarity between the spectra observed for the ordered Xe and CO overlayers we conclude that the changes are primarily due to diffraction of the photoexcited Cu3d electrons by the adsorbate. This is supported also by calculated photoelectron energy distributions.

    • Regeneration of severed peripheral nerves is unfortunately often incomplete, due to loss of nerve fibers and neuroma formation. A new approach is presented with the intention of improving the conditions for nerve repair. In the first of the two stages, a pseudosynovial tube is formed around a silicone rubber rod, surrounded by a stainless steel spiral, which was placed in the backs of rats. This tube, in the second stage, is used as a free "tube graft" to bridge gaps of about 10-12 mm lengths in the severed sciatic nerve. The tube was kept open by the metal spiral. Regenerating nerve fibers with their sprouts grew into the initially open space in the tube. A new nerve trunk was formed, comprised of closely packed myelinated and unmyelinated axons, organized into fascicles. Demonstration by electron microscopy and by EMG recording of reinnervation of foot muscles supported successful long-term results. The fascicles were delimited by perineurial and epineurial sheaths and, furthermore, showed signs of maturation. It was also demonstrated that the nerve-fiber regeneration ceased after a few weeks if there was no distal nerve inserted into the tube. The importance of optimizing the interaction between local factors and regenerating nerve fibers for reestablishment of functionally valuable motor units is discussed.

  • A General Method for the Immobilization of Cells with Preserved Viability

    • Microbial, algal, plant and animal cells have been immobilized, with preserved viability, by entrapment in various matrices according to a new bead polymerization technique. The cell polymer/monomer mixture is kept suspended in a hydrophobic phase such as soy, paraffin, or silicon oil, tri-n-butylphosphate, or dibutyphtalate, which is compatible with the cells. The various monomers or polymers tested include agarose, agar, carrageenan, alginate, fibrin, and polyacrylamide. Furthermore, by adjustment of the stirring speed of the suspension, beads of desired diameter can easily be obtained. The entrapped cells are fully viable and biosynthetically active.

    • Applications of quantitative gas chromatography/mass spectrometry (selected ion monitoring) to clinical and ecological microbiology are described. The technique permits determination of selected microbial metabolites and can be used for the rapid diagnosis of, for example, tuberculosis by ion-focusing of specific branch-chain fatty acids. The technique is also used to determine diaminopimelic and muramic acids, and D- and L-amino acids separated by chiral gas chromatography, in the low picogram range. The latter method has the potential to detect and quantitate microbial populations.
