Electrolyte handling in the isolated perfused rat kidney : demonstration of vasopressin V2-receptor-dependent calcium reabsorption

- Background The most profound effect of vasopressin on the kidney is to increase water reabsorption through V-2-receptor (V2R) stimulation, but there are also data suggesting effects on calcium transport. To address this issue, we have established an isolated perfused kidney model with accurate pressure control, to directly study the effects of V2R stimulation on kidney function, isolated from systemic effects. Methods The role of V2R in renal calcium handling was studied in isolated rat kidneys using a new pressure control system that uses a calibration curve to compensate for the internal pressure drop up to the tip of the perfusion cannula. Results Kidneys subjected to V2R stimulation using desmopressin (DDAVP) displayed stable osmolality and calcium reabsorption throughout the experiment, whereas kidneys not administered DDAVP exhibited a simultaneous fall in urine osmolality and calcium reabsorption. Epithelial sodium channel (ENaC) inhibition using amiloride resulted in a marked increase in potassium reabsorption along with decreased sodium reabsorption. Conclusions A stable isolated perfused kidney model with computer-controlled pressure regulation was developed, which retained key physiological functions. The preparation responds to pharmacological inhibition of ENaC channels and activation of V2R. Using the model, the dynamic effects of V2R stimulation on calcium handling and urine osmolality could be visualised. The study thereby provides evidence for a stimulatory role of V2R in renal calcium reabsorption.

- 2017 har varit ett år med en del förändringar. Ny ÖB började vid årsskiftet och en ny avdelningschef på Campus Vallabiblioteket i september. Planering för nya avdelningschefer för Medicinska Bibliotek och Publicerings Infrastruktur har pågått under senare delen av året. Vi har också fått nya ansvariga för Undervisning/kontakbibliotekarieskapet och Forsknings-stöd.Studenthus Valla har varit en kristalliseringspunkt för nytt tänkande för hela biblioteket. Under sommaren flyttade vi en stor del av samlingen på Campus Vallabiblioteket till Bokladan för att skapa utrymme för flera studie-/läsplatser och även prova nya typer av studieplatser. Campus Norrköping Bibliotek skickade också en del av samlingen till Bokladan med samma syften. Sedan har det varit utveckling och provning av t.ex. Pop up-bibliotek, Digimaker, On-Call bibliotekarie, och söktorget.Årets tema var Visa Värdet, som egentligen är en fortsättning av påbörjat arbete mot systematiskt kvalitetsarbete.Ett år med stort engagemang från medarbetare!

- If one looks at the Constitutional Courts and their place in the architectural landscape of a constitution in a Western or Western-like democracy, they appear to lean into politics through their activism in the political game. This leaning appears to go against the (at least theoretical) nature of a court, which has the task of settling legal cases, not dealing with political decisions. In particular, many Constitutional Courts in the Western or Western-like democracies have grown more distant from the legal arena (as the ultimate authority on what constitutes valid law) and closer to the political one (by limiting and expanding the operating space of the political actors and their legislative tools). The basic thesis of this article is that, from a descriptive point of view, Constitutional Courts, though playing a bridging role between the political and legal worlds, are – from an institutional and functional perspective – primarily legal actors. Without a doubt, these Courts play a role in the political game; however, their position as institutional actors should be based upon the direct effects of their decisions (‘outputs’) in the legal arena, rather than on the indirect consequences (‘outcomes’) in the political arena. As a consequence, the article comes to the conclusion that the Constitutional Courts’ primary responsibility ought to be towards the legal community and the paradigms governing its discourse

- There is a broad consensus that better models for assessing the impact of information efforts are needed to inform information and knowledge management and digital preservation. In contrast to measuring the quality of information, less attention has been directed to the assessment of knowledge and information processes as its constituent. Using archaeology and archaeological information as a sample context, the aim of this conceptual paper is to probe in to the evaluation of the impact and usefulness of information by taking into account the practices of how it is produced, managed and used. On a basis of a review and discussion of earlier literature on the impact of information and evaluation and management of information in archaeology, it is proposed that a better understanding of how the impact of information unfolds as a part of its production, management and use could contribute to the development of infrastructures, repositories and procedures for the management of the preservation and use of these resources.

- Generalized Chebyshev iteration (GCI) applied for solving linear equations with nonselfadjoint operators is considered. Sufficient conditions providing the convergence of iterations imposed on the domain of localization of the spectrum on the complex plane are obtained. A minimax problem for the determination of optimal complex iteration parameters is formulated. An algorithm of finding an optimal iteration parameter in the case of arbitrary location of the operator spectrum on the complex plane is constructed for the generalized simple iteration method. The results are applied to numerical solution of volume singular integral equations (VSIEs) associated with the problems of the mathematical theory of wave diffraction by 3D dielectric bodies. In particular, the domain of the spectrum location is described explicitly for low-frequency scattering problems and in the general case. A prospective study of the immune system activation biomarker neopterin and colorectal cancer risk

- Background: Neopterin may be relevant for colorectal cancer (CRC) development, as a biomarker of cellular immune activity exerting pleiotropic effects on cellular ageing, oxidative stress, and inflammation. So far, the association between prediagnostic neopterin and colon and rectal cancer risk has not been evaluated in human populations. Methods: A nested case-control study was conducted within the European Prospective Investigation into Cancer and Nutrition cohort using data on plasma concentrations of total neopterin (T-N, sum of neopterin and 7,8-dihydroneopterin) in 830 incident CRC case patients (561 colon and 269 rectal) matched within risk sets to 830 control participants. A subsequent replication study used data from the Hordaland Health Study, where 173 CRC case patients have been diagnosed among 6594 healthy participants over 12 years of follow-up. Results: After multivariable adjustment for a priori chosen CRC risk factors, a "U-shaped" association of T-N with CRC was revealed. Compared with the second quintile of the T-N distribution, the relative risks for the first, third, fourth, and fifth quintiles were 2.37 (95% CI = 1.66 to 3.39), 1.24 (95% CI = 0.87 to 1.77), 1.55 (95% CI = 1.08 to 2.22), and 2.31 (95% CI = 1.63 to 3.27), respectively. Replication of these associations within the Hordaland Health Study yielded similar results. No differences have been observed when the associations were explored by colon and rectal cancer site (two-sided P-difference = .87) and after excluding case patients diagnosed within the first four follow-up years. Conclusions: These novel findings provide evidence of the role of both suppressed and activated cell-mediated immunity as reflected by prediagnostic T-N concentrations in the development of CRC.
