  • Skeletal muscle PGC-1α1 and KAT enzymes at the intersection between depression and metabolic disease

    • Depression and metabolic diseases are leading causes of disability and major contributors to socioeconomic burden worldwide. Physical activity exerts many beneficial effects that confer direct health improvement in individuals suffering from these disorders. However, the molecular mechanisms underlying the influence of different components of exercise interventions remain unknown. To isolate the impact of skeletal muscle conditioning on stress-induced depression, we used transgenic models that exhibit many of the chronic adaptations to aerobic exercise in skeletal muscle. Here, we show a mechanism by which skeletal muscle PGC-1α1 modulates kynurenine metabolism and mediates resilience to stress-induced depression. PGC-1α1 acts in concert with PPARα/δ transcription factors to induce the expression of kynurenine aminotransferases in skeletal muscle. The activity of this pathway diverts the metabolism of stress-induced kynurenine to kynurenic acid. Since kynurenic acid is unable to cross the blood brain barrier, this peripheral shift protects the brain from stress-induced accumulation of kynurenines. In addition, we further show that skeletal muscle PGC-1α1 and kynurenine aminotransferases are part of the physiological adaptations to aerobic exercise in both rodents and humans. Given that exercise-mediated activation of this pathway leads to peripheral accumulation of kynurenic acid, we evaluated the physiological role of this metabolite in modulating energy homeostasis. Here, we describe that kynurenic acid plays a role in systemic energy homeostasis through the regulation of adipose tissue function and inflammation. Kynurenic acid induces the expression of lipid metabolism, thermogenic and anti-inflammatory gene networks in the white adipose compartment. This reduces body-weight gain and improves glucose tolerance in animals fed a high-fat diet. Mechanistic studies in primary adipocytes show that kynurenic acid activates the G protein-coupled receptor 35. Downstream signaling of this activation is mediated through Ca2+, ERK, CREB and PGC-1α1 stabilization. Finally, activation of GPR35 by kynurenic acid induces the expression of RGS14, which sensitizes β-adrenergic response to specific agonists. In sum, this work uncovers a previously unknown function of PGC-1α1 in skeletal muscle and kynurenic acid in white adipose tissue. Targeting this metabolic node has great potential for the treatment of depression and metabolic diseases such as type-2 diabetes.

    • Objective: To describe and compare the consumption of the main groups and subgroups of vegetables and fruits (V&F) in men and women from the centres participating in the European Prospective Investigation into Cancer and Nutrition (EPIC). Design: Cross-sectional analysis. Dietary intake was assessed by means of a 24-hour dietary recall using computerised interview software and standardised procedures. Crude and adjusted means were computed for the main groups and sub-groups of V&F by centre, separately for men and women. Adjusted means by season, day of the week and age were estimated using weights and covariance analysis. Setting: Twenty-seven centres in 10 European countries participating in the EPIC project. Subjects: In total, 35 955 subjects (13 031 men and 22 924 women), aged 35-74 years, randomly selected from each EPIC cohort. Results: The centres from southern countries had the highest consumption of V&F, while the lowest intake was seen in The Netherlands and Scandinavia for both genders. These differences were more evident for fruits, particularly citrus. However, slightly different patterns arose for some sub-groups of vegetables, such as root vegetables and cabbage. Adjustment for body mass index, physical activity, smoking habits and education did not substantially modify the mean intakes of vegetables and fruits. vegetable and fruit intake follows a south-north gradient in both Conclusions:. Total, genders, whereas for several sub-groups of vegetables a different geographic distribution exists. Differences in mean intake of V&F by centre were not explained by lifestyle factors associated with VF intake.

    • Hereditary hemochromatosis (HH) is a strong risk factor for hepatocellular cancer, and mutations in the HFE gene associated with HH and iron overload may be related to other tumors, but no studies have been reported for gastric cancer (GC). A nested case-control study was conducted within the European Prospective Investigation into Cancer and Nutrition (EPIC), including 365 incident gastric adenocarcinoma and 1284 controls matched by center, sex, age and date of blood collection. Genotype analysis was performed for two functional polymorphisms (C282Y/rs1800562 and H63D/rs1799945) and seven tagSNPs of the HFE genomic region. Association with all gastric adenocarcinoma, and according to anatomical localization and histological subtype, was assessed by means of the odds ratio (OR) and 95% confidence interval (CI) estimated by unconditional logistic regression adjusted for the matching variables. We observed a significant association for H63D with OR (per rare allele) of 1.32 (CI = 1.03-1.69). In subgroup analyses, the association was stronger for non-cardia anatomical subsite (OR = 1.60, CI = 1.16-2.21) and intestinal histological subtype (OR = 1.82, CI = 1.27-2.62). Among intestinal cases, two tagSNPs (rs1572982 and rs6918586) also showed a significant association that disappeared after adjustment for H63D. No association with tumors located in the cardia or with diffuse subtype was found for any of the nine SNPs analyzed. Our results suggest that H63D variant in HFE gene seems to be associated with GC risk of the non-cardia region and intestinal type, possibly due to its association with iron overload although a role for other mechanisms cannot be entirely ruled out.

  • Immune persistence and response to booster dose of Vi-DT vaccine at 27.5 months post-first dose

    • Vaccination with typhoid conjugate vaccines (TCV) is a major part of typhoid prevention. However, little is known about long-term immune persistence following vaccination with TCVs. In this phase-2, randomized double-blind trial (NCT03527355), 285 children aged 6–23 months were randomized to one of three groups: (1) the group that received a first dose of Vi polysaccharide conjugated to diphtheria-toxoid (Vi-DT) vaccine followed by an “early booster” at 24 weeks, (2) the group that which received a first dose of Vi-DT followed by a “late booster” at 96 or 110 weeks, and (3) comparator group. Safety and immunogenicity of anti-Vi IgG GMTs were assessed at weeks 0, 4, 24, 28, 60, 96, 110, and 114 since the first dose. Here, we describe persistence of immune responses at weeks 60, 96, 110, and 114 post first dose. The anti-Vi IgG seroconversion rate after 27.5 months of follow-up was 88.16% (95% CI: 79.00, 93.64) in late-booster and 94.76% (95% CI: 86.91, 97.88) in early booster Vi-DT groups (p = 0.081). Whereas anti-Vi IgG GMTs were significantly higher in the early booster group (11.95 [95% CI: 9.65, 14.81]) than prebooster GMTs in the late booster group (5.50 [95% CI: 4.44, 6.80], p < 0.0001). GMT in the late booster group significantly increased to 351.76 (95% CI: 265.01, 466.93) (p < 0.0001) when measured 4 weeks after they received their “late-booster” shot. In conclusion, late booster dosing with Vi-DT at 27.5 months post first dose was safe and elicited robust anti-Vi IgG immune responses. Anti-Vi IgG seroconversion rates were persistently comparable in early and late-booster Vi-DT groups.

  • Cortical Atrophy is Associated with Accelerated Cognitive Decline in Mild Cognitive Impairment with Subsyndromal Depression

    • Objectives: To investigate the association between cognitive decline and cortical atrophy in individuals with mild cognitive impairment (MCI) and chronic subsyndromal symptoms of depression (SSD) over a 4-year period. Design: Prospective cohort study. Setting: Multicenter, cwere observed for rate of decline on measures of attention, learning, and confrontation naming or for rate of atrophy in any other regions. Accelerated frontal lobe and anterior cingulate atrophy was associated with cognitive decline on measures of global cognition, information processing speed, and semantic fluency (all p < 0.05), but not memory. Conclusions: Individuals with chronic SSD may represent an MCI subgroup that is highly vulnerable to accelerated cognitive decline, an effect that may be governed by frontal lobe and anterior cingulate atrophy.linic-based. Participants: Within the Alzheimer's Disease Neuroimaging Initiative repository, the Neuropsychiatric Inventory was used to identify individuals with MCI and stable endorsement (SSD group N = 32) or no endorsement (non-SSD group N = 69) of depressive symptoms across time points. Measurements: Repeated measures of cognitive outcomes, cortical atrophy, and their associations were evaluated with mixed effects models adjusting for age, education, sex, and APOE genotype. Results: The SSD group demonstrated accelerated decline on measures of global cognition (Alzheimer Disease Assessment Scale; df = 421, t = 2.242, p = 0.025), memory (Wechsler Memory Scale-Revised Logical Memory II; df = 244, t = -2.525, p = 0.011), information processing speed (Trail Making Test Parts A [df = 421, t = 2.376, p = 0.018] and B [df = 421, t = 2.533, p = 0.012]), and semantic fluency (Category Fluency; df = 424, t = -2.418, p = 0.016), as well as accelerated frontal lobe (df = 341, t = -2.648, p = 0.008) and anterior cingulate (df = 341, t = -3.786, p < 0.001) atrophy. No group differences

    •  The characteristics of policy-based management make it an interesting candidate for managing Ambient Networks, which are characterized for being highly dynamic and heterogeneous. However, current policy-based approaches are not scalable, which is a must for such dynamic scenarios. A key aspect for developing scalable systems is policy distribution, the mechanism that provides the right policies at the right locations in the network when they are needed. In this paper, we present a scalable framework for policy distribution for Ambient Networks. The framework is based on aggregating the addresses of the policies and applying multipoint communication techniques. The aggregation is based on grouping the managed elements by the role they play in the network and distributing policies that apply to all the elements in a group. We show the validity of the framework by applying it to a study case.

  • The ALMA Discovery of the Rotating Disk and Fast Outflow of Cold Molecular Gas in NGC 1275

    • We present observations using the Atacama Large Millimeter/submillimeter Array of the CO(2-1), HCN(3-2), and HCO+(3-2) lines in the nearby radio galaxy/brightest cluster galaxy (BCG) NGC 1275 with a spatial resolution of similar to 20 pc. In previous observations, the CO(2-1) emission was detected as radial filaments lying in the east west direction on a kiloparsec scale. We resolved the inner filament and found that it cannot be represented by a simple infalling stream on a sub-kiloparsec scale. The observed complex nature of the filament resembles the cold gas structure predicted by numerical simulations of cold chaotic accretion. Within the central 100 pc, we detected a rotational disk of molecular gas whose mass is similar to 10(8) M-circle dot. This is the first evidence of the presence of a massive cold gas disk on this spatial scale for BCGs. A crude estimate suggests that the accretion rate of the cold gas can be higher than that of hot gas. The disk rotation axis is approximately consistent with the radio-jet axis. This probably suggests that the cold gas disk is physically connected to the innermost accretion disk, which is responsible for jet launching. We also detected absorption features in the HCN(3-2) and HCO+(3-2) spectra against the radio continuum emission mostly radiated by a jet of size similar to 1.2 pc. The absorption features are blueshifted from the systemic velocity by similar to 300-600 km s(-1), suggesting the presence of outflowing gas from the active galactic nucleus (AGN). We discuss the relation of the AGN feeding with cold accretion, the origin of blueshifted absorption, and an estimate of the black hole mass using molecular gas dynamics.

    • It is the aim of the present study to assess factors associated with time spent in class among working college students. Eighty-two working students from 21 to 26 years old participated in this study. They were enrolled in an evening course of the University of São Paulo, Brazil. Participants answered a questionnaire on living and working conditions. During seven consecutive days, they wore an actigraph, filled out daily activity diaries (including time spent in classes) and the Karolinska Sleepiness Scale every three hours from waking until bedtime. Linear regression analyses were performed in order to assess the variables associated with time spent in classes. The results showed that gender, sleep length, excessive sleepiness, alcoholic beverage consumption (during workdays) and working hours were associated factors with time spent in class. Thus, those who spent less time in class were males, slept longer hours, reported excessive sleepiness on Saturdays, worked longer hours, and reported alcohol consumption. The combined effects of long work hours (>40 h/week) and reduced sleep length may affect lifestyles and academic performance. Future studies should aim to look at adverse health effects induced by reduced sleep duration, even among working studentswho spent more time attending evening classes.
