    • Previous studies on wake flow visualization of live animals using DPIV have typically used low repetition rate lasers and 2D imaging. Repetition rates of around 10 Hz allow similar to 1 image per wingbeat in small birds and bats, and even fewer in insects. To accumulate data representing an entire wingbeat therefore requires the stitching-together of images captured from different wingbeats, and at different locations along the wing span for 3D-construction of wake topologies. A 200 Hz stereo DPIV system has recently been installed in the Lund University wind tunnel facility and the high-frame rate can be used to calculate all three velocity components in a cube, whose third dimension is constructed using the Taylor hypothesis. We studied two bat species differing in body size, Glossophaga soricina and Leptonycteris curasoa. Both species shed a tip vortex during the downstroke that was present well into the upstroke, and a vortex of opposite sign to the tip vortex was shed from the wing root. At the transition between upstroke/downstroke, a vortex loop was shed from each wing, inducing an upwash. Vorticity iso-surfaces confirmed the overall wake topology derived in a previous study. The measured dimensionless circulation, I"/Uc, which is proportional to a wing section lift coefficient, suggests that unsteady phenomena play a role in the aerodynamics of both species.

  • White clover pollinators and seed set in relation to local management and landscape context

    • Bees are declining, which is worrisome since they both have intrinsic conservation value and play a major role as pollinators in both natural and managed ecosystems. Land use change and lack of suitable habitats are often suggested as driving forces of bee decline. To propose mitigation measures to halt bee decline, it is important to understand how land use relates to bee abundance and diversity, and to explore consequences for their provision of pollination services. White clover, Trifolium repens, is an outcrossing mass-flowering crop, which could serve as an abundant, although ephemeral, food resource for bees. We investigated how the bee community in 39 fields of white clover grown for seed, related to local field management (organic, conventional without insecticides and conventional with insecticides) and landscape context (proportion semi-natural land), and how this pollinator community related to white clover seed set. The honey bee, Apis mellifera, was the most commonly observed bee species, and two generalist bumble bee species, Bombus terrestris and B. lapidarius, were the subsequently most common. We observed fewer non-Apis bees, and a lower bee species richness in organic white clover seed fields compared to conventional fields independent of insecticide treatment. Bee species richness in both conventional and organic fields were positively related to the proportion of semi-natural land in the landscape, likely because of a larger species pool in such landscapes. Initial seed set in immature inflorescences was positively related to bee abundance, whereas final seed set in mature inflorescences was unrelated to bee abundance, possibly as a consequence of seed-eating weevils consuming a large proportion of the seeds. We conclude that both bee visitation and seed set in white clover benefit from conventional management and that landscapes rich in semi-natural habitats will make future crop production more resilient. The observed positive relationship between bee abundance and initial seed set suggests that if we can mitigate pest impacts and increase bee abundance in clover seed fields, the final seed yield can be increased. Thus, bee decline should be considered and mitigated both to maintain biodiversity in general and for crop seed production specifically.

  • Measuring power input, power output and energy conversion efficiency in un-instrumented flying birds

    • Cost of flight at various speeds is a crucial determinant of flight behaviour in birds. Aerodynamic models, predicting that mechanical power (Pmech) varies with flight speed in a U-shaped manner, have been used together with an energy conversion factor (efficiency) to estimate metabolic power (Pmet). Despite few empirical studies, efficiency has been assumed constant across flight speeds at 23%. Ideally, efficiency should be estimated from measurements of both Pmech and Pmet in un-instrumented flight. Until recently, progress has been hampered by methodological constraints. The main aim of this study was to evaluate recently developed techniques and estimate flight efficiency across flight speeds. We used the 13C-labelled sodium bicarbonate method (NaBi) and particle image velocimetry (PIV) to measure Pmet and Pmech in blackcaps flying in a wind tunnel. We also cross-validated measurements made by NaBi with quantitative magnetic resonance (QMR) body composition analysis in yellow-rumped warblers. We found that Pmet estimated by NaBi was ∼12% lower than corresponding values estimated by QMR. Pmet varied in a U-shaped manner across flight speeds in blackcaps, but the pattern was not statistically significant. Pmech could only be reliably measured for two intermediate speeds and estimated efficiency ranged between 14% and 22% (combining the two speeds for raw and weight/lift-specific power, with and without correction for the ∼12% difference between NaBi and QMR), which were close to the currently used default value. We conclude that NaBi and PIV are viable techniques, allowing researchers to address some of the outstanding questions regarding bird flight energetics.

  • Structural properties of recombinant nonrepetitive and repetitive parts of major ampullate spidroin 1 from Euprosthenops australis : implications for fiber formation

    • Spider dragline silk proteins, spidroins, have a tripartite composition; a nonrepetitive N-terminal domain, a central repetitive region built up from many iterated poly-Ala and Gly rich blocks, and a C-terminal nonrepetitive domain. It is generally believed that the repetitive region forms intermolecular contacts in the silk fibers, while precise functions of the terminal domains have not been established. Herein, thermal, pH, and salt effects on the structure and aggregation and/or polymerization of recombinant N- and C-terminal domains, a repetitive segment containing four poly-Ala and Gly rich coblocks, and combinations thereof were studied. The N- and C-terminal domains have mainly alpha-helical structure, and interestingly, both form homodimers. Dimerization of the end domains allows spidroin multimerization independent of the repetitive part. Reduction of an intersubunit disulfide in the C-terminal domain lowers the thermal stability but does not affect dimerization. The repetitive region shows helical secondary structure but appears to lack stable folded structure. A protein composed of this repetitive region linked to the C-terminal domain has a mainly alpha-helical folded structure but shows an abrupt transition to beta-sheet structures upon heating. At room temperature, this protein self-assembles into macroscopic fibers within minutes. The secondary structures of none of the domains are altered by pH or salt. However, high concentrations of phosphate affect the tertiary structure and accelerate the aggregation propensity of the repetitive region. Implications of these results for dragline spidroin behavior in solution and silk fiber formation are discussed.

  • Single-cell quantification and dose-response of cytosolic siRNA delivery

    • Endosomal escape and subsequent cytosolic delivery of small interfering RNA (siRNA) therapeutics is believed to be highly inefficient. Since it has not been possible to quantify cytosolic amounts of delivered siRNA at therapeutic doses, determining delivery bottlenecks and total efficiency has been difficult. Here, we present a confocal microscopy-based method to quantify cytosolic delivery of fluorescently labeled siRNA during lipid-mediated delivery. This method enables detection and quantification of sub-nanomolar cytosolic siRNA release amounts from individual release events with measures of quantitation confidence for each event. Single-cell kinetics of siRNA-mediated knockdown in cells expressing destabilized eGFP unveiled a dose-response relationship with respect to knockdown induction, depth and duration in the range from several hundred to thousands of cytosolic siRNA molecules. Accurate quantification of cytosolic siRNA, and the establishment of the intracellular dose-response relationships, will aid the development and characterization of novel delivery strategies for nucleic acid therapeutics.

    • In a retrospective study of 51 consecutive patients undergoing transcatheter closure of secundum type atrial septal defects (ASDs), we examined the reliability of transesophageal echocardiography (TEE) prior to catheterization and compared the diameter with that obtained by balloon measurement during catheterization. The TEE diameter was 16.3+/-4.6 mm compared with 22.5+/-6.0 mm for the stretched diameter obtained during catheterization (p<0.001). There was no gender difference. The degree of left-to-right shunting correlated poorly with the size of the defect. We conclude that although TEE is accurate for diagnosis of an ASD, the measurement of its size to determine the size of the closure device is at best inaccurate.

  • Tracking the intrusion of unwanted memories into awareness with event-related potentials

    • Involuntary retrieval of unwanted memories is a common symptom in several clinical disorders, including post-traumatic stress disorder. With an aim to track the temporal dynamics of such memory intrusions, we recorded electrophysiological measures of brain activity while participants engaged in a Think/No-Think task. We presented the left hand word (the cue) of previously encoded word pairs in green or red font. We asked participants to think of the associated right hand word (the associate) when the cue appeared in green (Think condition) and to avoid thinking of the associate when the cue appeared in red (No-Think condition). To isolate cases when participants experienced an intrusive memory, at the end of each trial, participants judged whether the response had come to mind; we classified memories that came to mind during No-Think trials, despite efforts to stop retrieval, as intrusions. In an event-related potential (ERP) analysis, we observed a negative going slow wave (NSW) effect that indexed the duration of a trace in mnemonic awareness; whereas voluntary retrieval and maintenance of the associate was related to a sustained NSW that lasted throughout the 3-s recording epoch, memory intrusions generated short-lived NSWs that were rapidly truncated. Based on these findings, we hypothesize that the intrusion-NSW reflects the associate briefly penetrating working memory. More broadly, these findings exploit the high temporal resolution of ERPs to track the online dynamics of memory intrusions.
