    • We present the design and performance characteristics of a platelet analysis platform based on a microfluidic impedance cytometer. Dielectrophoretic focusing is used to centre cells in a fluid stream, which then forms the core of a two-phase flow (dielectric focusing). This flow then passes between electrodes for analysis by differential impedance spectroscopy at multiple frequencies from 280 kHz to 4 MHz. This approach increases the signal-to-noise ratio relative to a single-phase, unfocused stream, while minimising the shear forces to which the cells are subjected. The percentage of activated platelets before and after passage through the chip was measured using flow cytometry, and no significant change was measured. Measuring the in-phase amplitude at a single frequency is sufficient to distinguish platelets from erythrocytes. Using multi-frequency impedance measurements and discriminant analysis, resting platelets can be discriminated from activated platelets. This multifrequency impedance cytometer therefore allows ready determination of the degree of platelet activation in blood samples.

    • Multiplexed spatial profiling of mRNAs has recently gained traction as a tool to explore the cellular diversity and the architecture of tissues. We propose a sensitive, open-source, simple and flexible method for the generation of in situ expression maps of hundreds of genes. We use direct ligation of padlock probes on mRNAs, coupled with rolling circle amplification and hybridization-based in situ combinatorial barcoding, to achieve high detection efficiency, high-throughput and large multiplexing. We validate the method across a number of species and show its use in combination with orthogonal methods such as antibody staining, highlighting its potential value for developmental and tissue biology studies. Finally, we provide an end-to-end computational workflow that covers the steps of probe design, image processing, data extraction, cell segmentation, clustering and annotation of cell types. By enabling easier access to high-throughput spatially resolved transcriptomics, we hope to encourage a diversity of applications and the exploration of a wide range of biological questions.

    • Cancer and bacterial infection are the most serious problems threatening people's lives worldwide. However, the overuse of antibiotics as antibacterial and anticancer treatments can cause side effects and lead to drug-resistant bacteria. Therefore, developing natural materials with excellent antibacterial and anticancer activity is of great importance. In this study, different concentrations of chitosan (CS), graphene oxide (GO), and graphene oxide-chitosan composite (GO-CS) were tested to inhibit the bacterial growth of gram-positive (Bacillus cereus MG257494.1) and gram-negative (Pseudomonas aeruginosa PAO1). Moreover, we used the most efficient natural antibacterial material as an anticancer treatment. The zeta potential is a vital factor for antibacterial and anticancer mechanism, at pH 3–7, the zeta potential of chitosan was positive while at pH 7–12 were negative, however, the zeta potential for GO was negative at all pH values, which (p < 0.05) increased in the GO-CS composite. Chitosan concentrations (0.2 and 1.5%) exhibited antibacterial activity against BC with inhibition zone diameters of 4 and 12 mm, respectively, and against PAO1 with 2 and 10 mm, respectively. Treating BC and PAO1 with GO:CS (1:2) and GO:CS (1:1) gave a larger (p < 0.05) inhibition zone diameter. The viability and proliferation of HeLa cells treated with chitosan were significantly decreased (p < 0.05) from 95.3% at 0% to 12.93%, 10.33%, and 5.93% at 0.2%, 0.4%, and 0.60% concentrations of chitosan, respectively. Furthermore, CS treatment increased the activity of the P53 protein, which serves as a tumor suppressor. This study suggests that chitosan is effective as an antibacterial and may be useful for cancer treatment.

  • Influence of structural damping on sound radiation from a point-excited beam

    • The track is the main noise source of a railway when the frequency is below about 1250 Hz. Quite a few efforts have been made to increase the attenuation of the railway track in order to reduce the radiated sound. The attempt of this paper is to investigate the influence of the structural damping on the sound pressure level at the area close to the excitation point instead of the total radiated sound power. For this purpose, the simplest model, i.e. an infinite beam, is adapted. Both near field solution and the propagating wave are included. Results show that when the sound pressure levels at a certain areas close to the excitation is concerned only, the influence of the structural damping is somewhat lower than what expected on the total sound power, partly due to the contribution of the near field solution and partly due to the wave propagation. If the area concerned is small, this influence is also dependent on frequency, because of the length of the bending wave. It is difficult to estimate the influence of the structural damping on wayside noise from laboratory tests with a short track and a point excitation.

  • Mass Spectrometry Proteotyping of Streptococcus pneumoniae and commensal Streptococcus: identification of biomarkers for infectious strain characterization

    • Background: Streptococcus pneumoniae (pneumococcus) is the leading cause of community-acquired pneumonia, with morbidity and mortality worldwide. S. pneumoniae belongs to the S. mitis-Group (viridans streptococci), phenotypically and genotypically similar to commensal species of the upper respiratory tract, S. mitis, S. oralis, and S. pseudopneumoniae, causing problems for identifications in clinical laboratories. In this project, we apply state-of-the-art proteomics for Streptococcus spp. 'proteotyping'; identifying and characterizing protein biomarkers for species-level identification, antibiotic resistance, virulence and strain typing for epidemiological analyses (1). Material/methods: Bacterial proteins, from intact bacteria or cell fractions, are bound to a membrane surface, using patented (WO2006068619) FlowCell (LPITM) technology. Peptides are generated from the bound proteins, by enzymatic digestion, separated and analyzed, using LC-MS/MS. The mass spectra profiles are compared to reference peptide sequences and whole genome sequence (wgs) data of the NCBI RefSeq Database. The S. mitis-Group specie, S. pneumoniae, S. mitis, S. oralis, S. psedopneumoniae, as well as the more distantly-related, Group A Streptococcus (GAS) species, S. pyogenes , were analyzed individually and in mixtures, to demonstrate the resolution of proteotyping for differentiating bacteria. Results: Using proteotyping protocols, S. pneumoniae were detected and differentiated from other streptococci, S. mitis, S. oralis, S. psedopneumoniae and the more distant relative, S. pyogenes, by identification of unique discriminatory peptides. Metabolic protein biomarkers were identified, including for antibiotic resistance and virulence. It was possible to find discriminatory biomarkers for a target species when analyzing 1:1 mixes of S. pneumoniae and other species from the S. mitis-Group. The different strains of S. pneumoniae, analyzed in different ratio combinations, were successfully differentiated and identified. For successful proteotyping, a comprehensive and accurate genomic database was observed to be key for obtaining reliable peptide matching and proteotyping data. Importantly, because of observed high rates of misclassified wgs data in the public databases, the taxonomic classifications of genomes in GenBank were analyzed against reference type strain genomes of target species by calculating wgs similarities, using Average Nucleotide Identity with BLAST (ANIb). While wgs data for S. pneumoniae were confirmed to be classified correctly, approximately one-third of wgs data for other species of the S. mitis-Group were determined to be misclassified. Streptococci strains that could not be identified, using standard genotypic and phenotypic approaches, were characterized by proteotyping and genome sequencing to establish their taxonomy and biomarker features to enhance species database matching. Conclusions: Proteotyping enables differentiation, identification and characterization of pneumococcus from the most closely related species attaining, as well, strain-level discrimination from single LC-MS/MS analyses. The protocol enhances identification and characterization of pathogenic bacterial isolates through identifications of expressed biomarkers, ultimately for cultivation-independent analyses of clinical samples. 1) Karlsson et al., 2015.     • Device-independent quantum key distribution (DI-QKD) offers the strongest form of security against eavesdroppers bounded by the laws of quantum mechanics. However, a practical implementation is still pending due to the requirement of combinations of visibility and detection efficiency that are beyond those possible with current technology. This mismatch motivates the search for DI-QKD protocols that can close the gap between theoretical and practical security. In this work, we present two DI-QKD protocols whose security relies on Bell inequalities with more than two inputs and two outputs. We show that, for maximally entangled states and perfect visibility, a protocol based on a Bell inequality with three inputs and four outputs requires a slightly lower detection efficiency than the protocols based on Bell inequalities with two inputs and two outputs.

  • Steering Magnetic Skyrmions with Currents : A Nonequilibrium Green's Functions Approach

    • Magnetic skyrmions, topologically protected vortex-like configurations in spin textures, are of wide conceptual and practical appeal, notably in relation to the making of so-called race-track memory devices. Skyrmions can be created, steered, and destroyed with magnetic fields and/or (spin) currents. Here the authors focus on the latter mechanism, analyzed via a microscopic treatment of the skyrmion–current interaction. The system considered is an isolated skyrmion in a square-lattice cluster, interacting with electron spins in a current-carrying quantum wire. For the theoretical description, a quantum formulation of spin-dependent currents via nonequilibrium Green's functions (NEGF) within the generalized Kadanoff–Baym ansatz (GKBA) is employed. This is combined with a treatment of skyrmions based on classical localized spins, with the skyrmion motion described via Ehrenfest dynamics. With the mixed quantum–classical scheme, the authors assess how time-dependent currents can affect the skyrmion dynamics, and how this in turn depends on electron–electron and spin–orbit interactions in the wire. This study shows the usefulness of a quantum–classical treatment of skyrmion steering via currents, as a way for example to validate/extract an effective, classical-only, description of skyrmion dynamics from a microscopic quantum modeling of the skyrmion–current interaction.

  • Extracting band structure characteristics of GaSb/InAs core-shell nanowires from thermoelectric properties

    • Nanowires with a GaSb core and an InAs shell (and the inverted structure) are interesting for studies of electron-hole hybridization and interaction effects due to the bulk broken band-gap alignment at the material interface. We have used eight-band k·p theory together with the envelope function approximation to calculate the band structure of such nanowires. For a fixed core radius, as a function of shell thickness the band structure changes from metallic (for a thick shell) to semiconducting (for a thin shell) with a gap induced by quantum confinement. For intermediate shell thickness, a different gapped band structure can appear, where the gap is induced by hybridization between the valence band in GaSb and the conduction band in InAs. To establish a relationship between the nanowire band structures and signatures in thermoelectrical measurements, we use the calculated energy dispersions as input to the Boltzmann equation and to ballistic transport equations to study the diffusive limit and the ballistic limit, respectively. Our theoretical results provide a guide for experiments, showing how thermoelectric measurements in a gated setup can be used to distinguish between different types of band gaps, or tune the system into a regime with few electrons and few holes, which can be of interest for studies of exciton physics.

    • There is a considerable amount of literature dealing with inference about the parameters in a heteroscedastic one-way random-effects ANOVA model. In this paper, we primarily address the problem of improved quadratic estimation of the random-effect variance component. It turns out that such estimators with a smaller mean squared error compared with some standard unbiased quadratic estimators exist under quite general conditions. Improved estimators of the error variance components are also established.
