  • Fish bile used to detect estrogenic substances in treated sewage water

    • Endocrine disrupting effects on fish associated with sewage treatment effluents have been demonstrated in several studies. To investigate if the effluents from two modem Swedish sewage treatment plants contained estrogenic substances, juvenile rainbow trout were exposed to sewage water before and after the last treatment step which is a sand filter. As a biomarker for estrogenic effect, vitellogenin was analysed in the blood plasma of the exposed fish. To identify substances possibly responsible for the effect, bile fluid from the exposed fish were analysed with GUMS. Elevated levels of vitellogenin were only seen in the fish exposed at one of the sewage treatment plants, the one with shorter residence time in the biological treatment steps, which suggests that the residence time is of importance for the ability to reduce the amount of estrogenic substances in the sewage water. The highest elevation of vitellogenin was seen in the fish exposed to water before the sand filter, which indicates that the sand filter contributes to further reduction of estrogenic substances in the sewage water. In bile from the same group of fish, considerably higher concentrations of estrone, bisphenol A and 4-nonylphenol (4.0 mu g/g bile, 23 mu g/g bile and 24 mu g/g bile, respectively) were observed compared to bile from control fish (< 0.04 mu g/g bile, 0.21 mu g/g bile, and 3.5 mu g/g bile, respectively). The more potent steroidal estrogens were suggested to be major contributors to the observed estrogenic effect, although xenoestrogens were detected at higher concentrations in the bile fluid.   • Effects of the brominated flame retardants hexabromocyclododecane (HBCDD), and tetrabromobisphenol A (TBBPA), on hepatic enzymes and other biomarkers in juvenile rainbow trout and feral eelpout

    • Brominated flame retardants (BFRs) leak out in the environment, including the aquatic one. Despite this, sublethal effects of these chemicals are poorly investigated in fish. In this study, a screening of selected biomarkers in juvenile rainbow trout (Oncorhynchus mykiss) and feral eelpout (Zoarces viviparus) was performed after exposure to hexabromocyclododecane (HBCDD) and tetrabromobisphenol A (TBBPA). Rainbow trout was injected intraperitoneally (i.p.) with HBCDD or TBBPA. Two out of four short-term experiments with HBCDD showed an increase in the activity of catalase. A 40% increase in liver somatic index (LSI) could be observed after 28 days. HBCDD did also seem to have an inhibitory effect on CYPIA's activity (ethoxyresorufin-O-deethylase (EROD)). A putative peroxisome proliferating activity of the compound was investigated without giving a definite answer. HBCDD did not seem to be estrogenic or genotoxic. TBBPA increased the activity of glutathione reductase (GR) after 4, 14 and 28 days in rainbow trout suggesting a possible role of this compound in inducing oxidative stress. The compound did not seem to be estrogenic. TBBPA seemed to compete with the artificial substrate ethoxyresorufin in vitro, during the EROD assay. In eelpout, only one 5 days in vivo experiment was performed. Neither of the compounds gave rise to any effect in this fish. This was the first screening of sublethal effects of the two chemicals in fish, using high doses. Our results indicate that there is a need for further studies of long-term, low-dose effects of these two widely used flame retardants. 2004 Elsevier B.V. All rights reserved.

    • The eelpout (Zoarces viviparus) is used in Swedish and German environmental monitoring of pollutant effects in fish and is being considered for the same purpose in other countries, mainly around the Baltic Sea. The activities of several hepatic biotransformation and antioxidant enzymes were monitored during the period of 12 months from August 1996 to July 1997, using newly caught feral eelpout from Kattegat, off the west coast of Sweden. In females, the biotransformation enzyme activities showed significant seasonal variations, although the amplitude of the changes was limited; CYP1A-activity, measured as ethoxyresorufin-O-deethylase (EROD), was highest in February-March and decreased afterwards, possibly due to rising estrogen levels. UDP-glucuronosyltransferase activity was lowest in November, while the glutathione-S-transferase activity markedly decreased during the winter. The antioxidant enzymes, catalase, glutathione peroxidase and glutathione reductase showed small variations in their activity. The number of male fish was limited and the seasonal variations less clear. However, in both sexes, glutathione reductase and peroxidase activities were highest in autumn and May, respectively. This study establishes a baseline for the activities of the selected enzymes in female eelpout, which is important for the environmental monitoring using this fish species.

    • Several biomarkers in female and male eelpout (Zoarces viviparus) were measured at Fjällbacka, a reference site on the Swedish west coast from 1989 to 2001. Standardised procedures for sampling and measurements were used. Some of the biomarkers were largely stable through the years, for example, condition factor, blood plasma chloride concentration, white blood cell count, percentage of lymphocytes, activities of glutathione-S-transferase and glutathione reductase. Others showed greater variations but not significant trends, for example, percentage of granulocytes and activity of catalase and ethoxyresorufin-O-deethylase activity. Three variables in female eelpout showed a statistically significant downward trend from 1989 to 2001, hematocrit, body weight and length. The change in hematocrit was probably related to decreased stress due to improved sampling techniques, but the reason for changes in weight and length remain unknown. There is a need to evaluate the area for pollutants. The results indicate that there have not been any major large-scale changes in the aquatic environment affecting the fish health at the investigated site

    • Treated sewage effluents contain complex mixtures of micropollutants, raising concerns about effects on aquatic organisms. The addition of advanced treatment steps has therefore been suggested. However, some of these could potentially produce effluents affecting exposed organisms by unknown modes of action. Here, (1)H NMR (proton nuclear magnetic resonance spectroscopy) metabolomics of fish blood plasma was used to explore potential responses not identified by more targeted (chemical or biological) assays. Rainbow trout was exposed in parallel to six differently treated effluents (e.g., conventional activated sludge, addition of sand filter, further addition of ozonation and/or a moving bed biofilm reactor or a separate membrane bioreactor line). Multivariate data analysis showed changes in the metabolome (HDL, LDL, VLDL and glycerol-containing lipids, cholesterol, glucose, phosphatidylcholine, glutamine, and alanine) between treatment groups. This formed the basis for postulating a hypothesis on how exposure to effluent treated by certain processes, including ozonation, would affect the metabolic profiles of exposed fish. The hypothesis withstood testing in an independent study the following year. To conclude, (1)H NMR metabolomics proved suitable for identifying physiological responses not identified by more targeted assays used in parallel studies. Whether these changes are linked to adverse effects remains to be tested.

    • Nuclear magnetic resonance (NMR) based metabolomics in combination with multivariate data analysis may become valuable tools to study environmental effects of pharmaceuticals and other chemicals in aquatic organisms. To explore the usefulness of this approach in fish, we have used H-1 NMR metabolomics to compare blood plasma and plasma lipid extracts from rainbow trout exposed to the synthetic contraceptive estrogen ethinylestradiol (EE2) with plasma from control fish. The plasma metabolite profile was affected in fish exposed to 10 ng/L but not 0.87 ng/L of EE2, which was in agreement with an induced vitellogenin synthesis in the high dose group only, as measured by ELISA. The main affected metabolites were vitellogenin, alanine, phospholipids and cholesterol. The responses identified by this discovery-driven method could be put in context with previous knowledge of the effects of estrogens on fish. This adds confidence to the approach of using NMR metabolomics to identify environmental effects of pharmaceuticals and other contaminants. (c) 2006 Elsevier B.V. All rights reserved.

  • Precautionary defaults - A new strategy for chemical risk management

    • In order to give adequate support to risk managers, new risk assessment methods should be developed that are (1) scientifically sound, (2) simplified, and (3) suited for precautionary risk management. In this Perspective we propose that the notion of a precautionary default can be a useful tool in the development of such methods. A precautionary default is a cautious or pessimistic assumption that is used in the absence of adequate information and that should be replaced when such information is obtained. Furthermore, we point out some promising research areas for the development of such indicators, viz. connections between chemical characteristics such as persistence and effect parameters, monitoring of contaminants in polar regions, monitoring of contaminants in breast milk, application of results from (human) toxicology in ecotoxicology and vice versa, (eco) toxicological test systems that are sensitive to effects on reproduction, and the application of bioinformatic methods to complex data, both in genomic research and in ecotoxicology. We conclude that precautionary decision-making does not require less science, but to the contrary it requires more science and improved communication between scientists and risk managers.

  • Three decades of Swedish experience demonstrates the need for integrated long-term monitoring of fish in marine coastal areas

    • The first attempts to monitor coastal fish in Sweden were made in the 1960s and 1970s. Ecological, physiological, biochemical and environmental chemistry data were collected in separate projects. When the National Marine Monitoring Programme was revised in 1992, a new strategy was introduced for assessments of long-term trends in coastal fish communities. Annual integrated monitoring of contaminants, biomarkers and population and community indicators of ecosystem health was started in selected areas using common sentinel species. Data from one monitoring area at the coast of the Baltic proper are analyzed in this paper. The results have shown a shift in fish community structure indicating changes in ecosystem productivity. Trends have been detected in growth rate (positive) and relative gonad size (negative) in perch (Perca fluviatilis), suggesting a metabolic disturbance according to the predictive response model developed for interpretations. One factor which may have contributed to the reduced GSI was a decrease in mean age of sampled fish during the period of study. Chemical exposure was indicated by a 3-fold increase of EROD activity during the monitored 15-year period. However, concentrations of most measured contaminants in perch have decreased during the same period. The experience of the integrated approach has shown that a tentative analysis of cause and environmental significance could be made, improving the assessment, but there still remain unsolved questions to be answered in follow-up studies. The analysis has also shown the importance of long-term monitoring at several levels of biological organization to distinguish between natural variation and low-level effects on ecosystems.

    • Rubber tire material contains toxic compounds including oils rich in polycyclic aromatic hydrocarbons (PAH), so-called highly aromatic (HA) oils, as well as other reactive additives used as antioxidants, antiozonants, and vulcanization accelerators. The toxicity of rubber tire leachates to aquatic organisms has been demonstrated before. However, previous Studies have focused on lethal rather than sublethal effects. We kept rainbow trout (Oncorhynchus mykiss) in tanks with two types of fires: a tire containing HA oils in the tread or a tire free of HA oils in the tread. After 1 d of exposure. an induction of cytochrome P4501A1 (CYP1A1) was evident in both exposed groups, measured as elevated ethoxyresorufin-O-deethylase (EROD) activity and increased CYP1A1 mRNA levels. After two weeks of exposure, EROD activity and CYP1A1 mRNA were still high in fish exposed to leachate from HA oil-containing tire, whereas the effect was somewhat lower in fish exposed to leachate from HA oil-free tread tire. Compounds in the tire leachates also affected antioxidant parameters. Total glutathione concentration in liver as well as hepatic glutathione reductase, glutathione S-transferase, and glucose-6-phosphate dehydrogenase activities were markedly elevated after two weeks of exposure in both groups. The responses were greater in the group exposed to leachate from HA oil-free tread tire. Vitellogenin measurements did not indicate leakage of estrogenic compounds from the tires. Chemical analyses of bile from exposed fish revealed the presence of hydroxylated PAH as well as aromatic nitrogen compounds indicating uptake of these compounds by the fish.
