    • OBJECTIVE: Atherosclerotic lesions have regions that are hypoxic. Because the lesion contains macrophages that are loaded with lipid, we investigated whether hypoxia can influence the accumulation of lipids in these cells. METHODS AND RESULTS: Exposure of human macrophages to hypoxia for 24 hours resulted in an increased formation of cytosolic lipid droplets and an increased accumulation of triglycerides. Exposure of the macrophages to oxidized low-density lipoprotein (oxLDL) increased the accumulation of cytosolic lipid droplets because of an increase in cellular cholesterol esters. The accumulation of lipid droplets in oxLDL-treated cells was further increased after hypoxia, caused by an increased level of triglycerides. Expression analyses combined with immunoblot or RT-PCR demonstrated that hypoxia increased the expression of several genes that could promote the accumulation of lipid droplets. Hypoxia increased the mRNA and protein levels of adipocyte differentiation-related protein (ADRP). It is well known that an increased expression of ADRP increases the formation of lipid droplets. Hypoxia decreased the expression of enzymes involved in beta-oxidation (acyl-coenzyme A synthetase and acyl-coenzyme A dehydrogenase) and increased the expression of stearoyl-coenzyme A desaturase, an important enzyme in the fatty acid biosynthesis. Moreover, exposure to hypoxia decreased the rate of beta-oxidation, whereas the accumulation of triglycerides increased. CONCLUSIONS: The results demonstrate that exposure of human macrophages to hypoxia causes an accumulation of triglyceride-containing cytosolic lipid droplets. This indicates that the hypoxia present in atherosclerotic lesions can contribute to the formation of the lipid-loaded macrophages that characterize the lesion and to the accumulation of triglycerides in such lesions.

    • The Swedish infinitive marker "att" is disappearing after "kommer", the major construction for indicating future time. "Kommer" is originally a main verb, but the loss of the infinitive marker of the second verb in the phrase gives it the chief characteristic of an auxiliary, which makes the proecess one of grammaticalization. Based on a survey among Swedish students it is shown that both the age of speakers/writers and the syntax of the sentence in question are crucial factors for the choice between "att" and omission.

    • In Swedish there is no inflectional verb form to indicate future time. To some extent the present tense can be used, but two constructions use auxiliaries. The main forms are traditionally given as skall and kommer att, with the latter cited as having the infinitive marker att of the main verb linked to it, much like the tradition for ought to in English grammar. Bona fide modal auxiliaries in Swedish take an infinitive without the infinitive marker att, and kommer has been developing in that direction for the last few decades. Standard grammars and usage guides still tend to regard the use of kommer plus a bare infinitive as only marginally acceptable. The present article discusses the position today of kommer on a kline where the extremes are the full main verb and the bona fide auxiliary. Corpora of newspaper texts show that the dropping of att after kommer occurred marginally in the period from the 1960s through the 1990s, with strong acceleration starting around 2000. A recent experiment in which Swedish university students translated two sentences from English into Swedish, demonstrated two things very clearly: (1) The dropping of att after kommer is characteristic of young speakers and very much age-related; (2) The dropping of att increases for all age-groups when there is a distance (created by interposed words) between kommer and the infinitive of the main verb.
