  • Enhanced bone healing around nanohydroxyapatite-coated polyetheretherketone implants : An experimental study in rabbit bone

    • Objective: To investigate the bone response to threaded polyetheretherketone (PEEK) implants coated with nanohydroxyapatite. Materials and methods: A total of 39 PEEK implants were coated with nanocrystalline hydroxyapatite and 39 uncoated implants were used as controls. The implant surface was characterized by optical interferometry and scanning electron microscope. The implants were inserted in the tibia and femur of 13 rabbits. After 6 weeks of healing, quantitative and qualitative analyses were performed. Results: The test implants showed significantly higher removal torque test values compared with the control group. Histomorphometric evaluation demonstrated higher bone-to-implant contact for the test implants; however, there were no differences in bone area between the groups. Qualitative histological analyses demonstrated inflammatory cellular reactions in close vicinity of both implant surfaces. A two-cell layer of foreign body giant cells was observed irrespective of sample type. Conclusion: Our findings demonstrate that implants with a threaded design render good stability to PEEK in both coated and uncoated implants. Nanohydroxyapatite-coated PEEK implants demonstrated improved bone formation compared with uncoated controls.

    • 5'-AMP-activated protein kinase (AMPK) is a metabolic stress sensor present in all eukaryotes. A dominant missense mutation (R225Q) in pig PRKAG3, encoding the muscle-specific gamma3 isoform, causes a marked increase in glycogen content. To determine the functional role of the AMPK gamma3 isoform, we generated transgenic mice with skeletal muscle-specific expression of wild type or mutant (225Q) mouse gamma3 as well as Prkag3 knockout mice. Glycogen resynthesis after exercise was impaired in AMPK gamma3 knock-out mice and markedly enhanced in transgenic mutant mice. An AMPK activator failed to increase skeletal muscle glucose uptake in AMPK gamma3 knock-out mice, whereas contraction effects were preserved. When placed on a high fat diet, transgenic mutant mice but not knock-out mice were protected against excessive triglyceride accumulation and insulin resistance in skeletal muscle. Transfection experiments reveal the R225Q mutation is associated with higher basal AMPK activity and diminished AMP dependence. Our results validate the muscle-specific AMPK gamma3 isoform as a therapeutic target for prevention and treatment of insulin resistance.

    • Today, the drive in many organizations is to focus on reducing production costs while increasing customer satisfaction. One key to succeed with these goals is to develop and improve both quality and maintenance in production as well as quality and maintenance in early phases of the development processes. The purpose of this paper is to discuss how and motivate why research within quality and maintenance development may interact, in order to help companies meet customer demand while at the same time increase productivity. The paper is based on ideas and research perspectives of the newly formed competence group on ‘Quality- and Maintenance Development’ at the School of Innovation, Design and Engineering at the Malardalen University, Sweden. This paper elaborates on the concepts of Quality and Maintenance, its important integration, and provides some examples of ongoing research projects within the competence group.

  • The brominated flame retardants TBECH and DPTE alter prostate growth, histology and gene expression patterns in the mouse

    • The brominated flame retardants (BFRs), 1,2-dibromo-4-(1,2 dibromoethyl)cyclohexane (TBECH) and 2,3-dibromopropyl-2,4,6-tribromophenyl ether (DPTE) bind to the androgen receptor (AR). In vitro bioassays have shown that TBECH is a potent androgen agonist while DPTE is a potent AR antagonist. Both TBECH and DPTE alter gene expression associated with AR regulation. However, it remains to be determined if TBECH and DPTE can affect the prostate. For this reason, we exposed CD1 mice to a 1:1 mixture of TBECH diastereomers α and β, a 1:1 mixture of γ and δ, and to DPTE, and tested their effects on prostate growth, histology and gene expression profiles. Castrated (C) mice were used to study the androgenic effects of TBECHαβ and TBECHγδ while the antagonistic effects of DPTE were studied in non-castrated (NC) mice. We observed that testosterone and TBECHγδ increased body and prostate weights while TBECHαβ affected neither of them; and that DPTE had no effect on body weight but reduced prostate weight drastically. Histomorphometric analysis of the prostate revealed epithelial and glandular alterations in the TBECHγδ group comparable to those in testosterone group while alterations in the TBECHαβ group were less pronounced. DPTE displayed androgen antagonist activity reminiscent of castration. The transcription profile of the prostate was altered by castration and exposure to testosterone and to TBECHγδ reversed several of these changes. Testosterone and TBECHγδ also regulated the expression of several androgen responsive genes implicated in prostate growth and cancer. While DPTE resulted in a drastic reduction in prostate weight, it only affected a small number of genes. The results indicate that TBECHγδ and DPTE are of high human health concern as they may contribute to changes in prostate growth, histology and function.

  • Chronic widespread pain and its associations with quality of life and function at a 20-year follow-up of individuals with chronic knee pain at inclusion

    • Objective: To study the prevalence of chronic widespread pain (CWP) and chronic regional pain (CRP), and their association to quality of life, pain, physical function at a 20-year follow-up in a population based cohort with chronic knee pain at inclusion. Methods: 121 individuals (45% women, mean age 64 years, range 54-73) with chronic knee pain from a population-based cohort study, answered a questionnaire and had radiographic knee examination at a 20-year follow-up. The responders were divided into three groups according to reported pain; individuals having no chronic pain (NCP), chronic widespread pain (CWP) and chronic regional pain (CRP). Pain and physical function were assessed using Knee injury and Osteoarthritis Outcome Score (KOOS). Health related quality of life (HRQL) was assessed with Euroqol-5D-3 L (EQ5D) and Short form 36 (SF36). The associations between pain groups and KOOS, EQ5D, and SF36 were analysed by multiple logistic regression, controlled for age, gender and radiographic changes indicating knee osteoarthritis (OA). Results: The prevalence of CWP was 30%, and CWP was associated to worse scores in all KOOS subscales, controlled for age, gender and radiographic changes. CWP was also associated to worse scores in EQ-5D and in seven of the SF-36 subgroups, controlled for age, gender and radiographic changes. Conclusion: One third of individuals with chronic knee pain met the criteria for CWP. CWP was associated with patient reported pain, function and HRQL. This suggest that it is important to assess CWP in the evaluation of patients with chronic knee pain, with and without radiographic knee OA.

  • Same-same but different : Gendering forest ownership in Sweden

    • Globally, gender has been identified in research as a salient dimension embedded in the social relations of forests. While research related to the Global South is abundant on this topic, the scholarly output from the Global North is sparser. Based on the theoretical understanding of gendering as ongoing contested spatial and constitutive differencing practices, this study, through a qualitative approach, aims to examine and analyse the constitution of private forest ownership in the boreal and production-oriented setting of Sweden. A thematic analysis of semi-structured interviews with 25 female and 26 male forest owners was conducted. Many of the interviewees did not express a gendered experience of their forest ownership, and a diversity in practices of gendering was demonstrated. Also, the analysis highlighted how the gendering of activities, experiences, expectations, and forest values was constructed by emphasising differences through a complementary or dichotomy-related understanding of gender, and by associating specific bodies (women/men) with specific spaces (forest/household), tasks (manual forest labour/domestic labour), characteristics (strong/caring), and perspectives (economic/ecological). This construction contributes to a reproduction of the power of specific production-oriented masculinities and values, e.g. by marking distance or difference to femininities. In the gendering of forest ownership, doing ‘difference’ was highlighted both as a means of ‘othering’ and as a positive and innovative way of resisting and negotiating, as well as a way of reasserting and constituting the current gendered forest ownership and the production-oriented context of forestry in Sweden.
