    • An interference quenching of the m=1 final state vibrational line in the resonant Auger decay of N 1s-->pi(*) core-excited N-2 is observed and analyzed. The intensity ratio between the m=1 and m=0 vibrational levels of the X (2)Sigma(g)(+) final state shows a surprising nonmonotonic variation as a function of frequency detuning, going through a minimum with a complete suppression of m=1. We have developed a simple model which indicates a linear relation between the value of the detuning frequency for this minimum and the equilibrium bond distance of the core-excited state. This implies the possibility of determining the equilibrium bond distances for core-excited states to a high degree of accuracy. Simultaneously with the simple model we present a strict theory of the studied effect. This strict theory allows us to explore the accuracy of determining the bond length of the core-excited state from resonant Auger spectra. We obtain a weak influence of the core-hole lifetime on the determined bond length, whereas the number of intermediate vibrational states accounted for in the numerical simulations seems to be quite important.

  • Automated Nano-Electrospray Mass Spectrometry for Protein-Ligand Screening by Noncovalent Interaction Applied to Human H-FABP and A-FABP

    • A method for ligand screening by automated nano-electrospray ionization mass spectrometry (nano-ESI/MS) is described. The core of the system consisted of a chip-based platform for automated sample delivery from a 96-well plate and subsequent analysis based on noncovalent interactions. Human fatty acid binding protein, H-FABP (heart) and A-FABP (adipose), with small potential ligands was analyzed. The technique has been compared with a previously reported method based on nuclear magnetic resonance (NMR), and excellent correlation with the found hits was obtained. In the current MS screening method, the cycle time per sample was 1.1 min, which is approximately 50 times faster than NMR for single compounds and approximately 5 times faster for compound mixtures. High reproducibility was achieved, and the protein consumption was in the range of 88 to 100 picomoles per sample. Furthermore, a novel protocol for preparation of A-FABP without the natural ligand is presented. The described screening approach is suitable for ligand screening very early in the drug discovery process before conventional high-throughput screens (HTS) are developed and/or used as a secondary screening for ligands identified by HTS.

  • Vestibular evoked myogenic potentials in response to skull taps for patients with vestibular neuritis

    • In recent years it has been demonstrated that loud clicks generate short latency vestibular evoked myogenic potentials (VEMP). It has also been demonstrated that skull tap stimulation evokes similar VEMP. In the present study, the differences between the click-induced and the skull-tap induced VEMP were studied in 18 patients at onset of vestibular neuritis. Gentle skull taps were delivered manually above each ear on the side of the skull and on the forehead midline. The muscular responses were recorded over both sternocleidomastoid muscles using skin electrodes. Abnormal skull tap VEMP were found in the majority of the patients (10/18, 56%). However, only 4/18 (22%) showed asymmetry in the click-induced VEMP. The high percentage of abnormal skull tap VEMP might suggest that this response is not only dependent on the inferior division of the vestibular nerve, because the inferior division of this nerve is usually spared in vestibular neuritis. Moreover, the patients with abnormal skull tap VEMP differed from those with normal VEMP in their settings of the subjective visual horizontal with static head tilt in the roll plane. This might suggest that skull tap VEMP are (also) related to utricular function.

    • A reaction system consisting of terephthalic acid, NaOH, inorganic Mn(II) or Mn(III) salt, and salicylidene alkylimine resulted in dinuclear manganese complexes (salpn)(2)Mn-2(mu-phth)(CH3OH)(2) (1, salpn = N,N'-1,3-propylene-bis(salicylideneiminato); phth = terephthalate dianion), (salen)(2)Mn-2(mu-phth)(CH3OH)(2) (2, salen = N,N'-ethylene-bis(salicylideneiminato)), (salen)(2)Mn-2(mu-phth)(CH3OH)(H2O) (3), and (salen)(2)Mn-2(mu-phth) (4), while the absence of NaOH in the reaction led to a mononuclear Mn complex (salph)Mn(CH3OH)(NO3) (5, salph = N,N'-1,2-phenylene-bis(salicylideneiminato)). In addition, a trinuclear mixed metal complex H{Mn2Na(salpn)(2)(mu-OAc)(2)(H2O)(2)}(OAc)(2) (6) was obtained from the reaction system by using maleic acid instead of terephthalic acid. Five-coordinate Mn ions were found in 4 giving rise to an intermolecular interaction and constructing a one-dimensional linear structure. Antiferromagnetic exchange interactions were observed for 1-3, and a total ferromagnetic exchange of 4 was considered to stem from intermolecular magnetic coupling. H-1 NMR signals of phenolate ring and alkylene (or phenylene) backbone of the diamine are similar to those reported in the literature, and the phth protons are at -2.3 to -10.1 ppm. Studies on structure, bond valence sum analysis, and magnetic properties indicate the oxidation states of the Mn ions in 6 to be +3, which are also indicated by ESR spectra in dual mode. Ferromagnetic exchange interaction between the Mn(III) sites was observed with J = 1.74 cm(-1). A quasireversible redox pair at -0.29V/0.12V has been assigned to the redox of Mn-2(III)/Mn(III)Mn(II), implying the intactness of the complex backbone in solution.

  • Real life testing of a hybrid PEM fuel cell bus

    • Fuel cells produce low quantities of local emissions, if any, and are therefore one of the most promising alternatives to internal combustion engines as the main power source in future vehicles. It is likely that urban buses will be among the first commercial applications for fuel cells in vehicles. This is due to the fact that urban buses are highly visible for the public, they contribute significantly to air pollution in urban areas, they have small limitations in weight and volume and fuelling is handled via a centralised infrastructure. Results and experiences from real life measurements of energy flows in a Scania Hybrid PEM Fuel Cell Concept Bus are presented in this paper. The tests consist of measurements during several standard duty cycles. The efficiency of the fuel cell system and of the complete vehicle are presented and discussed. The net efficiency of the fuel cell system was approximately 40% and the fuel consumption of the concept bus is between 42 and 48% lower compared to a standard Scania bus. Energy recovery by regenerative braking saves up 28% energy. Bus subsystems such as the pneumatic system for door opening, suspension and brakes, the hydraulic power steering, the 24 V grid, the water pump and the cooling fans consume approximately 7% of the energy in the fuel input or 17% of the net power output from the fuel cell system. The bus was built by a number of companies in a project partly financed by the European Commission's Joule programme. The comprehensive testing is partly financed by the Swedish programme "Den Grona Bilen" (The Green Car). A 50 kW(el) fuel cell system is the power source and a high voltage battery pack works as an energy buffer and power booster. The fuel, compressed hydrogen, is stored in two high-pressure stainless steel vessels mounted on the roof of the bus. The bus has a series hybrid electric driveline with wheel hub motors with a maximum power of 100 kW. Hybrid Fuel Cell Buses have a big potential, but there are still many issues to consider prior to full-scale commercialisation of the technology. These are related to durability, lifetime, costs, vehicle and system optimisation and subsystem design. A very important factor is to implement an automotive design policy in the design and construction of all components, both in the propulsion system as well as in the subsystems.

    • The objective of the present study was to assess the performance and recent improvements of the land surface scheme used operationally in the European Centre for Medium-Range Weather Forecasts (ECMWF) in a Scandinavian boreal forest climate/ecosystem. The previous (the 1999 scheme of P. Viterbo and A. K. Betts) and the new (Tiled ECMWF Surface Scheme for Exchange Processes over Land, TESSEL) surface schemes were validated by single-column runs against data from NOPEX (Northern Hemisphere Climate-Processes Land-Surface Experiment). Driving and validation datasets were prepared for a 3-yr period (1994-96). The new surface scheme, with separate surface energy balances for subgrid fractions (tiling), improved predictions of seasonal as well as diurnal variation in surface energy fluxes in comparison with the old scheme. Simulated wintertime evaporation improved significantly as a consequence of the introduced additional aerodynamic resistance for evaporation from snow lying under high vegetation. Simulated springtime evaporation also improved because the limitation of transpiration in frozen soils was now accounted for. However, downward sensible heat flux was still underestimated during winter, especially at nighttime, whereas soil temperatures were underestimated in winter and overestimated in summer. The new scheme also underestimated evaporation during dry periods in summer, whereas soil moisture was overestimated. Sensitivity tests showed that further improvements of simulated surface heat fluxes and soil temperatures could be obtained by calibration of parameters governing the coupling between the surface and the atmosphere and the ground heat flux, and parameters governing the water uptake by the vegetation. Model performance also improved when the seasonal variation in vegetation properties was included.

  • Eccentric correction for off-axis vision in central visual field loss

    • Background. Subjects with absolute central visual field loss use. eccentric fixation and magnifying devices to utilize their residual vision. This preliminary study investigated the importance of an accurate eccentric correction of off-axis refractive errors to optimize the residual visual function for these subjects. Methods. Photorefraction using the PowerRefractor instrument was used to evaluate the ametropia in eccentric fixation angles. Methods were adapted for measuring visual acuity outside the macula using filtered optotypes from high-pass resolution perimetry. Optical corrections were implemented, and the visual function of subjects with central visual field loss was measured with and without eccentric correction. Results. Of the seven cases reported, five experienced an improvement in visual function in their preferred retinal locus with eccentric refraction. Conclusions. The main result was that optical correction for better image quality on the peripheral retina is important for the vision of subjects with central visual field loss, objectively as well as subjectively.
