    • An efficient method to synthesize positional scanning synthetic combinatorial libraries (PS-SCLs) for studying the specificity of protein kinases is presented. Isokinetic ratios for pentafluorophenyl esters were determined iteratively using a new approach incorporating high performance liquid chromatography (HPLC) quantification and statistical experimental design. In the development process a large amount of work was put in to find efficient ways of screening for new isokinetic mixtures and to optimize the process of PS-SCL synthesis. The newly developed methods for the screening of isokinetic mixtures could be used for the screening of other interesting mixtures, but more importantly, the isokinetic ratios determined for the preactivated pentafluorophenyl esters were incorporated into a new efficient protocol. This straightforward protocol allows for a convenient synthesis of high quality PS-SCLs regardless of previous experience in solid phase synthesis.

    • We describe here a library aimed at automating the solution of partial differential equations using the finite element method. By employing novel techniques for automated code generation, the library combines a high level of expressiveness with efficient computation. Finite element variational forms may be expressed in near mathematical notation, from which low-level code is automatically generated, compiled, and seamlessly integrated with efficient implementations of computational meshes and high-performance linear algebra. Easy-to-use object-oriented interfaces to the library are provided in the form of a C++ library and a Python module. This article discusses the mathematical abstractions and methods used in the design of the library and its implementation. A number of examples are presented to demonstrate the use of the library in application code. © 2010 ACM.

    • The rapid development of computer hardware in the last two decades provides a platform for solving large and complex problems numerically. In computational electromagnetics (CEM), extensive work has been contributed to the development of the finite-difference time domain (FDTD) method [1] with a significant interest of modeling electromagnetic phenomena concerning human bodies. In the microwave frequency (few hundred MHz to GHz region), dielectric property of human bodies is highly heterogeneous and varies as a function of frequency [2] . Analytical solutions of electromagnetic wave propagations inside such complex environments are usually not available and thus numerical solutions become important. © 2010 IEEE.

    • This is a collection of different citations and my own naïve pedagogical reasoning, as a mathematics teacher in higher education, concerning the teaching of mathematics at an introductory university level. I will start with some comments about the present calculus-teaching situation, from both a teacher and a student viewpoint. Then I will present an interview with one of my favorite teachers. I will also comment on some books and texts that I found helpful to me, when trying to better understand the current calculus teaching challenges. Finally, I will briefly discuss two challenges didactics research are facing today: How to manifest itself as an accepted science, and how to convince active teachers about its usefulness in daily teaching.

  • Two novel fusion inhibitors of human respiratory syncytial virus

    • To search for novel drugs against human respiratory syncytial virus (RSV), we have screened a diversity collection of 16,671 compounds for anti-RSV activity in cultures of HEp-2 cells. Two of the hit compounds, i.e., the N-(2-hydroxyethyl)-4-methoxy-N-methyl-3-(6-methyl[1,2,4]triazolo[3,4-a]phthalazin-3-yl)benzenesulfonamide (designated as P13) and the 1,4-bis(3-methyl-4-pyridinyl)-1,4-diazepane (designated as C15), reduced the virus infectivity with IC₅₀ values of 0.11 and 0.13μM respectively. The concentration of P13 and C15 that reduced the viability of HEp-2 cells by 50% was 310 and 75μM respectively. Both P13 and C15 exhibited no direct virucidal activity or inhibitory effects on the virus attachment to cells. However, to inhibit formation of RSV-induced syncytial plaques P13 and C15 had to be present during the virus entry into the cells and the cell-to-cell transmission of the virus. The RSV multiplication in HEp-2 cells in the presence of P13 or C15 resulted in rapid selection of viral variants that were ∼1000 times less sensitive to these drugs than original virus. Sequencing of resistant viruses revealed presence of amino acid substitutions in the F protein of RSV, i.e., the D489G for C15-selected, and the T400I and N197T (some clones) for the P13-selected virus variants. In conclusion, we have identified two novel fusion inhibitors of RSV, and the detailed understanding of their mode of antiviral activity including selection for the drug resistant viral variants may help to develop selective and efficient anti-RSV drugs.

    • The duality theorem for Coleff-Herrera products on a complex manifold says that if $f = (f_1,\dots,f_p)$ defines a complete intersection, then the annihilator of the Coleff-Herrera product $\mu^f$ equals (locally) the ideal generated by $f$. This does not hold unrestrictedly on an analytic variety $Z$. We give necessary, and in many cases sufficient conditions for when the duality theorem holds. These conditions are related to how the zero set of $f$ intersects certain singularity subvarieties of the sheaf $\O_Z$.

    • This thesis concerns residue currents on analytic spaces. In the first paper, we construct Coleff-Herrera products and Bochner-Martinelli type currents associated with a weakly holomorphic mapping, and show that these currents satisfy well-known properties from the strongly holomorphic case. This includes the transformation law, the Poincaré-Lelong formula and the equivalence of the Coleff-Herrera product and the Bochner-Martinelli current associated with a complete intersection of weakly holomorphic functions. In the second paper, we discuss the duality theorem on singular varieties. In the case of a complex manifold, the duality theorem, proven by Dickenstein-Sessa and Passare, says that the annihilator of the Coleff-Herrera product associated with a complete intersection $f$ equals the ideal generated by $f$. We give sufficient and in many cases necessary conditions in terms of certain singularity subvarieties of the sheaf $\mathcal{O}_Z$ for when the duality theorem holds on a singular variety $Z$.
