    • Increased automation and use of computer-aided decision support systems are attractive options for hydrologic forecasting agencies faced with growing product complexity and institutional resourcing pressures. Although the hydrologic literature has been nearly silent on the roles of expertise and automation in forecasting, other disciplines such as meteorology have had decades of open discussion on the topic. To address the lack of dialogue in hydrology on automation, this article seeks to contextualize relevant findings from similar disciplines, including meteorology, psychology, decision support systems, and interface design. We predict which aspects of operational hydrology have the greatest chance for successfully increasing automation in the near future. Some applications have employed higher levels of automation, notably flash flood forecasting which requires rapid response times, and extended prediction which requires heavy emphasis on uncertainty quantification. Short-range flood forecasting may be more challenging to automate and traditionally has been less automated than other types of forecasts, partly because of existing practices of interfacing with meteorologists and water system operators, and the difficulties in modeling human impacts on the water cycle. Overall, we suggest that the design of computer-aided decision support systems for forecasting systems should consider three factors: (1) processes change under automation and people may require new roles; (2) automation changes the way people behave, sometimes negatively; and (3) people may not have accurate perceptions of the quality of the automated guidance. Seven lessons learned from automation in meteorology are highlighted and translated into a hydrologic forecasting context, leading to a set of recommendations for how to make best use of expertise in increasingly automated systems.

  • Quinoline-galactose hybrids bind selectively with high affinity to a galectin-8 N-terminal domain

    • Quinolines, indolizines, and coumarins are well known structural elements in many biologically active molecules. In this report, we have developed straightforward methods to incorporate quinoline, indolizine, and coumarin structures into galactoside derivatives under robust reaction conditions for the discovery of glycomimetic inhibitors of the galectin family of proteins that are involved in immunological and tumor-promoting biological processes. Evaluation of the quinoline, indolizine and coumarin-derivatised galactosides as inhibitors of the human galectin-1, 2, 3, 4N (N-terminal domain), 4C (C-terminal domain), 7, 8N, 8C, 9N, and 9C revealed quinoline derivatives that selectively bound galectin-8N, a galectin with key roles in lymphangiogenesis, tumor progression, and autophagy, with up to nearly 60-fold affinity improvements relative to methyl β-d-galactopyranoside. Molecular dynamics simulations proposed an interaction mode in which Arg59 had moved 2.5 Å and in which an inhibitor carboxylate and quinoline nitrogen formed structure-stabilizing water-mediated hydrogen bonds. The compounds were demonstrated to be non-toxic in an MTT assay with several breast cancer cell lines and one normal cell line. The improved affinity, selectivity, and low cytotoxicity suggest that the quinoline-galactoside derivatives provide an attractive starting point for the development of galectin-8N inhibitors potentially interfering with pathological lymphangiogenesis, autophagy, and tumor progression.

  • Characterization of InGaN/GaN quantum well growth using monochromated valence electron energy loss spectroscopy

    • The early stages of InGaN/GaN quantum wells growth for In reduced conditions have been investigated for varying thickness and composition of the wells. The structures were studied by monochromated STEM–VEELS spectrum imaging at high spatial resolution. It is found that beyond a critical well thickness and composition, quantum dots (>20 nm) are formed inside the well. These are buried by compositionally graded InGaN, which is formed as GaN is grown while residual In is incorporated into the growing structure. It is proposed that these dots may act as carrier localization centers inside the quantum wells.

  • Detection and Doppler monitoring of K2-285 (EPIC 246471491), a system of four transiting planets smaller than Neptune

    • Context. The Kepler extended mission, also known as K2, has provided the community with a wealth of planetary candidates that orbit stars typically much brighter than the targets of the original mission. These planet candidates are suitable for further spectroscopic follow-up and precise mass determinations, leading ultimately to the construction of empirical mass-radius diagrams. Particularly interesting is to constrain the properties of planets that are between Earth and Neptune in size, the most abundant type of planet orbiting Sun-like stars with periods of less than a few years. Aims. Among many other K2 candidates, we discovered a multi-planetary system around EPIC 246471491, referred to henceforth as K2-285, which contains four planets, ranging in size from twice the size of Earth to nearly the size of Neptune. We aim here at confirming their planetary nature and characterizing the properties of this system. Methods. We measure the mass of the planets of the K2-285 system by means of precise radial-velocity measurements using the CARMENES spectrograph and the HARPS-N spectrograph. Results. With our data we are able to determine the mass of the two inner planets of the system with a precision better than 15%, and place upper limits on the masses of the two outer planets. Conclusions. We find that K2-285b has a mass of M b = 9.68 -1.37+1.21 M · and a radius of R b = 2.59 -0.06+0.06 R · , yielding a mean density of ρ b = 3.07 -0.45+0.45 g cm -3 , while K2-285c has a mass of M c = 15.68 -2.13+2.28 M · , radius of R c = 3.53 -0.08+0.08 R · , and a mean density of ρ c = 1.95 -0.28+0.32 g cm -3 . For K2-285d (R d = 2.48 -0.06+0.06 R · ) and K2-285e (R e = 1.95 -0.05+0.05 R · ), the upper limits for the masses are 6.5 M · and 10.7 M · , respectively. The system is thus composed of an (almost) Neptune-twin planet (in mass and radius), two sub-Neptunes with very different densities and presumably bulk composition, and a fourth planet in the outermost orbit that resides right in the middle of the super-Earth/sub-Neptune radius gap. Future comparative planetology studies of this system would provide useful insights into planetary formation, and also a good test of atmospheric escape and evolution theories.

  • Renomedullary Blood Flow And Blood Volume Are Increased During Vasopressin Escape

    • Hyponatremia is a common electrolyte disorder usually caused by inappropriate vasopressin (AVP) levels relative to serum osmolality. The degree of the hyponatremia is limited by "escape" from AVP-induced antidiuresis, characterized by increased urine volume and decreased urine osmolality independently of circulating AVP. The mechanisms mediating escape are not fully understood, but we have hypothesized that increased renomedullary blood flow (BF) contributes to this process. We therefore investigated intrarenal BF and blood volume distribution in rats with and without escape.Adult male Sprague-Dawley rats (n=10) were infused with DDAVP (5 ng/h) to produce maximal antidiuresis. Half were fed a liquid diet (AIN-76) to produce escape; half were fed a solid diet to prevent escape. After 5 days, all rats were anesthetized with Inactin and high resolution images (voxel size 97x97x600 µm) of renal BF were acquired using a Siemens Definition Dual Source CT. Iopromide (0.15 ml/rat) was rapidly injected iv and the contrast over the kidney area was collected during 30 s. Data were evaluated by analyzing local renal contrast density utilizing the Siemens Syngo body perfusion tool and correlated to the aorta. Cortical and medullary BF were 709±41 and 251±50 ml/100ml/min respectively in non-escaped rats. Cortical BF in escaped rats was similar (588±81 ml/100ml/min), but medullary BF was increased compared to non-escaped rats (666±105 ml/100ml/min). Blood volumes were similar in the two groups in cortex (42±1 vs. 49±6 ml/100ml), but elevated in the medulla of escaped rats (70±3 vs 18±3 ml/100ml).Our results demonstrate that escape is accompanied by markedly elevated renomedullary BF and volume. Elevated BF to the renal medulla results in reduced interstitial osmolality, and may also contribute to the down-regulation of aquarporin-2 water channels known to accompany escape. These results therefore provide a potential mechanistic explanation for the reduced ability to concentrate urine during AVP escape.

    • Interleukin (IL)-4 and IL-13 are cytokines that inhibit bone resorption. Data showing an inhibitory effect of IL-4 and IL-13 on RANK mRNA in mouse calvariae were first reported at the 22nd American Society for Bone and Mineral Research Meeting (Lerner, U.H., and Conaway, H. H. 2000) J. Bone Min. Res. 15, Suppl. 1, Abstr. SU 230). In the present study, release of 45Ca from cultured mouse calvarial bones stimulated by different cytokines, peptides, and steroid hormones was inhibited by IL-4 and IL-13. IL-4 and IL-13 decreased receptor activator of nuclear factor-kappaB ligand (RANKL) and RANK mRNA and increased osteoprotegerin (OPG) mRNA in calvariae. Additionally, the cytokines decreased RANKL protein and increased OPG protein in calvarial bones. In osteoblasts isolated from calvariae, both an increase in RANKL mRNA and a decrease in OPG mRNA and protein elicited by vitamin D3 were reversed by IL-4 and IL-13. IL-4 and IL-13 decreased the number of tartrate-resistant acid phosphatase positive multinucleated cells and the mRNA expression of calcitonin receptor, tartrate-resistant acid phosphatase, and cathepsin K in mouse spleen cells and bone marrow macrophages (BMM) treated with macrophage colony-stimulating factor and RANKL. Inhibition of mRNA for RANK and the transcription factor NFAT2 was also noted in spleen cell and BMM cultures treated with IL-4 and IL-13. In addition, RANK mRNA and RANK protein were decreased by IL-4 and IL-13 in RAW 264.7 cells. Osteoblasts, spleen cells, and BMM expressed mRNA for the four proteins making up the IL-4 and IL-13 receptors. No effects by IL-4 on bone resorption and osteoclast formation or on RANKL and RANK mRNA expression were seen in Stat6-/- mice. The data indicate that IL-4 and IL-13, via a STAT6-dependent pathway, inhibit osteoclast differentiation and bone resorption by activating receptors on osteoblasts and osteoclasts that affect the RANKL/RANK/OPG system.

    • Background: Three to five percent of patients with acute myocardial infarction (AMI) have normal coronary arteries on invasive coronary angiography (ICA). The aim of this study was to assess the presence and characteristics of atherosclerotic plaques on computed tomography coronary angiography (CTCA) and describe the clinical characteristics of this group of patients. Methods: This was a multicentre, prospective, descriptive study on CTCA evaluation in thirty patients fulfilling criteria for AMI and without visible coronary plaques on ICA. CTCA evaluation was performed head to head in consensus by two experienced observers blinded to baseline patient characteristics and ICA results. Analysis of plaque characteristics and plaque effect on the arterial lumen was performed. Coronary segments were visually scored for the presence of plaque. Seventeen segments were differentiated, according to a modified American Heart Association classification. Echocardiography performed according to routine during the initial hospitalisation was retrieved for analysis of wall motion abnormalities and left ventricular systolic function in most patients. Results: Twenty-five patients presented with non ST-elevation myocardial infarction (NSTEMI) and five with ST-elevation myocardial infarction (STEMI). Mean age was 60.2 years and 23/30 were women. The prevalence of risk factors of coronary artery disease (CAD) was low. In total, 452 coronary segments were analysed. Eighty percent (24/30) had completely normal coronary arteries and twenty percent (6/30) had coronary atherosclerosis on CTCA. In patients with atherosclerotic plaques, the median number of segments with plaque per patient was one. Echocardiography was normal in 4/22 patients based on normal global longitudinal strain (GLS) and normal wall motion score index (WMSI); 4/22 patients had normal GLS with pathological WMSI; 3/22 patients had pathological GLS and normal WMSI; 11/22 patients had pathological GLS and WMSI and among them we could identify 5 patients with a Takotsubo pattern on echo. Conclusions: Despite a diagnosis of AMI, 80 % of patients with normal ICA showed no coronary plaques on CTCA. The remaining 20 % had only minimal non-obstructive atherosclerosis. Patients fulfilling clinical criteria for AMI but with completely normal ICA need further evaluation, suggestively with magnetic resonance imaging (MRI).

    • I en kontext av en generellt ökande välfärdspluralism där medborgaren ställs inför betydligt fler val än tidigare, har det nyliberala tänkesättet också fått ett stort genomslag i det svenska utbildningssystemet. På skolområdet, som traditionellt dominerats av offentliga huvudmän, ser vi idag en blandning mellan offentliga och privata aktörer. Denna utveckling får som konsekvens att såväl de som är verksamma inom utbildningssystemet som elever och deras föräldrar behöver hitta nya förhållningssätt. För de yrkesverksamma innebär ökad konkurrens att de befinner sig i en verksamhet som antar rollen som marknadsaktör där elever och deras föräldrar alltmer blir betraktade som kunder. Utbildningssystemet har skapat lokala skolmarknader. Elever och deras föräldrar åtnjuter ökade valmöjligheter men får samtidigt krav på sig att sätta sig in i och värdera olika alternativ. Ett tydligt exempel på avreglering och marknadsanpassning i utbildningssystemet utgörs av gymnasievalet. I detta kapitel uppmärksammas den period som föregår gymnasiestudier då grundskoleelever med stöd från skolpersonal måste välja den fortsatta studievägen på sekundär nivå. Vi inriktar oss på gymnasieval genom att analysera vad ökade valmöjligheter kan innebära för representanter för grundskolan i förhållande till elevernas gymnasieval. Vi kommer att fokusera på hur rektorer och studie- och yrkesvägledare reflekterar kring sin roll utifrån de förändringar som skett gällande gymnasievalet. Övergripande pekar svaren att inslaget av informationsförmedling har intensifierats samtidigt som många elever behöver ett mer handfast och omfattande stöd i att förhålla sig till valet. Det finns en osäkerhet i hur skolrepresentanter ska förhålla sig till gymnasievalet. Den tidigare rollen som informatör utmanas av nya krav, förväntningar och dilemman vilket leder till att skolrepresentanter kan ta på sig rollen som rådgivare, vägledare eller mäklare för att matcha elevens preferenser och förutsättningar med det komplexa utbud av program, inriktningar och skolor som erbjuds.

  • The missing pieces for better future predictions in subarctic ecosystems: A Torneträsk case study

    • Arctic and subarctic ecosystems are experiencing substantial changes in hydrology, vegetation, permafrost conditions, and carbon cycling, in response to climatic change and other anthropogenic drivers, and these changes are likely to continue over this century. The total magnitude of these changes results from multiple interactions among these drivers. Field measurements can address the overall responses to different changing drivers, but are less capable of quantifying the interactions among them. Currently, a comprehensive assessment of the drivers of ecosystem changes, and the magnitude of their direct and indirect impacts on subarctic ecosystems, is missing. The Torneträsk area, in the Swedish subarctic, has an unrivalled history of environmental observation over 100 years, and is one of the most studied sites in the Arctic. In this study, we summarize and rank the drivers of ecosystem change in the Torneträsk area, and propose research priorities identified, by expert assessment, to improve predictions of ecosystem changes. The research priorities identified include understanding impacts on ecosystems brought on by altered frequency and intensity of winter warming events, evapotranspiration rates, rainfall, duration of snow cover and lake-ice, changed soil moisture, and droughts. This case study can help us understand the ongoing ecosystem changes occurring in the Torneträsk area, and contribute to improve predictions of future ecosystem changes at a larger scale. This understanding will provide the basis for the future mitigation and adaptation plans needed in a changing climate.
