- The Lebanese are the largest group of Middle Eastern immigrants in the United States, and Lebanese immigrants are also prominent across Europe and the Americas. Based on over eighty interviews with first-generation Lebanese immigrants in the global cities of New York, Montreal and Paris, this book shows that the Lebanese diasporaolike all diasporasoconstructs global relations connecting and transforming their new societies, previous homeland and world-wide communities. Taking Lebanese immigrants' forms of identification, community attachments and cultural expression as manifestations of diaspora experiences, the book delves into the ways members of Lebanese diasporic communities move beyond nationality, ethnicity and religion, giving rise to global solidarities and negotiating their social and cultural spaces. The Lebanese Diaspora explores new forms of identities, alliances and cultural expressions, elucidating the daily experiences of Lebanese immigrants and exploring new ways of thinking about immigration, ethnic identity, community, and culture in a global world. By criticizing and challenging our understandings of nationality, ethnicity and assimilation, the book shows that global immigrants are giving rise to new forms of cosmopolitan citizenship.

We hope that this document will be a useful reference for those interested in a more nuanced and complex view of women’s “position” in the Gulf.

Glypican-1: Structural and functional analysis of the N-glycosylated human protein

- Glypicans are multifunctional cell surface heparan sulphate proteoglycans co-regulating numerous signalling pathways, and are thereby involved in the control of cellular division, differentiation, and morphogenesis. The heparan sulphate (HS) chains are responsible for many of those biological functions; nevertheless recent studies suggest functional roles for the glypican core proteins in mediating the signalling of various growth factors. Glypican-1 (GPC1) is the predominant HS proteoglycan in the developing and adult human brain. In addition, GPC1 is involved in Alzheimer’s disease and scrapie, among others. There is a shortage of detailed structural knowledge regarding the GPC1 core protein and accordingly, we proposed in this thesis to structurally and functionally characterize the human GPC1 core protein and to elucidate its overall topology with respect to the membrane. First, we determined the crystal structure of the human N-glycosylated GPC1 core protein by the two-wavelength MAD method on a SeMet-substituted protein crystal. The GPC1 structure revealed a quite rigid, cylindrical single-domain all α-helical fold with three substantial loops. Shortly afterwards, we achieved improvements of GPC1 crystal diffraction properties by controlled crystal dehydration using a humidity control device (HC1b) and generated better electron density for crystals of GPC1, allowing the building of previously disordered parts of the structure. Using small angle X-ray scattering and other biophysical approaches, we found that the GPC1 core protein lies on the membrane in a transverse orientation, directing a surface evolutionarily conserved in GPC1 orthologues towards the membrane, where it can interact with enzymes involved in HS substitution in the Golgi apparatus. Furthermore, the N-linked glycans are shown to extend the protein stability and lifetime by protection against proteolysis and aggregation. The EXTL3 protein, a member of the exostosin family, functions mainly as an initiator for HS assembly on the glypicans. We have investigated the spectroscopic and structural characteristics of the catalytic region of EXTL3, which exhibits a quite stable extended monomeric structure with two functional domains containing a majority of β sheets. Additionally, it was found that catalytic EXTL3 is occupied with N-glycans at least at two sites and these N-glycans seem critical for proper EXTL3 biosynthesis. To precisely determine how the GPC1 core protein regulates HS assembly through interactions with EXTL3, investigations of the GPC1-EXTL3 complexes are ongoing, and some preliminary results are presented here.

- The aim of this licentiate thesis is twofold: 1) to describe old people’s interest in and prospects of using the television for something other than traditional TV viewing, and 2) to describe the nonlinearities in the design process in which this research was performed. A concept called The Extended TV was developed and tested (by old people, their relatives and care workers) in a new nursing home in the process of moving in, hiring employees, making final adjustments on the premises and in the technology. A total of thirty old people, between 60 and 100 years old, were included in the process which was carried out between June 2006 and February 2007. A pivotal background factor was that the initial resistance to the technology itself was low when it came to such a frequently used product as the television set. This proved to also be the case in this study when it came to TVs with flat screens (installed in all rooms and entirely independent of this project). The functions in The Extended TV included both personal ones and ones in common with other residents. The common functions consisted of an internal TV channel and two media centres with the necessary equipment to view common internal productions, other programmes of general interest and material that relatives supplied. The personal function dealt with communication based on one individual’s photo album presented on her own TV. The function and usage of The Extended TV were developed in and through an interweaving of visions, expertise, relationships, responsibilities and logistical prerequisites. The results deal with the interface, the people themselves in action (the old people, their relatives, the staff, IT consultants, technology providers, myself and many others), our tools and the scope of our action. What undoubtedly took the most time was the nonlinear design process. It was complex and by no means followed the initial plans. This is indeed more the rule than the exception in situated human design processes, but is documented all too seldom. I hope that the lessons from the usage of The Extended TV will contribute to innovative ideas concerning old people’s TV viewing.
