    • This book brings together life stories from five generations of Balts, living through the diverse and recurring transformations of the twentieth century: occupations, war, independence, totalitarianism, and democratic rule and market economy. The twentieth century history of the Baltic countries has often been deeply tragic. Lying on the coastline of the Baltic Sea, these rather small but strategically well located territories have historically found themselves in the middle of many power struggles between larger states, empires and other power-holders: the Teutonic Knights, Swedish kings, Tsarist Russia, Nazi Germany, the Soviet Union. Today, they are once again forced to stand up to the Russian Federation.Biographical interviewing is a field focused on individuals, and on how those individuals choose to re-create and present their lived lives, make meaning of it through the narratives they tell. To interpret the biographical narrations of Lithuanians, Latvians and Estonians, shaped by complex and controversial historical background, the authors use Pierre Bourdieu’s concept of social and cultural capitals, the principles of Erving Goffman’s framing analysis and Alessandro Portelli’s distinction of private and public spheres, Anton Steen’s investigations of post-Socialist elites and Piotr Sztompka’s theory of cultural trauma, etc. Given analyses of particular biographical narrations are supplemented by brief historical and sociological overviews, which allow the reader to better understand the contexts of lived lives, and the mental atmosphere in which the interviews were conducted.

  • Weak Boundary and Interface Procedures for Wave and Flow Problems

    • In this thesis, we have analyzed the accuracy and stability aspects of weak boundary and interface conditions (WBCs) for high order finite difference methods on Summations-By-Parts (SBP) form. The numerical technique has been applied to wave propagation and flow problems.The advantage of WBCs over strong boundary conditions is that stability of the numerical scheme can be proven. The boundary procedures in the advection-diffusion equation for a boundary layer problem is analyzed. By performing Navier-Stokes calculations, it is shown that most of the conclusions from the model problem carries over to the fully nonlinear case.The work was complemented to include the new idea of using WBCs on multiple grid points in a region, where the data is known, instead of at a single point. It was shown that we can achieve high accuracy, an increased rate of convergence to steady-state and non-reflecting boundary conditions by using this approach.Using the SBP technique and WBCs, we have worked out how to construct conservative and energy stable hybrid schemes for shocks using two different approaches. In the first method, we combine a high order finite difference scheme with a second order MUSCL scheme. In the second method, a procedure to locally change the order of accuracy of the finite difference schemes is developed. The main purpose is to obtain a higher order accurate scheme in smooth regions and a low order non-oscillatory scheme in the vicinity of shocks.Furthermore, we have analyzed the energy stability of the MUSCL scheme, by reformulating the scheme in the framework of SBP and artificial dissipation operators. It was found that many of the standard slope limiters in the MUSCL scheme do not lead to a negative semi-definite dissipation matrix, as required to get pointwise stability.Finally, high order simulations of shock diffracting over a convex wall with two facets were performed. The numerical study is done for a range of Reynolds numbers. By monitoring the velocities at the solid wall, it was shown that the computations were resolved in the boundary layer. Schlieren images from the computational results were obtained which displayed new interesting flow features.

  • Engineering of the Ultra-stable Cystine Knot Framework of Microproteins : Design, Chemical Synthesis and Structural Studies

    • Ultra-stable cystine knotted microproteins, in which two disulfides and their connecting backbones form a circle that is penetrated by the third disulfide bonds, have attracted high interest due to their resistance to degradation in vitro and potential for the development of peptide drugs. This thesis gives new insights into engineering of that framework of microproteins, including approaches to their chemical synthesis, backbone engineering, structural and biological evaluations. Synthetic and oxidative folding approaches for bracelet cyclotides, a family of cyclic cystine knotted microproteins, was developed using a model peptide, cycloviolacin O2. Following assembly of the peptide chain, protected peptide was generated by mild cleavage that was subsequently thioesterified and cyclized in solution. The cyclic peptide was oxidatively folded under optimized conditions containing co-solvent and non-ionic detergent affording native cycloviolacin O2 as a major product. To gain further insights into the heterogeneity, efficiency and kinetics of cyclotides’ oxidative folding, the intermediates that accumulate in oxidative refolding pathways of all cyclotide subfamilies: Möbius, bracelet and the hybrid cyclotides were quantitatively determined under four different folding conditions. The results were used for defining major folding pathways, which indicated that Möbius cyclotides might accumulate heterogeneous folding intermediates with one-, two- and three-disulfides, whereas bracelet tend to accumulate a homogenous intermediate with three-disulfides, depending on the buffer systems used. Furthermore, to probe the internal factors contributing to inefficiency of oxidative folding, as well as undesired bioactivities of bracelet cyclotides (e.g., cytotoxic activity), polymer-hybridized cyclotides were designed by replacing non-conserved residues with small isosteric polymers. The designed hybrid analogs in which hybridization involved replacement of loop 3 with isosteric polymers showed improved synthetic and oxidative folding properties. The cytoxicity of a model hybrid designed with replacement of loop 3 and 5 exhibited no cytotoxic activity at concentration of 128-fold relative to that of native peptide. Furthermore, 1D and 2D 1H NMR analysis of this hybrid showed that it had well structured fold.

    • One of the main challenges in order to make electric cars competitive with gaspowered cars is in the improvement of the electric power system. Although many of the energy sources currently used in electric vehicles have sufficiently high specific energy, their applicability is limited due to low specific power. It would therefore be advantageous to create a driveline with the main energy storage separated from a smaller energy buffer, designed to have high power capabilities and to withstand frequent and deep discharge cycles. It has been found that rotating kinetic energy storage in flywheels is very well suited for this type of application. The work presented in this thesis and the included papers span a number of topcis Introductory overview - This section explains the concept of the modern flywheel, and investigates some of its properties. It illustrates the concepts with a number of examples, relevant for the usage of flywheels in vehicular applications. Experimental set-up - The construction of a complete electric driveline is ongoing within the division for Electricity at Uppsala University. An optimized electric machine has been constructed and connected with a programmable load, as well as with a DC power source through power electronics controlled by PWM. As a part of this system, an experimental set-up of an active magnetic bearing for two degrees-of-freedom has been constructed. The work with this device is described in detail and some preliminary results are presented. Self-bearing machine - The electric machine developed for the existing driveline is coreless, double wound and with a double rotor. In order to achieve magnetic bearing functionality in this device for all DOF, a novel Lorentz force self-bearing machine is suggested. The design is analyzed analytically and numerically.
