  • Direct repeat-mediated deletion of a type IV pilin gene results in major virulence attenuation of Francisella tularensis
  • 2006
  • Ingår i: Molecular Microbiology. - : Wiley-Blackwell. - 0950-382X .- 1365-2958. ; 59:6, s. 1818-1830
  • Tidskriftsartikel (refereegranskat)abstract
    • Francisella tularensis, the causative agent of tularaemia, is a highly infectious and virulent intracellular pathogen. There are two main human pathogenic subspecies, Francisella tularensis ssp. tularensis (type A), and Francisella tularensis ssp. holarctica (type B). So far, knowledge regarding key virulence determinants is limited but it is clear that intracellular survival and multiplication is one major virulence strategy of Francisella. In addition, genome sequencing has revealed the presence of genes encoding type IV pili (Tfp). One genomic region encoding three proteins with signatures typical for type IV pilins contained two 120 bp direct repeats. Here we establish that repeat-mediated loss of one of the putative pilin genes in a type B strain results in severe virulence attenuation in mice infected by subcutaneous route. Complementation of the mutant by introduction of the pilin gene in cis resulted in complete restoration of virulence. The level of attenuation was similar to that of the live vaccine strain and this strain was also found to lack the pilin gene as result of a similar deletion event mediated by the direct repeats. Presence of the pilin had no major effect on the ability to interact, survive and multiply inside macrophage-like cell lines. Importantly, the pilin-negative strain was impaired in its ability to spread from the initial site of infection to the spleen. Our findings indicate that this putative pilin is critical for Francisella infections that occur via peripheral routes.

  • Public Advisory Services - Theory and Practice
  • 2003
  • Ingår i: Entrepreneurship and Regional Development. - : Informa UK Limited. - 0898-5626 .- 1464-5114. ; 15:1, s. 83-98
  • Tidskriftsartikel (refereegranskat)abstract
    • Public advisory service to SMEs is a multibillion pound activity throughout the industrialized world. Yet very little research has been done on the theoretical basis for this field. This paper proposes some elements in a theoretical understanding of the rationale behind public measures. The authors argue that public intervention should be considered at two levels, as a public market intervention and as a consultant-client relation at the micro level. At the market intervention level, public advisory service is seen in the perspective of economic theory, comparing neo-classical and neo-Austrian theory. Two different kinds of services are identified and discussed: operational and strategic. At a micro level, the concepts of client identity and clientifying power relations serve to understand the small business manager's way of responding to services. In combining both levels - the market perspective and the micro level - it is argued that the neo-classical theory is connected to operational/expert services and objectifying power technologies. The neo-Austrian theory corresponds with the empirical findings at the micro level showing strategic services embedded in a subjectifying power technology. With the neo-Austrian perspective the rather symmetrical relations between client and consultant at the micro level is comprehensible

  • Consulting as story-making
  • 2004
  • Ingår i: Journal of Management Development. - : Emerald. - 0262-1711 .- 1758-7492. ; 23:4, s. 339-354
  • Tidskriftsartikel (refereegranskat)abstract
    • Story‐telling has been identified as a metaphor for management consulting which focuses on the consultant as “impression manager”. The convincing power of strong stories told by consultants is emphasised. In this article management consulting is conceptualised through the metaphor of story‐making, identifying co‐operation and reflection as characteristic features. A specific consultancy process is interpreted as a co‐operative effort between the consultant and the client in which plot lines and characters are constructed. Plots can be opened or closed, and characters can be settled or negotiated. Thus story‐making as a reflective act moves between enhancing reflection and preparing for action.
