    • This study was designed to quantitatively assess the mRNA expression of 36 important drug transporters in human jejunum, colon, liver, and kidney. Expression of these transporters in human organs was compared with expression in commonly used cell lines (Caco-2, HepG2, and Caki-1) originating from these organs to assess their value as in vitro transporter system models, and was also compared with data obtained from the literature on expression in rat tissues to assess species differences. Transporters that were highly expressed in the intestine included HPT1, PEPT1, BCRP, MRP2, and MDR1, whereas, in the liver, OCT1, MRP2, OATP-C, NTCP and BSEP were the main transporters. In the kidney, OAT1 was expressed at the highest levels, followed by OAT3, OAT4, MCT5, MDR1, MRP2, OCT2, and OCTN2. The best agreement between human tissue and the representative cell line was observed for human jejunum and Caco-2 cells. Expression in liver and kidney ortholog cell lines was not correlated with that in the associated tissue. Comparisons with rat transporter gene expression revealed significant species differences. Our results allowed a comprehensive quantitative comparison of drug transporter expression in human intestine, liver, and kidney. We suggest that it would be beneficial for predictive pharmacokinetic research to focus on the most highly expressed transporters. We hope that our comparison of rat and human tissue will help to explain the observed species differences in in vivo models, increase understanding of the impact of active transport processes on pharmacokinetics and distribution, and improve the quality of predictions from animal studies to humans.

  • FORTE: an extensible framework for robustness and efficiency in data transfer pipelines

    • In the age of big data and growing product complexity, it is common to monitor many aspects of a product or system, in order to extract well-founded intelligence and draw conclusions, to continue driving innovation. Automating and scaling processes in data-pipelines becomes essential to keep pace with increasing rates of data generated by such practices, while meeting security, governance, scalability and resource-efficiency demands.We present FORTE, an extensible framework for robustness and transfer-efficiency in data pipelines. We identify sources of potential bottlenecks and explore the design space of approaches to deal with the challenges they pose. We study and evaluate synergetic effects of data compression and in-memory processing as well as task scheduling, in association with pipeline performance.A prototype implementation of FORTE is implemented and studied in a use-case at Volvo Trucks for high-volume production-level data sets, in the order of magnitude of hundreds of gigabytes to terabytes per burst. Various general-purpose lossless data compression algorithms are evaluated, in order to balance compression effectiveness and time in the pipeline.All in all, FORTE enables to deal with trade-offs and achieve benefits in latency and sustainable rate (up to 1.8 times better), effectiveness in resource utilisation, all while also enabling additional features such as integrity verification, logging, monitoring and traceability, as well as cataloguing of transferred data. We also note that the resource efficiency improvements achievable with FORTE, and its extensibility, can imply further benefits regarding scheduling, orchestration and energy-efficiency in such pipelines.

  • Neutron guide optimisation for a time-of-flight neutron imaging instrument at the European Spallation Source

    • A neutron transport system for the planned imaging instrument ODIN at the future European Spallation Source (ESS) based on neutron optical components was designed and optimized. Different ways of prompt pulse suppression were studied. The spectral performance of the optimal neutron guide configuration is presented. In addition, the influence of the gaps in the guide system needed for the required chopper configuration was investigated. Given that the requirements for an imaging instrument located on a long guide system and hosting a complex chopper system are extremely demanding in terms of spectral and divergence needs, this study can be beneficial for a wide range of instruments in various ways.     • QCD sum rules for chiral partners in the open-charm meson sector are presented at nonzero baryon net density or temperature. We focus on the differences between pseudo-scalar and scalar as well as vector and axial-vector D mesons and derive the corresponding Weinberg-type sum rules. This allows for the identification of such QCD condensates which drive the nondegeneracy of chiral partners in the lowest order of the strong coupling alpha(s) and which, therefore, may serve as "order parameters" for chiral restoration (or elements thereof).

Referring to this, details concerning the cancelation of infrared divergences are presented and important technical and conceptional ingredients for an incorporation of four-quark condensates beyond factorization and of other mass dimension 6 condensates are collected.

  • The impact of chirally odd condensates on the rho meson

    • Based on QCD sum rules we explore the consequences of a scenario for the rho meson, where the chiral symmetry breaking condensates are set to zero whereas the chirally symmetric condensates remain at their vacuum values. This clean-cut scenario causes a lowering of the rho spectral moment by about 120 MeV. The complementarity of mass shift and broadening is discussed. A simple parametrization of the rho spectral function leads to a width of about 280 MeV if no shift of the peak position is assumed.
