Direct visualization of Propionibacterium acnes in prostate tissue by multicolor fluorescent in situ hybridization assay.

- Prostate tissues from patients with prostate cancer and benign prostatic hyperplasia (BPH) frequently contain histological inflammation, and a proportion of these patients show evidence of Propionibacterium acnes infection in the prostate gland. We developed a multicolor fluorescent in situ hybridization (FISH) assay targeting P. acnes 23S rRNA along with a 14-kb region of the P. acnes genome. This assay was used to analyze prostate tissues from patients with prostate cancer and BPH. P. acnes infection of the prostate gland was demonstrated in prostatic tissue in 5 of 10 randomly selected prostate cancer patients. FISH analysis and confocal laser microscopy imaging revealed intracellular localization and stromal biofilm-like aggregates as common forms of P. acnes infection in prostate tissues from both prostate cancer and BPH patients. A sequential analysis of prostate tissue from individual patients suggested that P. acnes can persist for up to 6 years in the prostate gland. These results indicate that P. acnes can establish a persistent infection in the prostate gland. Further study is needed to clarify the link between this bacterium and prostatic inflammation which may contribute to the development of BPH and prostate cancer.

Performing Co-production : On the logic and practice of shopping at IKEA

- Contemporary western society has often been described as a “consumer society” in relation to the producer oriented form that characterised the industrial society. While consumer habits used to be seen as a reflection of a person’s occupational status or place in a stable societal hierarchy, it has now become recognised as a practice through which people’s identity and status is partially defined by the choices they make as consumers. This has not only changed the view of the consumer but also created a new role for economic actors. Instead of merely responding to demand they have been recognised as active participants in ongoing processes of socio-cultural transformation. In this thesis I develop a particular understanding of the relation between the activities taking place within consumer markets and specific processes of home making. With theoretical reference to a performative perspective on economic markets and using my own empirical study of the multinational retailer IKEA’s operation in the North East of England, the thesis describes shopping at IKEA as a situated encounter where economic exchange becomes framed as meaningful in particular ways. At IKEA, this process is specifically organised around a principle of co-production. By doing part of the work in terms of picking up and assembling flat-packed furniture, the consumers are offered a low price. Rather than being limited to this work, co-productivity is here identified as a “calculative” logic and practice that characterises and frames the shopping experience as a whole and creates a relation between the consumer and the retailer through the medium of the retail brand. The co-productive calculation established through the shopping experience then continues to make sense as part of ongoing processes of home making. It does so in ways that enhances the immediate, temporary and mobile aspects of the home. Hence, it is by linking the experience of shopping at IKEA and processes of home making that the relation between economic exchange and the ongoing processes of socio-cultural transformation is identified. Rather than answering a particular question, the research and analysis constitutes a specific approach to how economic exchange may be understood and offers some suggestions as to what this may entail in terms of how relations within economic markets may be organised.

Endothelial dysfunction in growth hormone transgenic mice

- Acromegaly [overproduction of GH (growth hormone)] is associated with cardiovascular disease. Transgenic mice overexpressing bGH (bovine GH) develop hypertension and hypercholesterolaemia and could be a model for cardiovascular disease in acromegaly. The aims of the present study were to investigate the effects of excess GH on vascular function and to test whether oxidative stress affects endothelial function in bGH transgenic mice. We studied the ACh (acetylcholine)-induced relaxation response in aortic and carotid rings of young (9-11 weeks) and aged (22-24 weeks) female bGH transgenic mice and littermate control mice, without and with the addition of a free radical scavenger {MnTBAP [Mn(III)tetrakis(4-benzoic acid)porphyrin chloride]}. We also measured mRNA levels of eNOS (endothelial nitric oxide synthase) and EC-SOD (extracellular superoxide dismutase). Intracellular superoxide anion production in the vascular wall was estimated using a dihydroethidium probe. Carotid arteries from bGH transgenic mice had an impaired ACh-induced relaxation response (young, 46 +/- 7% compared with 69 +/- 8%; aged, 52 +/- 5% compared with 80 +/- 3%; P < 0.05), whereas endothelial function in aorta was intact in young but impaired in aged bGH transgenic mice. Endothelial dysfunction was corrected by addition of MnTBAP in carotid arteries from young mice and in aortas from aged mice; however, MnTBAP did not correct endothelial dysfunction in carotid arteries from aged bGH transgenic mice. There was no difference in intracellular superoxide anion production between bGH transgenic mice and control mice, whereas mRNA expression of EC-SOD and eNOS was increased in aortas from young bGH transgenic mice compared with control mice (P < 0.05). We interpret these data to suggest that bGH overexpression is associated with a time- and vessel-specific deterioration in endothelial function, initially caused by increased oxidative stress and later by other alterations in vascular function.

- Human growth hormone (GH) excess is linked to increased cardiovascular morbidity and mortality. However, little is known about the effect of GH excess on atherosclerosis. We developed a new mouse model to assess the hypothesis that GH overexpression accelerates atherosclerotic lesion formation. apoE(-/-) mice were crossed with bovine GH (bGH) transgenic mice to yield apoE(-/-) mice overexpressing bGH (apoE(-/-)/bGH). The mice were fed either standard or Western diet. At 22 weeks, atherosclerotic lesion area of thoracic aorta was larger in apoE(-/-)/bGH mice compared with littermate apoE(-/-) mice fed either diet (standard: +161+/-50%, Western: +430+/-134%). Aortic sinus lesions were more severe in apoE(-/-)/bGH mice fed standard diet compared with littermate apoE(-/-) mice. apoE(-/-)/bGH mice had lower (VLDL+LDL)/HDL ratios compared with littermate apoE(-/-) mice, while systolic blood pressure was higher in apoE(-/-)/bGH mice, irrespective of diet. The levels of serum amyloid A and hepatic CRP mRNA were higher in apoE(-/-)/bGH mice than in littermate apoE(-/-) mice. In conclusion, this study shows that excess GH augments the development of atherosclerosis in apoE(-/-) mice. The mechanisms could be direct effects of GH on cellular processes in the vessel wall or the result of concomitant processes such as hypertension or a general inflammatory state.

- Abstract This paper is a critical reflection on the basic ideas underpinning the new teacher education programmes at Malmö University. The main idea has been to combine subject studies, didactics and pedagogy in teacher education. We claim that personal encounters are one of the prerequisites for student teachers to learn and in depth understand in order to be able to translate into teaching. In the paper we will present and discuss some examples of personal encounters in relation to Citizenship Education within the main-subject “Social Science Subject and Children’s Learning”. This will illustrates a possible way of the construction of meaning in teacher education.
