    • OBJECTIVE: To investigate the impact of time since injury on neuropsychological and psychosocial outcome after serious TBI in childhood or adolescence. METHODS: The subjects were eight patients with serious TBI sustained at a mean age of 14 years who had been assessed neuropsychologically at 1, 7 and 14 years after TBI. A retrospective longitudinal design was chosen to describe the development in six neuropsychological domains on the basis of the assessments. Psychosocial data were gathered from clinical knowledge and a semi-structured interview 14 years after TBI. RESULTS: Performance of verbal IQ shows a declining trend over the three assessments, that the performance of attention and working memory is low and that verbal learning is the cognitive domain which exhibits the largest impairments. The main psychosocial result is that three of the eight subjects went from a school situation with no adjustments to adult life with early retirement. CONCLUSIONS: Time since insult is an important factor when assessing outcome after TBI in childhood and adolescence and that assessment of final outcome should not be done before adulthood.

    • We report on the first experimental study of the lifetime of a bound excited state of a negative ion. A new experimental technique was developed and used to measure the radiative lifetime of the 5p(5) P-2(1/2) level of Te-. The experiment was performed in a magnetic storage ring, where a laser beam was applied along one of the straight sections. In the experiment the population of the excited J=1/2 level was probed each time the Te- ions passed through the laser field. A decay curve was built up by sampling the population of the excited level of the Te- ions as a function of time after injection into the ring. A multiconfiguration Dirac-Hartree-Fock calculation was performed in conjunction with the experiment. The calculation yielded a radiative lifetime of 0.45 s, in excellent agreement with the measured value of 0.42(5) s.

  • Seagrass ecosystems in the Western Indian Ocean

    • Seagrasses are marine angiosperms widely distributed in both tropical and temperate coastal waters creating one of the most productive aquatic ecosystems on earth. In the Western Indian Ocean (WIO) region, with its 13 reported seagrass species, these ecosystems cover wide areas of near-shore soft bottoms through the 12 000 km coastline. Seagrass beds are found intertidally as well as subtidally, sometimes down to about 40 m, and do often occur in close connection to coral reefs and mangroves. Due to the high primary production and a complex habitat structure, seagrass beds support a variety of benthic, demersal and pelagic organisms. Many fish and shellfish species, including those of commercial interest, are attracted to seagrass habitats for foraging and shelter, especially during their juvenile life stages. Examples of abundant and widespread fish species associated to seagrass beds in the WIO belong to the families Apogonidae, Blenniidae, Centriscidae, Gerreidae, Gobiidae, Labridae, Lethrinidae Lutjanidae, Monacanthidae, Scaridae, Scorpaenidae, Siganidae, Syngnathidae and Teraponidae. Consequently, seagrass ecosystems in the WIO are valuable resources for fisheries at both local and regional scales. Still, seagrass research in the WIO is scarce compared to other regions and it is mainly focusing on botanic diversity and ecology. This article reviews the research status of seagrass beds in the WIO with particular emphasis on fish and fisheries. Most research on this topic has been conducted along the East African coast, i.e. in Kenya, Tanzania, Mozambique and eastern South Africa, while less research was carried out in Somalia and the Island States of the WIO (Seychelles, Comoros, Reunion (France), Mauritius and Madagascar). Published papers on seagrass fish ecology in the region are few and mainly descriptive. Hence, there is a need of more scientific knowledge in the form of describing patterns and processes through both field and experimental work. Quantitative seagrass fish community studies in the WIO such as the case study presented in this paper are negligible, but necessitated for the perspective of fisheries management. It is also highlighted that the pressure on seagrass beds in the region is increasing due to growing coastal populations and human disturbance from e.g. pollution, eutrophication, sedimentation, fishing activities and collection of invertebrates, and its effect are little understood. Thus, there is a demand for more research that will generate information useful for sustainable management of seagrass ecosystems in the WIO.

  • Structural modeling of dual-affinity purified Pho84 phosphate transporter

    • The phosphate transporter Pho84 of Saccharomyces cerevisiae is predicted to contain 12 transmembrane (TM) regions, divided into two partially duplicated parts of 6 TM segments. The three-dimensional (3D) organization of the Pho84 protein has not yet been determined. However, the 3D crystal structure of the Escherichia coli MFS glycerol-3-phosphate/phosphate antiporter, GlpT, and lactose transporter, LacY, has recently been determined. On the basis of extensive prediction and fold recognition analyses (at the MetaServer), GlpT was proposed as the best structural template on which the arrangement of TM segments of the Pho84 transporter was fit, using the comparative structural modeling program MODELLER. To initiate an evaluation of the appropriateness of the Pho84 model, we have performed two direct tests by targeting spin labels to putative TM segments 8 and 12. Electron paramagnetic resonance spectroscopy was then applied on purified and spin labeled Pho84. The line shape from labels located at both positions is consistent with the structural environment predicted by the template-generated model, thus supporting the model.

    • An efficient and short route has been elaborated for the aminopropyl spacer equipped Leb hexasaccharide. For the preparation of HSA-conjugates of this oligosaccharide, the use of disuccinimidyl suberate (DSS) and disuccinimidyl glutarate (DSG) as cross-linker reagents has been evaluated. This conjugation method emerged as being faster and easier to monitor by standard MALDI-TOF spectrometry than squarate ester based conjugations of similar efficiency if DSS is used as cross-linker.

    • Several brominated flame retardants (BFRs) were analyzed in peregrine falcon eggs collected in 1987-1999, including the constituents of the technical polybrominated diphenyl ether (PBDE) products Penta (BDE-47, -99, -100, -153, -154), Octa (BDE-183), and Deca (BDE-209), hexabrominated biphenyl (BB-153), and hexabromocyclododecane (HBCD). The eggs represented females from three different breeding populations, northern Sweden, southwestern Sweden, and a captive breeding population. All BFRs analyzed for were found, including BDE-183 and -209, and concentrations were much higher in wild falcons (geometric mean SigmaPBDE, BB-153, and HBCD for northern/southern populations of 2200/2700, 82/77, and 150/250 ng/g lw, respectively) than in captive falcons (39, 8 ng/g lw, and not detected, respectively). This is the first time, to our knowledge, that BDE-183 and -209 have been quantified in high trophic level wildlife.

    • In Saccharomyces cerevisiae, the Pho84 high-affinity transport system is the major phosphate transporter activated when the cells experience a limitation in external phosphate. In this study, we have compared the phosphate-responsive mechanism of cells expressing PHO84 with a Deltapho84 strain by use of a phosphate analogue, methylphosphonate, which was judged to be suitable for assessment of phosphate homeostasis in the cells. Intracellular levels of the analogue, which in several respects mimicks phosphate, were monitored by (31)P NMR spectroscopy. Results show that methylphosphonate is a nonhydrolyzable and nonutilizable analogue that cannot be used to replenish phosphate or polyphosphate in yeast cells grown under conditions of phosphate limitation. However, the presence of methylphosphonate under such conditions represses the Pho5 acidic phosphatase activity of PHO84 cells, a finding that implies a direct role of the analogue in the regulation of phosphate-responsive genes and/or proteins. Likewise, accumulation of the Pho84 protein at the plasma membrane of the same cells is inhibited by methylphosphonate, although the derepressive expression of the PHO84 gene is unperturbed. Thus, a post-transcriptional regulation is suggested. Supportive of this suggestion is the fact that addition of methylphosphonate to cells with abundant and active Pho84 at the plasma membrane causes enhanced internalization of the Pho84 protein. Altogether, these observations suggest that the Pho84 transporter is regulated not only at the transcriptional level but also by a direct molecule-sensing mechanism at the protein level.

  • Chemical settlement inhibition versus post-settlement mortality as an explanation for differential fouling of two congeneric seaweeds

    • It has been proposed that seaweed secondary metabolites, e.g. brown algal phlorotannins, may have an ecologically important function as a chemical defence against epiphytes, by acting against colonisation of epiphytic organisms. We tested whether the low epiphytic abundance on the invasive brown seaweed Fucus evanescens, compared to the congeneric F. vesiculosus, is due to a more effective chemical defence against epiphyte colonisation. A field survey of the distribution of the common fouling organism Balanus improvisus (Cirripedia) showed that the abundance was consistently lower on F. evanescens than on F. vesiculosus. However, contrary to expectations, results from experimental studies indicated that F. vesiculosus has a more effective anti-settlement defence than F. evanescens. In settlement experiments with intact fronds of the two Fucus species, both species deterred settlement by barnacle larvae, but settlement was lower on F. vesiculosus both in choice and no-choice experiments. Phlorotannins from F. vesiculosus also had a stronger negative effect on larval settlement and were active at a lower concentration than those from F. evanescens. The results show that Fucus phlorotannins have the potential to inhibit settlement of invertebrate larvae, but that settlement inhibition cannot explain the lower abundance of the barnacle Balanus improvisus on F. evanescens compared to F. vesiculosus. Assessment of barnacle survival in the laboratory and in the field showed that this pattern could instead be attributed to a higher mortality of newly settled barnacles. Observation suggests that the increased mortality was due to detachment of young barnacles from the seaweed surface. This shows that the antifouling mechanism of F. evanescens acts on post-settlement stages of B. improvisus.
