  • A deep proteome and transcriptome abundance atlas of 29 healthy human tissues

    • Genome-, transcriptome- and proteome-wide measurements provide insights into how biological systems are regulated. However, fundamental aspects relating to which human proteins exist, where they are expressed and in which quantities are not fully understood. Therefore, we generated a quantitative proteome and transcriptome abundance atlas of 29 paired healthy human tissues from the Human Protein Atlas project representing human genes by 18,072 transcripts and 13,640 proteins including 37 without prior protein-level evidence. The analysis revealed that hundreds of proteins, particularly in testis, could not be detected even for highly expressed mRNAs, that few proteins show tissue-specific expression, that strong differences between mRNA and protein quantities within and across tissues exist and that protein expression is often more stable across tissues than that of transcripts. Only 238 of 9,848 amino acid variants found by exome sequencing could be confidently detected at the protein level showing that proteogenomics remains challenging, needs better computational methods and requires rigorous validation. Many uses of this resource can be envisaged including the study of gene/protein expression regulation and biomarker specificity evaluation.

    • A Myxococcus xanthus gene, MXAN3487, was identified by transposon mutagenesis to be required for the expression of mcuABC, an operon coding for part of the chaperone-usher (CU) system in this bacterium. The MXAN3487 protein displays sequence and structural homology to adenosine 5'-phosphosulphate (APS) kinase family members and contains putative motifs for ATP and APS binding. Although the MXAN3487 locus is not linked to other sulphate assimilation genes, its protein product may have APS kinase activity in vivo and the importance of the ATP-binding site for activity was demonstrated. Expression of MXAN3487 was not affected by sulphate availability, suggesting that MXAN3487 may not function in a reductive sulphate assimilation pathway. Deletion of MXAN3487 significantly delayed fruiting body formation and the production of McuA, a spore coat protein secreted by the M. xanthus Mcu CU system. Based on these observations and data from our previous studies, we propose that MXAN3487 may phosphorylate molecules structurally related to APS, generating metabolites necessary for M. xanthus development, and that MXAN3487 exerts a positive effect on the mcuABC operon whose expression is morphogenesis dependent.

  • Adaptive Chromatic Dispersion Compensation for Coherent Communication Systems Using Delay-Tap Sampling Technique

    • A fast and low-complexity adaptive chromatic dispersion (CD) compensation algorithm based on delay-tap sampling technique for optical coherent systems is proposed. By generating a delay-tap plot from the signal powers after an adaptive CD equalizer, a parameter can be extracted from the delay-tap plot which can be used to estimate CD accurately. Simulation results for coherent polarization-division-multiplexed (PDM) quaternary phase-shift keying (QPSK) as well as 16-quadrature amplitude modulation (QAM) systems with various pulse shapes demonstrate that high CD estimation accuracies using relatively few data symbols can be achieved in presence of other transmission impairments.

  • Effect of basalt fiber inclusion on the mechanical properties and microstructure of cement-solidified kaolinite

    • The polypropylene fibers, which are currently attracting enormous attention in various geotechnical applications, carry a risk of aging under an integrated effect of heat, oxygen, light and other environmental factors, causing potentially infrastructure failure. An eco-friendly and biologically inactive material – basalt fiber, which has excellent natural resistance to aging and can eliminate aging-associated disasters, deserves more attention in geotechnical field. However, quite few studies are available on the beneficial reuse of basalt fibers to improve the engineering performance of soils. Therefore, this study aims to incorporate the sustainable basalt fiber and clarify how its inclusion impacts the mechanical properties and microstructure of cemented kaolinite. The experimental programs are comprised of three types of tests, i.e. two to examine the compressive strength and triaxial shear behavior and one to evaluate the microstructure properties. The results indicate that the basalt fiber reinforcement plays an essential role in enhancing the compressive strength and peak deviatoric stress of cemented and uncemented kaolinite. The inclusion of basalt fibers improves the ductility and weakens the brittleness of cemented kaolinite. The compressive strength increases with basalt fiber content and curing time, and reaches the peak at the fiber content of 0.2%, followed by a reduction due to the formation of weak zone at higher fiber content. The peak deviatoric stress is elevated until reaching the maximum at the basalt fiber content of 0.4%, after which further addition of basalt fiber tends to reduce its reinforcing effect. The peak deviatoric stress increases as the basalt fiber length is shortened and the confining pressure is raised. The strength gain of cement-basalt fiber inclusion is much more than the sum of strength increase induced by them individually. The combination of basalt fiber and cement has the virtues of both cement-stabilized and basalt fiber-reinforced kaolinite. The SEM analysis reveals that the mechanical interaction in the form of interface bonding and friction between kaolinite particle, cement hydration product and basalt fiber is the dominant mechanism controlling the reinforcement-cementation benefits. The bridging effect (reinforcement) of basalt fibers and binding effect (cementation) of hydration products make a major contribution to the formation of stable and interconnected microstructure, which results in an evident improvement in the mechanical behaviour of cemented kaolinite. The combination of basalt fiber and cement stabilization would be an innovative and effective method for geotechnical engineering works such as soft ground improvement.

    • Alkali-activated industrial by-products (granulated blast furnace slag, Class F fly ash) by traditional alkali activator (such as NaOH and Na2SiO3) serves as a partial replacement for Portland cement in soil stabilization projects and suffers from environmental and technical problems. Reactive MgO - a greener and more practical alternative has recently emerged as a potential activator for slag and fly ash, but its micromechanisms of alkaline activation still need to be deeply investigated for strength improvement of soils. Hence, this study focuses on the strength and hydration properties of reactive MgO-slag and MgO-fly ash solidified soils, especially incorporating the impact of elevated curing temperature. Reactive MgO is proved to be excellent as a novel activator for activation of slag and fly ash, and their activating efficiency increases with elevated curing temperature that helps to remarkably enhance the compressive strength of soils. The major hydration products for reactive MgO-slag solidified soils, detected jointly by X-ray diffraction, scanning electron microscopy and thermogravimetric/differential thermogravimetric tests, are calcium silicate hydrate gels and hydrotalcite-like phases. The primary hydration products for MgO-fly ash solidified soils are magnesium silicate hydrate gels and Mg(OH)(2). That is just the intrinsic reason why the microstructure of solidified soils becomes much denser and the mechanical behavior is significantly improved. The minor carbonate phases such as magnesium carbonate and/or calcite are also observed in reactive MgO-slag and MgO-fly ash solidified soils, depending on the period of exposure to air. The curing temperature and binder amount are proved to be the two major factors governing the hydration process of reactive MgO-slag and MgO-fly ash blends. A higher curing temperature and binder amount can generate more hydration products, but their chemical compositions such as accurate element ratios need to be investigated in the future study.

    • The lamprey central nervous system has been used extensively as a model system for investigating the networks underlying vertebrate motor behavior. The locomotor networks can be activated by application of glutamate agonists, such as N-methyl-D-aspartic acid (NMDA), to the isolated spinal cord preparation. Many spinal neurons are capable of generating pacemaker-like membrane potential oscillations upon activation of NMDA receptors. These oscillations rely on the voltage-dependent properties of NMDA receptors in interaction with voltage-dependent potassium and calcium-dependent potassium (KCa) channels, as well as low voltage-activated calcium channels. Upon membrane depolarization, influx of calcium will activate KCachannels, which in turn, will contribute to repolarization and termination of the depolarized phase. The appearance of the NMDA-induced oscillations varies markedly between spinal cord preparations; they may either have a pronounced, depolarized plateau phase or be characterized by a short-lasting depolarization lasting approximately 200–300 ms without a plateau. Both types of oscillations increase in frequency with increased concentrations of NMDA. Here, we characterize these two types of membrane potential oscillations and show that they depend on the level of endogenous release of 5-HT in the spinal cord preparations. In the lamprey, 5-HT acts to block voltage-dependent calcium channels and will thereby modulate the activity of KCachannels. When 5-HT antagonists were administered, the plateau-like oscillations were converted to the second type of oscillations lacking a plateau phase. Conversely, plateau-like oscillations can be induced or prolonged by 5-HT agonists. These properties are most likely of significance for the modulatory action of 5-HT on the spinal networks for locomotion.

  • Green synergy: Eco-friendly, high-performance anti-corrosion and wear-resistant coatings utilizing organosolv lignin and polydimethylsiloxane

    • Anti-corrosion and anti-wear coatings provide an effective solution. However, traditional coatings are often fossil-based and contain heavy metals, posing environmental concerns. The drive for eco-friendly coatings has led to the exploration of green materials. This study combined lignin, an abundant organic material, and polydimethylsiloxane (PDMS), a known hydrophobic material, to address the challenges. Organosolv lignin was functionalised with (3-Aminopropyl)triethoxysilane (APTES), then chemically grafted on PDMS for the final coating synthesis. The optimised coating achieved through an eco-friendly process, exhibiting enhanced hydrophobicity and barrier properties, showing excellent long-term corrosion resistance in NaCl solution. The optimal coating formulation contained 15 wt% lignin and 40 wt% PDMS, demonstrating a high corrosion resistance (measured impedance of 1010 Ω·cm2), which remains effective even after 3 weeks of immersion in 1 M NaCl solution. This coating also showed good wear resistance, with a low friction coefficient evident from nano scratch tests.
