  • Lipid-related markers and cardiovascular disease prediction
  • 2012
  • Ingår i: JAMA : the journal of the American Medical Association. - : American Medical Association (AMA). - 1538-3598 .- 0098-7484. ; 307:23, s. 2499-506
  • Tidskriftsartikel (refereegranskat)abstract
    • CONTEXT: The value of assessing various emerging lipid-related markers for prediction of first cardiovascular events is debated. OBJECTIVE: To determine whether adding information on apolipoprotein B and apolipoprotein A-I, lipoprotein(a), or lipoprotein-associated phospholipase A2 to total cholesterol and high-density lipoprotein cholesterol (HDL-C) improves cardiovascular disease (CVD) risk prediction. DESIGN, SETTING, AND PARTICIPANTS: Individual records were available for 165,544 participants without baseline CVD in 37 prospective cohorts (calendar years of recruitment: 1968-2007) with up to 15,126 incident fatal or nonfatal CVD outcomes (10,132 CHD and 4994 stroke outcomes) during a median follow-up of 10.4 years (interquartile range, 7.6-14 years). MAIN OUTCOME MEASURES: Discrimination of CVD outcomes and reclassification of participants across predicted 10-year risk categories of low (<10%), intermediate (10%-<20%), and high (>/=20%) risk. RESULTS: The addition of information on various lipid-related markers to total cholesterol, HDL-C, and other conventional risk factors yielded improvement in the model's discrimination: C-index change, 0.0006 (95% CI, 0.0002-0.0009) for the combination of apolipoprotein B and A-I; 0.0016 (95% CI, 0.0009-0.0023) for lipoprotein(a); and 0.0018 (95% CI, 0.0010-0.0026) for lipoprotein-associated phospholipase A2 mass. Net reclassification improvements were less than 1% with the addition of each of these markers to risk scores containing conventional risk factors. We estimated that for 100,000 adults aged 40 years or older, 15,436 would be initially classified at intermediate risk using conventional risk factors alone. Additional testing with a combination of apolipoprotein B and A-I would reclassify 1.1%; lipoprotein(a), 4.1%; and lipoprotein-associated phospholipase A2 mass, 2.7% of people to a 20% or higher predicted CVD risk category and, therefore, in need of statin treatment under Adult Treatment Panel III guidelines. CONCLUSION: In a study of individuals without known CVD, the addition of information on the combination of apolipoprotein B and A-I, lipoprotein(a), or lipoprotein-associated phospholipase A2 mass to risk scores containing total cholesterol and HDL-C led to slight improvement in CVD prediction.

  • Development and initial characterization of a novel ghrelin receptor CRISPR/Cas9 knockout wistar rat model
  • 2019
  • Ingår i: International Journal of Obesity. - : Nature Pub. Group. - 0307-0565 .- 1476-5497. ; 43:2, s. 344-354
  • Tidskriftsartikel (refereegranskat)abstract
    • Ghrelin, a stomach-derived hormone implicated in numerous behaviors including feeding, reward, stress, and addictive behaviors, acts by binding to the growth hormone secretagogue receptor (GHSR). Here, we present the development, verification, and initial characterization of a novel GHSR knockout (KO) Wistar rat model created with CRISPR genome editing.

  • Correlative Microstructure Analysis and In Situ Corrosion Study of AISI 420 Martensitic Stainless Steel for Plastic Molding Applications
  • 2017
  • Ingår i: Journal of the Electrochemical Society. - : Electrochemical Society. - 0013-4651 .- 1945-7111. ; 164:4, s. C85-C93
  • Tidskriftsartikel (refereegranskat)abstract
    • In this work, the corrosion behavior of tempered AISI 420 martensitic stainless steel (MSS) was studied by in-situ atomic force microscopy (AFM) in 0.1M NaCl and correlated with the microstructure. Thermocalc simulation, dilatometry, and X-ray diffraction (XRD) were performed to investigate phase transformation which showed the formation of M3C, M7C3, and M23C6 type of carbides and also retained austenite. Optical microscopy, scanning electron microscopy (SEM), and AFM characterization revealed undissolved carbides and tempering carbides in the martensitic matrix. Volta potential mapping measured by scanning Kelvin probe force microscopy (SKPFM) indicated higher electrochemical (practical) nobility of the carbides with respect to the martensitic matrix whereas regions adjacent to carbides showed lower nobilities due to chromium depletion. Open circuit potential and cyclic potentiodynamic polarization measurements showed metastable corrosion activities associated with a weak passive behavior and a risk for localized corrosion along certain carbide boundaries. In-situ AFM measurements revealed selective dissolution of certain carbide interphases and martensitic inter-lath regions indicating higher propensity to localized corrosion.

  • Halo excitations in fragmentation of He-6 at 240 MeV/u on carbon and lead targets
  • 2000
  • Ingår i: Nuclear Physics A. - 0375-9474. ; 669:1-2, s. 51-64
  • Tidskriftsartikel (refereegranskat)abstract
    • Dissociation of a 240 MeV/u beam of He-6, incident on carbon and lead targets, has been studied in kinematically complete experiments to investigate low-lying excitation modes in the halo nucleus He-6. It is shown that alignment effects characterize the inelastic scattering and allow an unambiguous assignment of the spin of a narrow resonance observed in the excitation energy spectrum. The differential cross sections for the He-6 inelastic scattering on carbon and lead targets were deduced from the measured moments of the two neutrons and the a-particle. An analysis of these distributions shows that quadrupole and, possibly, monopole excitations characterize the hadronic interaction, while the dipole mode is dominating in Coulomb dissociation. Neither theoretically predicted new resonance states in He-6 nor nuclear excitation of a dipole mode were found. Direct evidence has been obtained for strong suppression of Coulornb post-acceleration in direct Coulomb breakup in a lead target.
