  • Hypoxia increases LDL oxidation and expression of 15-lipoxygenase-2 in human macrophages

    • OBJECTIVE: Macrophage-mediated oxidation of low-density lipoprotein (LDL) by enzymes, such as the lipoxygenases, is considered of major importance for the formation of oxidized LDL during atherogenesis. Macrophages have been identified in hypoxic areas in atherosclerotic plaques. METHODS AND RESULTS: To investigate the role of hypoxia in macrophage-mediated LDL oxidation, we incubated human monocyte-derived macrophages with LDL under normoxic (21% O2) or hypoxic (0% O2) conditions. The results showed that hypoxic macrophages oxidized LDL to a significantly higher extent than normoxic cells. Interestingly, the mRNA and protein expression of 15-lipoxygenase-2 (15-LOX-2) as well as the activity of this enzyme are elevated in macrophages incubated at hypoxia. Both the unspliced 15-LOX-2 and the spliced variant 15-LOX-2sv-a are found in macrophages. In addition, 15-LOX-2 was identified in carotid plaques in some macrophage-rich areas but was only expressed at low levels in nondiseased arteries. CONCLUSIONS: In summary, these observations show for the first time that 15-LOX-2 is expressed in hypoxic macrophages and in atherosclerotic plaques and suggest that 15-LOX-2 may be one of the factors involved in macrophage-mediated LDL oxidation at hypoxia.

    • BACKGROUND: The study aimed to investigate patient and spouse perception of cognitive functioning 1 to 2 years after coronary artery bypass grafting. METHODS: Seventy-six married patients who had undergone coronary artery bypass grafting were selected and sex- and age-matched with 75 concurrent married patients who had undergone percutaneous transluminal coronary angioplasty. Couples received a letter of explanation and then completed telephone interviews. Forty-seven questions assessed memory, concentration, general health, social functioning, and emotional state. Response choices were: improved, unchanged, or deteriorated function after coronary artery bypass grafting/percutaneous transluminal coronary angioplasty. RESULTS: Patients who had undergone coronary artery bypass grafting did not differ in subjective ratings on any measure from patients who had undergone percutaneous transluminal coronary angioplasty. There were no differences between spouses in the respective groups; spouse ratings also did not differ from patient ratings. Only in memory function did patients and spouses report a postprocedural decline. CONCLUSIONS: No subjective differences were found in patients who had undergone either coronary artery bypass grafting or percutaneous transluminal coronary angioplasty. Spouse ratings agreed with each other and with patient ratings. Positive correlations were found between the questionnaire factors, suggesting that perceived health and well-being are associated with subjective cognition.

    • This paper deals with physical modelling and control of dynamic foaming in the LD-converter process. An experimental setup consisting of a water model, DSP and PC hardware is built up and shown to be useful for studying dynamic foaming. Furthermore, a foam height estimation algorithm is presented and validated through experiments. Finally, sound signals from the LD-converter and water model are compared and similarities between them are found.

    • Although the Normalized Difference Vegetation Index (NDVI) time-series data, derived from NOAA/AVFIRR, SPOT/VEGETATION, TERRA or AQUA/MODIS, has been successfully used in research regarding global environmental change, residual noise in the NDVI time-series data, even after applying strict pre-processing, impedes further analysis and risks generating erroneous results. Based on the assumptions that NDVI time-series follow annual cycles of growth and decline of vegetation, and that clouds or poor atmospheric conditions usually depress NDVI values, we have developed in the present study a simple but robust method based on the Savitzky-Golay filter to smooth out noise in NDVI time-series, specifically that caused primarily by cloud contamination and atmospheric variability. Our method was developed to make data approach the upper NDVI envelope and to reflect the changes in NDVI patterns via an iteration process. From the results obtained by applying the newly developed method to a 10-day MVC SPOT VGT-S product, we provide optimized parameters for the new method and compare this technique with the BISE algorithm and Fourier-based fitting method. Our results indicate that the new method is more effective in obtaining high-quality NDVI time-series.

    • Time-series of NASA/NOAA Pathfinder AVHRR Land (PAL) data have been analysed to extract parameters describing the seasonality of vegetation in Africa. Two methods have been developed to fit smooth curves to the time-series. The first method is based on an adaptive Savitzky-Golay filtering technique, and the second on non-linear least-squares fits of asymmetric Gaussian model functions. Both processing methods involve a preliminary definition of the number and timing of growing seasons using a least-squares fit of sinusoidal functions and a second order polynomial. The fit to the sinusoidal functions is used to determine the type of seasonal pattern (uni-modal or bi-modal) and to obtain starting values for the non-linear Gaussian function fits to the data. The processing incorporates qualitative information on cloudiness from the CLAVR dataset. The resulting smooth curves are used for defining parameters describing the growing seasons. The method has been applied to PAL NDVI data, and resulting imagery have been generated that show parameters such as beginnings and ends of seasons, seasonal integrated NDVI, seasonal amplitudes etc. The results indicate that the two methods complement each other and that they may be suitable in different areas depending on the behaviour of the NDVI signal.

    • Objective: Glucose degradation products (GDPs) in peritoneal dialysis (PD) fluids are cytotoxic and affect the survival of the peritoneal membrane. One of the most reactive GDPs in PD fluids is 3,4-dideoxyglucosone-3-ene (3,4-DGE). 3,4-DGE has been reported as an intermediate between 3-deoxyglucosone (3-DG) and 5-hydroxymethyl furaldehyde (5-HMF) during degradation of glucose. In PD fluids, 3,4-DGE exists in a temperature-dependent equilibrium with a pool of unidentified substances. The aim of this study was to explore this equilibrium and its temperature dependence during the first months of storage after the sterilization procedure. Methods: GDPs and inhibition of cell growth (ICG) were measured directly after sterilization of the PD fluid and during storage at different temperatures for 60 days. The following GDPs were analyzed: 3-DG, 3,4-DGE, 5-HMF, formaldehyde, acetaldehyde, glyoxal, and methylglyoxal. Results: Immediately after sterilization, the concentration of 3,4-DGE was 125 mumol/L. During the first weeks of storage, it decreased by about 80%. At the same time, the 3-DG concentration increased. None of the other GDPs were significantly affected. Cytotoxicity correlated well with the concentration of 3,4-DGE. When pure 3,4-DGE was substituted for the lost amount of 3,4-DGE after 30 days of storage, the initial ICG was almost completely regained. Conclusions: Heat sterilization of PD fluids promotes the formation of large quantities of 3,4-DGE, rendering the fluid highly cytotoxic. During storage, the main part of 3,4-DGE is reversibly converted in a temperature-dependent manner to a less cytotoxic pool, consisting mainly of 3-DG. Cytotoxicity seems to be dependent exclusively on 3,4-DGE. In order to avoid higher levels of 3,4-DGE concentrations, PD fluids should not be used too soon after sterilization and should not be stored at temperatures above room temperature.

    • The serum and cerebrospinal fluid (CSF) pathology of patients with sudden sensorineural hearing loss (SHL), both seropositive and seronegative to Borrelia burgdorferi (Bb), was prospectively studied. Nineteen consecutive patients were included and trends between the degree of hearing recovery and serum/CSF pathology and given therapy were examined. The pilot study showed a high prevalence (68%) of pathology in serum and CSF in patients with SHL. In 54% of the patients, elevated levels of CSF proteins and/or pathological CSF cell counts were present without positive antibodies to Bb. Positive levels of antibodies against Bb or pathological proteins in CSF were associated with better hearing recovery (means of 47.2 and 51.7%, respectively). The audiometric configuration "high frequency sloping" hearing impairment was associated with the lowest degree of hearing recovery. Patients with SHL and positive serology to Bb who received antibiotic treatment (oral tetracycline), with or without steroids, had the best hearing recovery in this study (61.7 and 48.4%, respectively). In conclusion, we found a high prevalence of serum and CSF pathology in a consecutive group of patients with SHL. Early appropriate antibiotic treatment may prevent the development of major late complications of Lyme disease/borreliosis. We also find it justified to perform more general serological analyses, including CSF analysis, in patients with SHL. A more liberal approach to testing and development of test protocols for SHL patients will increase our knowledge in this field.

    • The role of the Laude g-factor in atomic spectroscopy is explored through theoretical studies for Ne I and Ne II where some experimental values are available. Extended correlation studies in the Breit-Pauli approximation for spectrum calculations are analysed and compared with experiment and other theory. For Ne II, the multiconfiguration Dirac-Hartree-Fock method was also applied, but correlation was restricted only to the effect of core-polarization on the outer electron.
