  • Suppression of the breakthrough of human immunodeficiency virus type 1 (HIV-1) in cell culture by thiocarboxanilide derivatives when used individually or in combination with other HIV-1-specific inhibitors (i.e., TSAO derivatives)

    • Five structurally related thiophene and furane analogues of the oxathiin carboxanilide derivative NSC 615985 (UC84) (designated UC10, UC68, UC81, UC42, and UC16) were identified as potent inhibitors of HIV-1 replication in cell culture and HIV-1 reverse transcriptase activity. These compounds were markedly active against a series of mutant HIV-1 strains, containing the Leu-100-->Ile, Val-106-->Ala, Glu-138-->Lys, or Tyr-181-->Cys mutations in their reverse transcriptase. However, the thiocarboxanilide derivatives selected for mutations at amino acid positions 100 (Leu-->Ile), 101 (Lys-->Ile/Glu), 103 (Lys-->Thr/Asp) and 141 (Gly-->Glu) in the HIV-1 reverse transcriptase. The compounds completely suppressed HIV-1 replication and prevented the emergence of resistant virus strains when used at 1.3-6.6 microM--that is, 10- to 25-fold lower than the concentration required for nevirapine and bis(heteroaryl)piperazine (BHAP) U90152 to do so. If UC42 was combined with the [2',5'-bis-O-(tert-butyldimethylsilyl)-3'-spiro-5"-(4"-amino-1",2"- oxathiole-2",2"-dioxide)]-beta-D-pentofuranosyl (TSAO) derivative of N3-methylthymine (TSAO-m3T), virus breakthrough could be prevented for a much longer time, and at much lower concentrations, than if the compounds were used individually. Virus breakthrough could be suppressed for even longer, and at lower drug concentrations, if BHAP was added to the combination of UC42 with TSAO-m3T, which points to the feasibility of two- or three-drug combinations in preventing virus breakthrough and resistance development.

  • Emerging streams of intitutional theory in accounting research : current status and future directions

    • More than 30 years have passed since first calls to see accounting as social and institutional practice (Burchell et al., 1980; Hopwood, 1987; Hopwood & Miller, 1994). During these years, a significant number of theories have been striving to extend and challenge the hegemony of particular ways of seeing accounting (Jacobs, 2012) with acknowledging the need for inter-disciplinary approach and theoretical pluralism in contemporary studies. Within this theoretical pluralism, the institutional theory was recognized as one of the dominating theories in accounting research during last decades (Jacobs, 2012; Lounsbury, 2008; Modell, 2015).However, despite acknowledging the paradigmatic developments of institutional theory into several streams in organisational science (DiMaggio, 1988; Lawrence et al., 2009, 2011; Oliver, 1991; Thornton et al., 2012), few attempts were done to make a separate review of these  streams and their value for accounting literature (Modell, 2009). Here, influential work of Lounsbury (2008) rather reflected organizational science literature with seeing accounting as a context which can give crucial insights into institutional and practice theories rather than vice versa. Jacobs (2012) in his study of theoretical pluralism in accounting research during 1992-2008 acknowledged the institutional theory domination but made few reflections on its progressions and implications for seeing accounting as social and institutional practice. And finally, more recent studies of Modell and his colleagues (Modell, 2015; Modell et al., 2017) made overviews of two streams of institutional theory (e.g. logic and work) and institutional theory combination possibility with other theories. While giving the valuable insights into understanding of issues related to theories combinations in accounting, these studies hardly review the accounting literature related to institutional theory streams which were criticised for apolitical nature but at the same time acknowledged as self-sufficient.Therefore, it seems that current studies rather ignore or criticize specific institutional theory streams, jumping into possible theory combinations agenda without revealing the value of these streams for accounting research. Such tendency may potentially lead to marginalization of accounting knowledge (Jacobs, 2016; Jacobs & Cuganesan, 2014) with the use of these streams under the quest for contribution to the interdisciplinary literature.In this regard, this paper aims to reflect upon the emerging streams of institutional theory, their application and influence on the accounting research of the last decade. Specifically, the paper’s ambition is to review and understand the role of institutional logics (IL), institutional entrepreneurship (IE) and institutional work (IW) in accounting research. These streams of institutional theory become increasingly valuable in accounting research for their explanatory capacities. But at the same time, but their application, especially in a combined form,  can also raise criticalities within theoretical pluralism agenda. Therefore, we construct our research questions as:How have the emerging stream of institutional theory (IL, IE and IW) been used by accounting scholars?How have these streams influenced accounting research? What could be done in the future accounting research using emerging streams  of institutional theory?In order to answer these research questions, we compile a structured literature review (Massaro et al., 2016) of articles published in scientific journals in the accounting field (levels 4 and 3 in ABS list). As a frame for analysis of the paper, we use conceptual clarification and analytical framework proposed by Lukka and Vinnari (2014) who made a distinction between domain theory and method theory in order to capture the relations between common knowledge about accounting as a social practice and the theories used to extend this knowledge. Applying to our paper, domain theory refers particularly to a set of knowledge about accounting as social and institutional practice. Method theory (i.e. IL, IE, IW) is a meta-level conceptual system, which can offer alternative perspectives and add new understanding/insights into domain theory. Following this analytical distinction, we examine the way how current accounting literature utilizes the selected institutional streams in domain-method dimension, how the theory or their combination (if any) are justified and used in a flow of argument for understanding of accounting practice.

    • Despite the introduction of 2-(2-nitro-4-trifluormethyl-benzoyl)-1,3-cyclohexandion into the treatment of hereditary tyrosinemia type I (HT1), patients remain at risk of developing hepatocellular carcinoma (HCC). Serial total alpha-fetoprotein (AFP) levels are used to monitor the individual patient. Lectin-reactive alpha-fetoprotein (L3-AFP) is an AFP isoform that is expressed by malignant liver tumors. We investigated whether the analysis of L3-AFP could lead to earlier detection of HCC in HT1 compared with judgement based on total AFP alone. AFP electrophoresis using lectin-containing agarose gel identifies L3-AFP by the affinity of its specific carbohydrate chain to lectin. We report the retrospective analysis of sequential serum samples of 12 patients with HT1 and histologically proven HCC. AFP isoforms could be identified in all 12 patients. In 6 patients, the L3-AFP increased before the total AFP. In 3 patients, the rise in L3-AFP was parallel to the rise of total AFP; and in 3 patients, the L3-AFP was raised after the total AFP or did not increase at all. We were able to identify 6 of 12 patients with an early increase in the new tumor marker. Lectin-affinity electrophoresis may have a role in discriminating benign liver disease from HCC in HT1. We suggest the further evaluation of L3-AFP in HT1.

  • The State of the Art of Spatial Interfaces for 3D Visualization

    • We survey the state of the art of spatial interfaces for 3D visualization. Interaction techniques are crucial to data visualization processes and the visualization research community has been calling for more research on interaction for years. Yet, research papers focusing on interaction techniques, in particular for 3D visualization purposes, are not always published in visualization venues, sometimes making it challenging to synthesize the latest interaction and visualization results. We therefore introduce a taxonomy of interaction technique for 3D visualization. The taxonomy is organized along two axes: the primary source of input on the one hand and the visualization task they support on the other hand. Surveying the state of the art allows us to highlight specific challenges and missed opportunities for research in 3D visualization. In particular, we call for additional research in: (1) controlling 3D visualization widgets to help scientists better understand their data, (2) 3D interaction techniques for dissemination, which are under-explored yet show great promise for helping museum and science centers in their mission to share recent knowledge, and (3) developing new measures that move beyond traditional time and errors metrics for evaluating visualizations that include spatial interaction.

    • Providing functionally effective sensory feedback to users of prosthetics is a largely unsolved challenge. Traditional solutions require high band-widths for providing feedback for the control of manipulation and yet have been largely unsuccessful. In this study, we have explored a strategy that relies on temporally discrete sensory feedback that is technically simple to provide. According to the Discrete Event-driven Sensory feedback Control (DESC) policy, motor tasks in humans are organized in phases delimited by means of sensory encoded discrete mechanical events. To explore the applicability of DESC for control, we designed a paradigm in which healthy humans operated an artificial robot hand to lift and replace an instrumented object, a task that can readily be learned and mastered under visual control. Assuming that the central nervous system of humans naturally organizes motor tasks based on a strategy akin to DESC, we delivered short-lasting vibrotactile feedback related to events that are known to forcefully affect progression of the grasp-lift-and-hold task. After training, we determined whether the artificial feedback had been integrated with the sensorimotor control by introducing short delays and we indeed observed that the participants significantly delayed subsequent phases of the task. This study thus gives support to the DESC policy hypothesis. Moreover, it demonstrates that humans can integrate temporally discrete sensory feedback while controlling an artificial hand and invites further studies in which inexpensive, noninvasive technology could be used in clever ways to provide physiologically appropriate sensory feedback in upper limb prosthetics with much lower band-width requirements than with traditional solutions.

  • Marketing the Past: A Literature Review and Future Directions for Researching Retro, Heritage, Nostalgia, and Vintage

    • Over the last decades, marketing literature has persistently been interested in how marketers and consumers alike engage with the past through past-themed brands, products, and services. However, our knowledge on past-themed marketing is scattered and treated in largely separated literature streams on retro, heritage, nostalgia, and vintage. This article reviews marketing literature on retro, heritage, nostalgia, and vintage – and their ontological, methodological, and axiological underpinnings – to synthesise a differential overview of these streams and contribute shared future research directions for researching and understanding past-themed marketing. Thus, based on our review, we identify and discuss areas for future research on marketing the past, specifically concerned with authenticity complexities and marketing management perspectives.

  • GLI1 modulation of the output of hedgehog signaling in human cancer

    • The Hedgehog (HH) signaling pathway has critical roles in embryonic development and tumorigenesis. Aberrant activation of HH signaling has been known to be involved in several types of malignant tumors. GLI1 is a transcription factor that acts not only as the terminal effector but also represents a pathway target gene, amplifying the HH signal. GLI1 expression/activity is modulated at different levels of gene regulation. The aim of the thesis is to explore the functional alterations in GLI1 transcriptional activity and further investigate the consequences on the output of HH signaling. Paper I. We discovered an antisense transcript, which is a long non-coding RNA located head-to-head with the gene encoding GLI1, and termed it GLI1 antisense (GLI1AS). The expression of GLI1AS in cancer cells was concordant with GLI1 levels. GLI1AS knockdown upregulated GLI1, increased cellular proliferation and tumor growth in a xenograft model system. GLI1AS overexpression decreased the levels of GLI1, its target genes and cellular proliferation. Additionally, we demonstrated that GLI1 knockdown reduced GLI1AS, while GLI1 overexpression increased GLI1AS, highlighting a regulatory feedback loop on GLI1/GLI1AS expression. GLI1AS acts by modifying the chromatin landscape and reduces the recruitment of RNA polymerase II at the GLI1/GLI1AS locus. Paper II. We examined the possible interplay of S6K1 and GLI1 signaling in neuroblastoma. mTOR/S6K1 signaling is known to have a role in the development of this disease and recent evidence also implicates the HH pathway. In this paper, it is shown that knockdown of either S6K1 or GLI1 reduces the cellular proliferation of neuroblastoma cells. However, there is little effect of S6K1 on the expression of GLI1 mRNA and protein and on the capacity of GLI1 to activate target genes. No detectable phosphorylation of GLI1 is observed prior or following S6K1 knockdown. Moreover, no additional growth inhibitory effects were detected when PI3K/mTOR and GLI signaling inhibitors were combined. Our results demonstrate that the impact of S6K1 kinase on neuroblastoma cells is not mediated through modulation of GLI1 expression/activity. Paper III. We investigated HH signaling activation in breast cancer and the contribution of GLI1 to tamoxifen resistance. Our results showed that GLI1 expression was higher in tamoxifen resistant compared to sensitive cells. Tamoxifen resistant cells had stronger ERα transcriptional activity relative to sensitive cells, even though the ERα expression was similar in both cell types. Knockdown of GLI1 attenuated cell proliferation and reduced ERα transcriptional activity in both resistant and sensitive cells, irrespective of estrogen stimulation. Moreover, a positive correlation between GLI1 and ERα expression was identified in breast cancer samples. High GLI1 expression predicted worse distant metastasis-free survival in breast cancer patients. These data suggest that the HH pathway may be a new candidate for therapeutic targeting and prognosis in ERα-positive breast cancer. Paper IV. We explored the common and differential target genes of GLI1 and edited GLI1, using single molecule RNA sequencing (RNA-seq), via overexpression and siRNA-mediated depletion approaches in rhabdomyosarcoma Rh36 cells. GO analysis revealed that GLI1 and edited GLI1 are involved in developmental and metabolic processes, cellular proliferation, KEGG pathways in cancer, basal cell carcinomas and thyroid cancer. 37 genes were differentially upregulated by edited GLI1, while 29 common target genes, including FOXS1, SOSTDC1 and SOX18, were identified by RNA-seq combined with correlation analysis to the FANTOM5 dataset. SOSTDC1 and FOXS1 expression was also modulated in HH signaling responsive medulloblastoma Daoy cells by GLI1 knockdown and Smoothened agonist (SAG) treatment. Reciprocally, GLI1 was downregulated in both Rh36 and Daoy cells following FOXS1 knockdown, highlighting a FOXS1/GLI1 regulatory loop.

    • Introduction: Obstructive Sleep Apnea (OSA) is a common co-morbidity in obese patients. OSA is also frequently associated with various metabolic complications. In this study, we have evaluated the associations between measures of OSA and abdominal and liver fat subjects with untreated OSA. Methods: A total of 470 subjects (73% females) were examined during a screening process at the Obesity unit at the Sahlgrenska University Hospital. OSA was determined by the ApneaLink system and visceral and liver fat content were determined by CT using a two slice technique at the liver and L4-5 level. The included subjects had a mean age of 42.4 years (SD: 13.5 years), mean weight of 116.6 kg (SD: 20.3 kg), and a mean BMI of 40.8 kg/m2 (SD: 5.7 kg/m2). From the ApneaLink examinations the Apnea - Hypopnea Index (AHI), Respiratory Distress Index (RDI), Oxygen Desaturation Index (ODI) and mean oxygen saturation (SO2) was determined. From the CT examinations visceral adipose tissue mass (VAT) and hepatic fat content (HFC) was determined. Results: VAT was strongly correlated to AHI, RDI, ODI, and SO2 (r = 0.397, 0.388, 0.449, and )0.424 respectively, P < 0.001). There was also a correlation between HFC and AHI, RDI, ODI, and SO2 (r = 0.193, 0.198, 0.214, and 0.173 respectively, P < 0.001). Conclusion: The present study demonstrates that untreated OSA in severe obesity is associated to both measures of visceral fat and hepatic fat content. Evaluations of abdominal fat content should be considered in obese subjects with OSA.
