  • Cell-based therapies for acute liver failure: human tissue engineering, pure cell implantation and stem cell application
  • 2007
  • Ingår i: Journal of Organ Dysfuntion. - : Informa UK Limited. - 1747-1060 .- 1747-1079. ; 3:4, s. 194-203
  • Tidskriftsartikel (refereegranskat)abstract
    • Improving the survival rate and quality of life of patients with acute and chronic hepatic dysfunction still represents a challenge. Cell-based therapeutic hepatocyte transplantation has great potential to serve as a temporary support and “bridge” prior to a donor organ becoming available, or even as therapy for replacing a compromised liver. In this review we describe potential applications of human tissue engineering, compare therapeutic effects of transplanted hepatocytes, with or without tissue engineering and highlight the exciting progress of stem cell transplantation in hepatic dysfunction. Hepatocytes can be transplanted in the form of pure cells or as cells engineered with devices (with or without pretreatment with growth factors), freshly isolated or preserved cells or adult matured or stem cells. The efficacy of different forms of cell-based therapy may be associated with the severity and duration of the disease and with the therapeutic strategy used to treat it.

  • Evidence for improved outcome following use of hemostatic fibrin sealants in HPB surgery?
  • 2012
  • Ingår i: European Journal of Trauma and Emergency Surgery. - : Springer Science and Business Media LLC. - 1863-9933 .- 1863-9941. ; 38:1, s. 33-36
  • Forskningsöversikt (refereegranskat)abstract
    • The development of fibrin sealants has been progressing; they are now often applied as fibrinogen-coated collagen patches. The concept has been increasingly applied in hepato-pancreato-biliary surgery, for example, following liver resection, and to some extent also in pancreatic surgery, both in order to reinforce the pancreatic anastomosis at pancreaticoduodenectomy and to seal the pancreatic stump following distal pancreatectomy. High quality evidence in the form of major prospective, randomized clinical studies is still lacking on in the field of HPB. There is also an the absence of proper cost-utility analyses.

  • Gemcitabine chemoresistance in pancreatic cancer: Molecular mechanisms and potential solutions.
  • 2009
  • Ingår i: Scandinavian Journal of Gastroenterology. - : Informa UK Limited. - 1502-7708 .- 0036-5521. ; 44, s. 782-786
  • Tidskriftsartikel (refereegranskat)abstract
    • Ductal pancreatic adenocarcinoma is associated with a very poor prognosis and most patients are given palliative care. Chemotherapy in the form of gemcitabine has been found to reduce disease-related pain, and the otherwise frequently occurring weight changes, to increase Karnofsky performance status and quality of life and has also resulted in a modest improvement in survival time. The intracellular uptake of gemcitabine is dependent on nucleoside transporters, predominantly human equilibrative nucleoside transporter-1 (hENT-1), which is over-expressed in human pancreatic adenocarcinoma cells. Cellular resistance to gemcitabine can be intrinsic or acquired during gemcitabine treatment. One of the mechanisms is a decrease in hENT-1 expression. Modifications of gemcitabine not rendering it dependent on the nucleoside transporter may be a successful future mode of chemotherapy treatment, and determination of the nucleoside receptor status at the time of diagnosis could potentially also contribute to a more targeted therapy in the future.

  • Gut barrier failure in critical illness: lessons learned from acute pancreatitis
  • 2006
  • Ingår i: Journal of Organ Dysfuntion. - : Informa UK Limited. - 1747-1060 .- 1747-1079. ; 2:2, s. 93-100
  • Tidskriftsartikel (refereegranskat)abstract
    • Gut barrier function is essential in critical illness and contributes to the systemic inflammatory response. Failure of the gut barrier includes both changes in microbial ecology, permeability and potential translocation, as well as local gut inflammatory response. The present review summarizes experiences made from acute pancreatitis, including pathophysiological mechanisms and ways of intervention.
