    • The present study examines pyruvate kinase-conjugated antibodies for potential use in EUSA applications. The conjugates had an acceptable stability, and the coupling inflicted only minor impairment on the kinase activity. To mimic the setup of an immunoassay under development, a test antigen (BSA) was attached to polystyrene nanoparticles. This arrangement was found to be suitable as solid support for presentation of antigens in sensitive bioluminescence assays. The nanoparticles were well characterized in terms of protein surface load and were used to establish the number of conjugate complexes needed to generate a detectable signal. Under the biochemical conditions employed here, the detection limit of the pyruvate kinase conjugate lies in the femtomole range.

  • Immune-Proteome Profiling in Classical Hodgkin Lymphoma Tumor Diagnostic Tissue

    • In classical Hodgkin Lymphoma (cHL), immunoediting via protein signaling is key to evading tumor surveillance. We aimed to identify immune-related proteins that distinguish diagnostic cHL tissues (=diagnostic tumor lysates, n = 27) from control tissues (reactive lymph node lysates, n = 30). Further, we correlated our findings with the proteome plasma profile between cHL patients (n = 26) and healthy controls (n = 27). We used the proximity extension assay (PEA) with the OlinkTM multiplex Immuno-Oncology panel, consisting of 92 proteins. Univariate, multivariate-adjusted analysis and Benjamini–Hochberg’s false discovery testing (=Padj) were performed to detect significant discrepancies. Proteins distinguishing cHL cases from controls were more numerous in plasma (30 proteins) than tissue (17 proteins), all Padj < 0.05. Eight of the identified proteins in cHL tissue (PD-L1, IL-6, CCL17, CCL3, IL-13, MMP12, TNFRS4, and LAG3) were elevated in both cHL tissues and cHL plasma compared with control samples. Six proteins distinguishing cHL tissues from controls tissues were significantly correlated to PD-L1 expression in cHL tissue (IL-6, MCP-2, CCL3, CCL4, GZMB, and IFN-gamma, all p ≤0.05). In conclusion, this study introduces a distinguishing proteomic profile in cHL tissue and potential immune-related markers of pathophysiological relevance

  • LED street lighting: A power quality comparison among street light technologies

    • High-pressure sodium lamps are currently the main lamps used in public lighting. However, the possibility of using high-power light emitting diode (LEDs) for street lighting is growing continuously due to their greater energy efficiency, robustness, long life and light control. The aim of this paper is to study the power quality of high-power lighting networks based on LED and high-pressure sodium lamps. Both electromagnetic and dimmable electronic ballasts, which can dim the lamp output smoothly and uniformly, have been used connected to high-pressure sodium lamps. High-pressure sodium lamps connected to electronic equipment have been tested with different arc power levels using dimming on a 230 V power supply. The study presented in this paper is completely based on measurements, including harmonic currents in the frequency range up to 150 kHz for all the technologies. The main results show a broadband spectrum in LED lamps which confirms other research in fluorescent lamps powered by high-frequency ballasts. Results also indicate a decrease in the harmonic value with increasing harmonic order, and a decrease in the harmonic value at half load (60%) compared with full load (100%). Although total harmonic distortion of the current is lower with high-pressure sodium lamps connected to electronic rather than electromagnetic ballasts, LED lamps achieved the lowest total harmonic distortion of current.

    • Aims: Cathepsin S has been reported to be a biomarker of spinal microglial activation, a process suggested to be involved in the pathophysiology of chronic neuropathic pain. So far this has been shown only in animal experiments. The aim of this study was to investigate the concentrations of cathepsin S in human cerebrospinal fluid (CSF) samples from a well-defined patient cohort suffering from neuropathic pain as compared to controls.Methods: CSF samples from patients suffering from chronic neuropathic pain (n = 14) were analyzed for cathepsin S levels using commercial sandwich ELISAs (DY1183, R&D Systems, Minneapolis, MN, USA). Control CSF was sampled from patients undergoing minor urological surgical procedures under spinal anaesthesia (n = 70), having no obvious pain suffering.Results: The neuropathic pain group had significantly higher levels of CSF cathepsin S (median 15189 pg/mL, range 3213–40,040), than the control group (median 5911 pg/mL, range 1909–17,188) (p < 0.005, Mann–Whitney U-test).Conclusion: The results support the existence of microglial activation in chronic neuropathic pain patients. CSF Cathepsin S may serve as a potential biomarker for this specific mechanism linked to neuropathic pain. In the future, Cathepsin S inhibiting drugs might become a new treatment alternative for neurophatic pain.

  • Urinary cytokines correlate with acute kidney injury in critically ill COVID-19 patients

    • BACKGROUND: Acute kidney injury is common in COVID-19 patients admitted to the ICU. Urinary biomarkers are a non-invasive way of assaying renal damage, and so far, urinary cytokines are not fully investigated. The current study aimed to assess urinary cytokine levels in COVID-19 patients.METHODS: Urine was collected from COVID-19 patients (n = 29) in intensive care and compared to a preoperative group of patients (n = 9) with no critical illness. 92 urinary cytokines were analyzed in multiplex using the Olink Target 96 inflammation panel and compared to clinical characteristics, and urinary markers of kidney injury.RESULTS: There were strong correlations between proinflammatory cytokines and between urinary cytokines and urinary kidney injury markers in 29 COVID-19 patients. Several cytokines were correlated to kidney injury, 31 cytokines to AKI stage and 19 cytokines correlated to maximal creatinine.CONCLUSIONS: Urinary inflammatory cytokines from a wide range of immune cell lineages were significantly upregulated during COVID-19 and the upregulation correlated with acute kidney injury as well as urinary markers of kidney tissue damage.

  • The approach of Swedish municipalities to the preservation of agricultural land in a planning context

    • Agricultural land is one of the primary natural resources for human life. Climate change, peak oil, peak soil and a growing world population are factors that are likely to increase the importance of land in relation to both food production and the cultivation of energy crops. The current legal protection for agricultural land in Sweden is considered weak. The aim of this paper is to present the ‘state of art' in respect of how Swedish municipalities approach the issue of preserving agricultural land. The topic is analysed in the context of municipal spatial planning, through which policies, strategies and motives pertaining to the preservation of agricultural land are asserted. Results from three empirical studies show that a relatively high degree of the responding municipalities stated an interest in the preservation of agricultural land. However, their actual planning practice did not confirm such an approach.
