    • The thermal conductivity λ and the heat capacity per unit volume, ρcp, have been measured for a number of common lubricating oils. The oils tested were paraffinic and naphthenic mineral oils and a 50/50 blend of these. Polyα-olefin, polyglycol, Santotrac, ester and rapeseed oils have also been tested. The measurements, using the transient hot-wire method, were carried out under isothermal conditions over a pressure range from atmospheric to 1.1 GPa and at two temperatures, 295 and 380 K (22 and 107°C respectively). The temperature had only a marginal effect on thermal conductivity; however, the thermal conductivity was doubled as the pressure was increased to 1 GPa. The heat capacity per unit volume was influenced by both the pressure and the temperature. Some of the lubricants solidified as the pressure increased and the transition from a fluid to a solid state could be detected in the measurements of ρcp,The relationship between the thermodynamic properties and the pressure and temperature are described by two empirical equations. These equations can be used in thermal elastohydrodynamic analyses.

  • Impacts of elevated terrestrial nutrient loads and temperature on pelagic food-web efficiency and fish production

    • Both temperature and terrestrial organic matter have strong impacts on aquatic food-web dynamics and production. Temperature affects vital rates of all organisms, and terrestrial organic matter can act both as an energy source for lower trophic levels, while simultaneously reducing light availability for autotrophic production. As climate change predictions for the Baltic Sea and elsewhere suggest increases in both terrestrial matter runoff and increases in temperature, we studied the effects on pelagic food-web dynamics and food-web efficiency in a plausible future scenario with respect to these abiotic variables in a large-scale mesocosm experiment. Total basal (phytoplankton plus bacterial) production was slightly reduced when only increasing temperatures, but was otherwise similar across all other treatments. Separate increases in nutrient loads and temperature decreased the ratio of autotrophic:heterotrophic production, but the combined treatment of elevated temperature and terrestrial nutrient loads increased both fish production and food-web efficiency. CDOM: Chl a ratios strongly indicated that terrestrial and not autotrophic carbon was the main energy source in these food webs and our results also showed that zooplankton biomass was positively correlated with increased bacterial production. Concomitantly, biomass of the dominant calanoid copepod Acartia sp. increased as an effect of increased temperature. As the combined effects of increased temperature and terrestrial organic nutrient loads were required to increase zooplankton abundance and fish production, conclusions about effects of climate change on food-web dynamics and fish production must be based on realistic combinations of several abiotic factors. Moreover, our results question established notions on the net inefficiency of heterotrophic carbon transfer to the top of the food web.

  • Targeting p53 in vivo : a first-in-man study with the p53-targeting compound APR-246 in refractory hematologic malignancies and prostate cancer

    • PURPOSE: APR-246 (PRIMA-1MET) is a novel drug that restores transcriptional activity of unfolded wild-type or mutant p53. The main aims of this first-in-human trial were to determine maximum-tolerated dose (MTD), safety, dose-limiting toxicities (DLTs), and pharmacokinetics (PK) of APR-246.PATIENTS AND METHODS: APR-246 was administered as a 2-hour intravenous infusion once per day for 4 consecutive days in 22 patients with hematologic malignancies and prostate cancer. Acute myeloid leukemia (AML; n = 7) and prostate cancer (n = 7) were the most frequent diagnoses. Starting dose was 2 mg/kg with dose escalations up to 90 mg/kg.RESULTS: MTD was defined as 60 mg/kg. The drug was well tolerated, and the most common adverse effects were fatigue, dizziness, headache, and confusion. DLTs were increased ALT/AST (n = 1), dizziness, confusion, and sensory disturbances (n = 2). PK showed little interindividual variation and were neither dose nor time dependent; terminal half-life was 4 to 5 hours. Tumor cells showed cell cycle arrest, increased apoptosis, and upregulation of p53 target genes in several patients. Global gene expression analysis revealed changes in genes regulating proliferation and cell death. One patient with AML who had a p53 core domain mutation showed a reduction of blast percentage from 46% to 26% in the bone marrow, and one patient with non-Hodgkin's lymphoma with a p53 splice site mutation showed a minor response.CONCLUSION: We conclude that APR-246 is safe at predicted therapeutic plasma levels, shows a favorable pharmacokinetic profile, and can induce p53-dependent biologic effects in tumor cells in vivo.

    • A multianvil cell assembly with octahedral edge length 25 mm has been adapted for high pressure investigations involving water-rich environments up to 6.5 GPa and 400°C. Water-rich samples are confined in Teflon containers with a volume up to 300 mm3. Applicability tests were performed between 250 and 400°C by investigating the transformation of amorphous titania particles close to the rutile–TiO2-II (∼5 GPa) phase boundary, and the transformation of amorphous silica particles close to the quartz–coesite (∼2.5 GPa) and coesite–stishovite (∼7 GPa) phase boundaries. The performed experiments employed 25.4 mm tungsten carbide anvils with a truncation edge length of 15 mm. The sample pressure at loads approaching 820 t was estimated to be around 6.5 GPa. The large volume multianvil cell is expected to have broad and varied application areas, ranging from the simulation of geofluids to hydrothermal synthesis and conversion/crystal growth in aqueous environments at gigapascal pressures.

  • Electric resistance of single-walled carbon nanotubes under hydrostatic pressure

    • The electric resistance of single-walled nanotube mats has been studied systematically under both ambient and high hydrostatic pressures up to 1.5 GPa. Both the temperature dependence of the resistance and the magnetoresistance indicate that electrical transport occurs by variable range hopping, apparently in 2D. We suggest that this unexpected dimensionality arises from a fractal network of tubes and bundles. Under hydrostatic pressure (HP) the resistance still shows 2D variable range hopping and decreases with increasing HP. An irreversible increase in localization length and DOS is induced below 0.5 GPa. The behavior is reversible and due to strong interaction of tubes from 0.5 GPa up to 1.05 GPa. These results indicate that 2D variable range hopping occurs within bundles.

    • The study of electrical transport properties of bulk single-walled carbon nanotube (SWNT) materials attracts a lot of attention. However, the transport results differ from sample to sample[l]. Some results show semiconducting behavior with different laws and whether the behavior is dominated by on-tube or on-bundle effects or by interbundle contacts is still unknown. Therefore, further investigation is still meaningful. The effect of hydrostatic pressure(HP), which is expected to significantly change the interaction between tubes or bundles, gives us a further possibility to understand the characteristic properties. Under HP, Raman spectra and electric resistance show pronounced changes at 1.5GPa[2]. In this study, we systematically studied the electric resistance up to 1.5 GPa, giving a different image of the transport behavior.

  • Expression and Genetic Variation in Neuroendocrine Signaling Pathways in Lethal and Nonlethal Prostate Cancer among Men Diagnosed with Localized Disease

    • BACKGROUND: Recent data suggest that neuroendocrine signaling pathways may play a role in the progression of prostate cancer, particularly for early-stage disease. We aimed to explore whether expression of selected genes in the adrenergic, serotoninergic, glucocorticoid, and dopaminergic pathways differs in prostate tumor tissue from men with lethal disease compared to men with nonlethal disease.METHODS: Based on the Swedish Watchful Waiting Cohort, we included 511 men diagnosed with incidental prostate cancer through TURP during 1977-1998 with follow-up up to 30 years. For those with tumor tissue (N=262), we measured mRNA expression of 223 selected genes included in neuroendocrine pathways. Using DNA from normal prostate tissue (N=396), we genotyped 36 SNPs from 14 receptor genes. Lethal prostate cancer was the primary outcome in analyses with pathway gene expression and genetic variants.RESULTS: Differential expression of genes in the serotoninergic pathway was associated with risk of lethal prostate cancer (P=0.007); similar but weaker associations were noted for the adrenergic (P=0.014) and glucocorticoid (P=0.020) pathways. Variants of the HTR2A (rs2296972; P=0.002) and NR3CI (rs33388; P=0.035) genes (within the serotoninergic and glucocorticoid pathways) were associated with lethal cancer in over-dominant models. These genetic variants were correlated with expression of several genes in corresponding pathways (P<0.05).CONCLUSIONS: Our findings lend support to hypothesis that the neuroendocrine pathways, particularly serotoninergic pathway, are associated with lethal outcome in the natural course of localized prostate cancer.IMPACT: The current study provides evidence of the role of neuroendocrine pathways in prostate cancer progression which may have clinical utility.
