  • Identification of Flt3(+) lympho-myeloid stem cells lacking erythro-megakaryocytic potential: A revised road map for adult blood lineage commitment

    • All blood cell lineages derive from a common hematopoietic stem cell (HSC). The current model implicates that the first lineage commitment step of adult pluripotent HSCs results in a strict separation into common lymphoid and common myeloid precursors. We present evidence for a population of cells which, although sustaining a high proliferative and combined lympho-myeloid differentiation potential, have lost the ability to adopt erythroid and megakaryocyte lineage fates. Cells in the Lin-Sca-1+c-kit+ HSC compartment coexpressing high levels of the tyrosine kinase receptor Flt3 sustain granulocyte, monocyte, and B and T cell potentials but in contrast to Lin-Sca-1(+)ckit(+)Flt3(-) HSCs fail to produce significant erythroid and megakaryocytic progeny. This distinct lineage restriction site is accompanied by downregulation of genes for regulators of erythroid and megakaryocyte development. In agreement with representing a lymphoid primed progenitor, Lin(-)Sca-l(+)c-kit(+)CD34(+)Flt3(+) cells display upregulated IL-7 receptor gene expression. Based on these observations, we propose a revised road map for adult blood lineage development.

    • The sedimentary record from the Ugleelv Valley on central Jameson Land, East Greenland, adds new information about terrestrial palaeoenvironments and glaciations to the glacial history of the Scoresby Sund fjord area. A western extension of a coastal ice cap on Liverpool Land reached eastern Jameson Land during the early Scoresby Sund glaciation (approximate to the Saalian). During the following glacial maximum the Greenland Ice Sheet inundated the Jameson Land plateau from the west. The Weichselian also starts with an early phase of glacial advance from the Liverpool Land ice cap, while polar desert and ice-free conditions characterised the subsequent part of the Weichselian on the Jameson Land plateau. The two glaciation cycles show a repeated pattern of interaction between the Greenland Ice Sheet in the west and an ice cap on Liverpool Land in the east. Each cycle starts with extensive glacier growth in the coastal mountains followed by a decline of the coastal glaciation, a change to cold and arid climate and a late stage of maximum extent of the Greenland Ice Sheet.

  • Evaluation of CPC-collector designs for stand-alone, roof- or wall installation

    • An asymmetrically truncated non-tracking compound parabolic concentrator type collector design concept has been developed. The collector type has a bi-facial absorber and is optimised for northern latitudes. The concept is based on a general reflector form that is truncated to fit different installation conditions. In this paper collectors for stand-alone, roof and wall mounting are studied. Prototypes of six different collectors have been built and outdoor tested. The evaluation gave high annual energy outputs for a roof mounted collector, 925 MJ/m2, and a stand-alone collector with Teflon, 781 MJ/m2, at an operating temperature of Top = 75 °C. A special design for roofs facing east or west was also investigated and gave an annual energy output of 349 (east) and 436 (west) MJ/m2 at Top = 75 °C. If a high solar fraction over the year is the objective, a load adapted collector with a high output during spring/fall and a low output during summer can be used. Such a collector had an output of 490 MJ/m2 at Top = 75 °C. Finally a concentrating collector for wall mounting was evaluated with an estimated annual output of 194 MJ/m2 at Top = 75 °C. The concentrator design concept can also be used for concentrators for PV-modules.

  • Compact medical fluorosensor for minimally invasive tissue characterization

    • A compact fiber-optic point-measuring fluorosensor fully adapted to clinical studies is described. The system can use two excitation wavelengths, 337 and 405 nm, obtained from a nitrogen laser directly, or after dye laser conversion, respectively. The image intensifier used in the spectrometer can be gated with a variable time delay, allowing also time-resolved spectra to be extracted, with a time resolution of about 4 ns. Moreover, diffusely scattered white light can be spectrally recorded. The system is fully computer controlled enabling short recording times in clinical application, which are illustrated.

  • In vivo absorption spectroscopy of tumor sensitizers with femtosecond white light

    • A system based on a femtosecond white-light continuum and a streak camera was used for recordings of the in vivo absorption spectra of the tumor-seeking agent disulphonated aluminum phthalocyanine. Measurements for different drug doses were performed on tumor tissue (muscle-implanted adenocarcinoma) and normal muscle tissue in rats. It was found that the shape of the spectrum is tissue dependent. The peak of the absorption spectrum is blueshifted in tumor tissue as compared with the muscle. Thus the contrast in the drug-related absorption can be altered by up to a factor of 2 from the primary drug molecular-concentration contrast between normal muscle and tumor by the proper selection of the illumination wavelength.
