  • The Proteome of Primary Prostate Cancer

    • Background: Clinical management of the prostate needs improved prognostic tests and treatment strategies. Because proteins are the ultimate effectors of most cellular reactions, are targets for drug actions and constitute potential biomarkers; a quantitative systemic overview of the proteome changes occurring during prostate cancer (PCa) initiation and progression can result in clinically relevant discoveries.Objectives: To study cellular processes altered in PCa using system-wide quantitative analysis of changes in protein expression in clinical samples and to identify prognostic biomarkers for disease aggressiveness.Design, setting, and participants: Mass spectrometry was used for genome-scale quantitative proteomic profiling of 28 prostate tumors (Gleason score 6-9) and neighboring nonmalignant tissue in eight cases, obtained from formalin-fixed paraffin-embedded prostatectomy samples. Two independent cohorts of PCa patients (summing 752 cases) managed by expectancy were used for immunohistochemical evaluation of proneuropeptide-Y (pro-NPY) as a prognostic biomarker.Results and limitations: Over 9000 proteins were identified as expressed in the human prostate. Tumor tissue exhibited elevated expression of proteins involved in multiple anabolic processes including fatty acid and protein synthesis, ribosomal biogenesis and protein secretion but no overt evidence of increased proliferation was observed. Tumors also showed increased levels of mitochondrial proteins, which was associated with elevated oxidative phosphorylation capacity measured in situ. Molecular analysis indicated that some of the proteins overexpressed in tumors, such as carnitine palmitoyltransferase 2 (CPT2, fatty acid transporter), coatomer protein complex, subunit alpha (COPA, vesicle secretion), and mitogen-and stress-activated protein kinase 1 and 2 (MSK1/2, protein kinase) regulate the proliferation of PCa cells. Additionally, pro-NPY was found overexpressed in PCa (5-fold, p < 0.05), but largely absent in other solid tumor types. Pro-NPY expression, alone or in combination with the ERG status of the tumor, was associated with an increased risk of PCa specific mortality, especially in patients with Gleason score <= 7 tumors.Conclusions: This study represents the first system-wide quantitative analysis of proteome changes associated to localized prostate cancer and as such constitutes a valuable resource for understanding the complex metabolic changes occurring in this disease. We also demonstrated that pro-NPY, a protein that showed differential expression between high and low risk tumors in our proteomic analysis, is also a PCa specific prognostic biomarker associated with increased risk for disease specific death in patients carrying low risk tumors.Patient summary: The identification of proteins whose expression change in prostate cancer provides novel mechanistic information related to the disease etiology.   • The occurrence of noncoagulating milk and the association of bovine milk coagulation properties with genetic variants of the caseins in 3 Scandinavian dairy breeds

    • Substantial variation in milk coagulation properties has been observed among dairy cows. Consequently, raw milk from individual cows and breeds exhibits distinct coagulation capacities that potentially affect the technological properties and milk processing into cheese. This variation is largely influenced by protein composition, which is in turn affected by underlying genetic polymorphisms in the major milk proteins. In this study, we conducted a large screening on 3 major Scandinavian breeds to resolve the variation in milk coagulation traits and the frequency of milk with impaired coagulation properties (noncoagulation). In total, individual coagulation properties were measured on morning milk collected from 1,299 Danish Holstein (DH), Danish Jersey (DJ), and Swedish Red (SR) cows. The 3 breeds demonstrated notable interbreed differences in coagulation properties, with DJ cows exhibiting superior coagulation compared with the other 2 breeds. In addition, milk samples from 2% of DH and 16% of SR cows were classified as noncoagulating. Furthermore, the cows were genotyped for major genetic variants in the alpha(S1)- (CSN1S1), beta- (CSN2), and kappa-casein (CSN3) genes, revealing distinct differences in variant frequencies among breeds. Allele I of CSN2, which had not formerly been screened in such a high number of cows in these Scandinavian breeds, showed a frequency around 7% in DH and DJ, but was not detected in SR. Genetic polymorphisms were significantly associated with curd firming rate and rennet coagulation time. Thus, CSN1S1 C, CSN2 B, and CSN3 B positively affected milk coagulation, whereas CSN2 A(2), in particular, had a negative effect. In addition to the influence of individual casein genes, the effects of CSN1S1-CSN2-CSN3 composite genotypes were also examined, and revealed strong associations in all breeds, which more or less reflected the single gene results. Overall, milk coagulation is under the influence of additive genetic variation. Optimal milk for future cheese production can be ensured by monitoring the frequency of unfavorable variants and thus preventing an increase in the number of cows producing milk with impaired coagulation. Selective breeding for variants associated with superior milk coagulation can potentially increase raw milk quality and cheese yield in all 3 Scandinavian breeds.

  • Development and results of the Swedish road deflection tester

    • A project to construct a high-speed road deflection tester was initiated in the 1991. A mid-sized truck was used as a carrier for the first prototype. The results were promising and it was decided to build a full-size truck system. The new vehicle, based on a Scania R143 ML, was completed in 1997. The Road Deflection Tester (RDT) is equipped with two arrays of twenty noncontact laser sensors that collects transversal surface profiles at normal traffic speeds. One profile, placed between the wheel axles, constitutes an unloaded case. The other profile, just behind the rear axle of the vehicle, constitutes the loaded case. By subtracting the front cross profile from the corresponding rear one, the "deflection profile" is assessed. The deflection is assumed to vary with the stiffness of the road. In order to produce a large load on the rear wheels the engine was mounted in the back of the vehicle, slightly behind the rear axle. In testing mode the rear axle force is approximately 112 kN, and the front axle force is about 30 kN. An incremental wheel pulse transducer, two force transducers and two accelerometers, an optical speedometer and a gyroscope are also mounted on the RDT. The first test programme was carried out in 1998. Due to the careful choice of test sections, data from these sections still produce the best results. A smaller test programme was carried out in 2001, and a larger one in 2002 when the RDT was taken to England and France for demonstration. Promising results, both on an aggregated scale and for individual test sections, have been obtained. The RDT compares favourably with the Falling Weight Deflectometer. Short histories of road construction and road research give some historical and cultural background to the more recent developments. A more comprehensive history of rolling deflectographs presents all devices found in the literature from the start in the mid-fifties when the California Traveling Deflectograph and Lacroix Deflectograph were constructed, to the latest laser based High-Speed Deflectograph. Many references are given for further reading. The data acquisition hardware on the RDT system consist of sensors, signal converters, signal processing cards, an industrial computer for data communication, and an ordinary PC for operating the equipment and data storage. The software used to evaluate the data is written entirely in Matlab. Many levels of pre-processing make evaluation relatively fast, and the modularised design makes it easy to implement new evaluation algorithms in a clean and efficient way. A literature survey on the deformations of solids under static and moving load is presented in Appendix A. The static case started with Boussinesq in 1885, was much developed in the sixties, but since the eighties only a very limited amount of new results have been published. The moving load case, on the other hand, is still an field of active research and development.
