  • The Proteome of Primary Prostate Cancer

    • Background: Clinical management of the prostate needs improved prognostic tests and treatment strategies. Because proteins are the ultimate effectors of most cellular reactions, are targets for drug actions and constitute potential biomarkers; a quantitative systemic overview of the proteome changes occurring during prostate cancer (PCa) initiation and progression can result in clinically relevant discoveries.Objectives: To study cellular processes altered in PCa using system-wide quantitative analysis of changes in protein expression in clinical samples and to identify prognostic biomarkers for disease aggressiveness.Design, setting, and participants: Mass spectrometry was used for genome-scale quantitative proteomic profiling of 28 prostate tumors (Gleason score 6-9) and neighboring nonmalignant tissue in eight cases, obtained from formalin-fixed paraffin-embedded prostatectomy samples. Two independent cohorts of PCa patients (summing 752 cases) managed by expectancy were used for immunohistochemical evaluation of proneuropeptide-Y (pro-NPY) as a prognostic biomarker.Results and limitations: Over 9000 proteins were identified as expressed in the human prostate. Tumor tissue exhibited elevated expression of proteins involved in multiple anabolic processes including fatty acid and protein synthesis, ribosomal biogenesis and protein secretion but no overt evidence of increased proliferation was observed. Tumors also showed increased levels of mitochondrial proteins, which was associated with elevated oxidative phosphorylation capacity measured in situ. Molecular analysis indicated that some of the proteins overexpressed in tumors, such as carnitine palmitoyltransferase 2 (CPT2, fatty acid transporter), coatomer protein complex, subunit alpha (COPA, vesicle secretion), and mitogen-and stress-activated protein kinase 1 and 2 (MSK1/2, protein kinase) regulate the proliferation of PCa cells. Additionally, pro-NPY was found overexpressed in PCa (5-fold, p < 0.05), but largely absent in other solid tumor types. Pro-NPY expression, alone or in combination with the ERG status of the tumor, was associated with an increased risk of PCa specific mortality, especially in patients with Gleason score <= 7 tumors.Conclusions: This study represents the first system-wide quantitative analysis of proteome changes associated to localized prostate cancer and as such constitutes a valuable resource for understanding the complex metabolic changes occurring in this disease. We also demonstrated that pro-NPY, a protein that showed differential expression between high and low risk tumors in our proteomic analysis, is also a PCa specific prognostic biomarker associated with increased risk for disease specific death in patients carrying low risk tumors.Patient summary: The identification of proteins whose expression change in prostate cancer provides novel mechanistic information related to the disease etiology.     • Zn-alpha2-glycoprotein (ZAG) is a serum protein implicated in cancer cachexia and lipolysis. Our aim was to investigate serum levels of ZAG and polymorphisms in the ZAG gene in relation to serum lipids in man. Serum levels of ZAG correlated with serum levels of cholesterol (P = .00088) in healthy subjects and during weight loss (P = .059). The ZAG genotype was associated with total cholesterol (P = .014) and low-density lipoprotein cholesterol (P = .026) in healthy subjects, and the associations were replicated in an additional cohort (P = .0017 and P = .060, respectively). Our data indicate that ZAG plays a role in lipid metabolism.

  • Steps and catalytic reactions : CO oxidation with preadsorbed O on Rh(553)

    • Industrial catalysts are often comprised of nanoparticles supported on high-surface-area oxides, in order to maximise the catalytically active surface area and thereby utilise the active material better. These nanoparticles expose steps and corners that, due to low coordination to neighboring atoms, are more reactive and, as a consequence, are often assumed to have higher catalytic activity. We have investigated the reaction between CO and preadsorbed O on a stepped Rh(553) surface, and show that CO oxidation indeed occurs faster than on the flat Rh(111) surface at the same temperature. However, we do find that this is not a result of reactions at the step sites but rather at the terrace sites close to the steps, due to in-plane relaxation enabled by the step. This insight can provide ways to optimize the shape of the nanoparticles to further improve the activity of certain reactions.

The suppressed formation of ammonia observed as the feed becomes net-oxidizing is accompanied by a considerable increase in the oxidation state of platinum as well as the formation of surface nitrates and the loss of NH-containing surface species. In the presence of (excess) oxygen, the ammonia formation is proposed to be limited by weak interaction between nitrogen oxide and the oxidized platinum surface. This leads to a slow dissociation rate of nitrogen oxide and thus low abundance of the atomic nitrogen surface species that can react with the adsorbed hydrogen species. In this case the consumption of hydrogen through the competing water formation reaction and decomposition/oxidation of ammonia are of less importance for the net ammonia formation.

    • Monolithic Pt/gamma-Al2O3 catalysts were prepd. by depositing of the platinum phase, either homogeneously or heterogeneously (i.e., local high platinum concn.), in the washcoat. The influence of the platinum distribution on the ignition and extinction processes for oxidn. of CO, propene, and propane, resp., was investigated by both temp.-programmed and oxygen step-response flow-reactor expts. In addn., in situ XANES spectroscopy was used to follow changes in the chem. state of platinum during propane oxidn. For samples with heterogeneous platinum distribution, the results show an improved low-temp. activity for CO oxidn., whereas no clear improvement is obsd. for oxidn. of propene or propane. Comparison of the results for CO and hydrocarbon oxidn. shows that the improved activity cannot be explained by thermal effects. Moreover, calcns. indicate that the enhanced activity for CO oxidn. can be due to mass-transfer phenomena. This could not be confirmed by our expts., however. Instead, the improved activity for CO oxidn. can be due to varying platinum particle size between the samples. Furthermore, the oxidn. of propane is shown to be highly influenced by the oxygen concn., showing an activity max. for gas compositions close to the stoichiometric. High oxygen levels result most probably in a predominantly oxygen-covered surface, which inhibits the reaction and, as supported by the in situ XANES experiments, passivates the surface by forming platinum oxide.

  • Metal dimer sites in ZSM-5 zeolite for methane-to-methanol conversion from first-principles kinetic modelling: is the [Cu-O-Cu]2+ motif relevant for Ni, Co, Fe, Ag, and Au?

    • Direct methane-to-methanol conversion is a desired process whereby natural gas is transformed into an energy-rich liquid. It has been realised at ambient pressure and temperature in metal ion-exchanged zeolites, where especially copper-exchanged ZSM-5 has shown promising results. The nature of the active sites in these systems is, however, still under debate. The activity has been assigned to a [Cu-O-Cu]2+ motif. One remaining question is whether this motif is general and also active in other metal-exchanged zeolites. Herein, we use first-principles microkinetic modelling to analyse the methane-to-methanol reaction on the [Cu-O-Cu]2+ motif, for Cu and other metals. First, we identify the cluster model size needed to accurately describe the dimer motif. Starting from the [Cu-O-Cu]2+ site, the metal ions are then systematically substituted with Ni, Co, Fe, Ag and Au. The results show that activation of Ag and Au dimer sites with oxygen is endothermic and therefore unlikely, whereas for Cu, Ni, Co and Fe, the activation is possible under realistic conditions. According to the kinetic simulations, however, the dimer motif is a plausible candidate for the active site for Cu only. For Ni, Co and Fe, close-to-infinite reaction times or unreasonably high temperatures are required for sufficient methane conversion. As Ni-, Co- and Fe-exchanged ZSM-5 are known to convert methane to methanol, these results indicate that the Cu-based dimer motif is not an appropriate model system for these metals.

    • The study aimed to examine if dysmetabolic subjects (MetS+) have lower adiponectin gene expression and lower circulating adiponectin levels than non-dysmetabolic obese subjects (MetS-) at baseline, if adiponectin expression and adiponectin concentration rise more in the dysmetabolic group during weight loss, and if v-SNARE Vti1a (vesicle transport soluble NSF attachment protein receptor vps10p tail interacting 1a) expression increases during the weight loss, as a mechanism for increased adiponectin secretion. Twenty-one obese MetS+ and 19 obese MetS- subjects underwent a very low-energy diet for 16 weeks followed by 2 weeks of refeeding. Abdominal subcutaneous adipose tissue biopsies and blood samples were taken before, during, and after dieting for DNA microarray, reverse transcriptase-polymerase chain reaction, and biochemical analyses. Serum adiponectin was also assessed in a sex- and age-matched healthy, nonobese reference group. Weight decreased by 26.3+/-9.8 kg in the MetS+ group and 28.2+/-8.4 kg in the MetS- group with concomitant reductions in insulin, hemoglobin A1c, and triglycerides that were more pronounced in the MetS+ group. Initially, the MetS+ subjects had lower serum adiponectin, but the differences disappeared at week 8, with a continuous increase in serum adiponectin throughout the study in both groups to a level that was higher than in the reference group. The expression of adiponectin and v-SNARE Vti1a did not differ between the groups or over time. In conclusion, obese subjects with the metabolic syndrome had lower circulating adiponectin than subjects without the syndrome. Weight loss increased serum levels of adiponectin without a parallel increase in adiponectin gene expression. The mechanisms involved in the regulation of adiponectin levels merits further investigation.
