    • Type 2 diabetes (T2D) is caused by insufficient insulin secretion from pancreatic β-cells. To identify candidates contributing to T2D pathophysiology, we studied human pancreatic islets from ~300 individuals. We found 395 differentially expressed genes (DEGs) in islets from individuals with T2D, including, to our knowledge, novel (OPRD1, PAX5, TET1) and previously identified (CHL1, GLRA1, IAPP) candidates. A third of the identified islet expression changes may predispose to diabetes, as they associated with HbA1c in individuals not previously diagnosed with T2D. Most DEGs were expressed in human β-cells based on single-cell RNA-sequencing data. Additionally, DEGs displayed alterations in open chromatin and associated with T2D-SNPs. Mouse knock-out strains demonstrated that T2D-associated candidates regulate glucose homeostasis and body composition in vivo. Functional validation showed that mimicking T2D-associated changes for OPRD1, PAX5, and SLC2A2 impaired insulin secretion. Impairments in Pax5-overexpressing β-cells were due to severe mitochondrial dysfunction. Finally, we discovered PAX5 as a potential transcriptional regulator of many T2D-associated DEGs in human islets. Overall, we identified molecular alterations in human pancreatic islets contributing to β-cell dysfunction in T2D pathophysiology.

  • Interacting with the arrowhead local cloud : On-boarding procedure

    • Industrial automation systems are advancing rapidly and a wide range of standards, communication protocols and platforms supporting the integration of devices are introduced. It is therefore necessary to design and build appropriate tools and frameworks that allow the integration of devices with multiple systems and services. In this work we present the Arrow-head Framework, used to enable collaborative IoT automation and introduce two support core systems, SystemRegistry and DeviceRegistry, which are needed to create a chain of trust from a hardware device to a software system and its associated services. Furthermore, we propose an on-boarding procedure of a new device interacting with the Arrowhead local cloud. This ensures that only valid and authorized devices can host software systems within an Arrowhead local cloud.

    • This paper discusses the design and implementation of a cooperative road infrastructure systems, which uses an intelligent road surface. Using an overtaking assist feature as an example it is shown how such a feature can be designed and implemented on a road infrastructure and integrated with drivers and passengers using IMS. The feasibility of this feature is assessed from a functional and communication perspective. Moreover, first results from real-life tests on the Swedish highway E4 are presented which motivate the next research and development steps.

  • Road surface networks technology enablers for enhanced ITS

    • The increased need for mobility has led to transportation problems like congestion, accidents and pollution. In order to provide safe and efficient transport systems great efforts are currently being put into developing Intelligent Transport Systems (ITS) and cooperative systems. In this paper we extend proposed solutions with autonomous on-road sensors and actuators forming a wireless Road Surface Network (RSN). We present the RSN architecture and design methodology and demonstrate its applicability to queue-end detection. For the use case we discuss the requirements and technological solutions to sensor technology, data processing and communication. In particular the MAC protocol is detailed and its performance assessed through theoretical verification. The RSN architecture is shown to offer a scalable solution, where increased node density offers more precise sensing as well as increased redundancy for safety critical applications. The use-case demonstrates that RSN solutions may be deployed as standalone systems potentially integrated into current and future ITS. RSN may provide both easily deployable and cost effective alternatives to traditional ITS (with a direct impact independent of penetration rate of other ITS infrastructures - i.e., smart vehicles, safe spots etc.) as well as provide fine grain sensory information directly from the road surface to back-end and cooperative systems, thus enabling a wide range of ITS applications beyond current state of the art.

    • The Arrowhead project aims to address the technical and applicative issues associated with cooperative automation based on Service Oriented Architectures. The problems of developing such kind of systems are mainly due to the lack of adequate development and service documentation methodologies, which would ease the burden of reusing services on different applications. The Arrowhead project proposes a technical framework to efficiently support the development of such systems, which includes several tools for documentation of services and to support the development of SOA-based installations. The work presented in this paper describes the approach which has been developed for the first generation pilots to support the documentation of their structural services. Each service, system and system- of-systems within the Arrowhead Framework must be documented and described in such way that it can be implemented, tested and deployed in an interoperable way. This paper presents the first steps of realizing the Arrowheadvision for interoperable services, systems and systems-of-systems.

    • Some of the common soil microorganisms are actinomycetes, Gram-positive bacteria with high GC content. Because of their mycelial habit they were initially believed to be fungi, which reected in their name (mucus (lat.) means fungus). In 1939, one year before rediscovery of penicillin by Florey and Chain, soil microbiologist Waksman has set his lab on a quest for new antimicrobial drugs. From the previous studies he knew that actinomycetes can inhibit the growth of other soil bacteria through secretion of bioactive compounds, which he named “antibiotics” (anti (lat.) against, bio (lat.) life). Systematic search for antibiotics produced by actinomycetes resulted in the discovery of actinomycin (1940), clavacin, and streptothricin (1942), all of them sadly turned out to be toxic in animal tests. In 1943 Waksman’s student Schatz isolated streptomycin-producing strain of Streptomyces griseus.1 Streptomycin was not particularly toxic to animals and humans, but remarkably was the rst compound active against tuberculosis bacteria. Many pharmaceutical companies and research laboratories started to collect soil samples from all over the world in search of antibiotics-producing organisms. Most of the discoveries were made in the rst ten years of the “hunt,” the larger part involved Streptomyces species. Streptomyces is a genus in the genera of actinomycetes, many of these bacteria produce volatile compounds that give the earth its characteristic odor. Streptomyces proved to be an excellent source of secondary metabolites, including antibiotics, anticancerous agents, antihelmintic drugs, and other useful compounds (Table 24.1). At present more than half of antibiotics in clinical use are produced in Streptomyces species.

  • Plasma dynamics at the Schwinger limit and beyond

    • Strong field physics close to or above the Schwinger limit are typically studied with vacuum as initial condition or by considering test particle dynamics. However, with a plasma present initially, quantum relativistic mechanisms such as Schwinger pair creation are complemented by classical plasma nonlinearities. In this work we use the Dirac-Heisenberg-Wigner formalism to study the interplay between classical and quantum mechanical mechanisms in the regime of ultrastrong electric fields. In particular, the effects of initial density and temperature on the plasma oscillation dynamics are determined. Finally, comparisons with competing mechanisms such as radiation reaction and Breit-Wheeler pair production are made.

    • Current challenges in production automation requires the involvement of new technologies like Internet of Things (IoT), Systems of Systems and local automation clouds. The objective of this paper is to address one of the challenges involved in establishing and managing a cloud based automation system. Three key capabilities have been identified as required to create the expected benefits of local automation clouds; 1) capturing of plant design 2) capturing and distributing configuration and deployment information 3) coordinating information exchange.This paper addresses the capturing and distribution of configuration and deployment information. For this purpose a system service is proposed, the ConfigurationStore, following the principles of the Arrowhead Framework. The service is accompanied by a deployment methodology and a bootstrapping procedure. These are discussed for several types of automation technology, e.g. controllers, sensors, actuators. A qualitative evaluation of the proposed approach is made for four use cases; Building automation, Manufacturing automation, Process automation and IoT devices. Concluding the usability for large-scale deployment and configuration of Industrial Internet of Things.

    • Major societal challenges such as environmental sustainability, availability of energy and raw materials, and globalisation are creating new requirements for many actors in society. These new requirements relate to efficiency, flexibility, sustainability, and competitiveness. While these aspects have all been around for some time, and many systems have been locally optimised with regards to one or a few of these requirements, recent advances in communication and processing capabilities promise increased possibilities for connecting different parts of society, allowing optimal operation on a much larger scale.While industrial production systems have been controlled electronically for decades, the digitisation of market channels and consumer systems, together with the possibility to interconnect different production facilities, now allow for automated interaction along the whole supply chain from raw materials to end users.Simultaneously, increased demand for efficiency forces increased specialisation among actors, which with increased possibilities of interconnectivity, creates large enterprises of cooperating, specialised stakeholders.One of the major remaining obstacles for a widespread adaptation of more intelligent, more connected systems, able to deliver these envisioned results, is a coherent approach to the engineering and management of Systems-of-Systems involving very large numbers of devices and operating across several automation domains.For traditional automation systems there are established engineering procedures and numerous standards for engineering data, although most are focused on the static processes that have traditionally been the norm. For full integration with a digital society many of the existing automation systems will need significant modifications and as many automation systems are ageing and in need of replacement, a suitable solution to this may be a large scale migration to new automation solutions.The work presented in this thesis includes some new approaches and methodologies to utilise the existing engineering procedures and standards, while introducing some of the flexibility proposed by the emerging technologies. The major technical solutions presented consist of a structure way to organise connected systems and how they are related, regardless of engineering standards used to design their interactions, and an approach to allow configuration of heterogeneous systems through service interactions.Further contributions include an approach for migrating certain categories of existing industrial control systems to a service oriented architecture, as a basic outline for adaption of the next generation of automation systems in industry.Certain remaining challenges have been identified, which have to be addressed for a successful launch of widespread interconnected automation systems based on Internet of Things and associated technologies.
