    • Experiments with a climate model (NorESM1) were performed to isolate the effects of aerosol particles and greenhouse gases on surface temperature and precipitation in simulations of future climate. The simulations show that by 2025-2049, a reduction of aerosol emissions from fossil fuels following a maximum technically feasible reduction (MFR) scenario could lead to a global and Arctic warming of 0.26 K and 0.84 K, respectively; as compared with a simulation with fixed aerosol emissions at the level of 2005. If fossil fuel emissions of aerosols follow a current legislation emissions (CLE) scenario, the NorESM1 model simulations yield a non-significant change in global and Arctic average surface temperature as compared with aerosol emissions fixed at year 2005. The corresponding greenhouse gas effect following the RCP4.5 emission scenario leads to a global and Arctic warming of 0.35 K and 0.94 K, respectively.The model yields a marked annual average northward shift in the inter-tropical convergence zone with decreasing aerosol emissions and subsequent warming of the northern hemisphere. The shift is most pronounced in the MFR scenario but also visible in the CLE scenario. The modeled temperature response to a change in greenhouse gas concentrations is relatively symmetric between the hemispheres and there is no marked shift in the annual average position of the inter-tropical convergence zone. The strong reduction in aerosol emissions in MFR also leads to a net southward cross-hemispheric energy transport anomaly both in the atmosphere and ocean, and enhanced monsoon circulation in Southeast and East Asia causing an increase in precipitation over a large part of this region.

  • Biomarkers in Natural Fish Populations Indicate Adverse Biological Effects of Offshore Oil Production

    • Background: Despite the growing awareness of the necessity of a sustainable development, the global economy continues to depend largely on the consumption of non-renewable energy resources. One such energy resource is fossil oil extracted from the seabed at offshore oil platforms. This type of oil production causes continuous environmental pollution from drilling waste, discharge of large amounts of produced water, and accidental spills. Methods and principal findings: Samples from natural populations of haddock (Melanogrammus aeglefinus) and Atlantic cod (Gadus morhua) in two North Sea areas with extensive oil production were investigated. Exposure to and uptake of polycyclic aromatic hydrocarbons (PAHs) were demonstrated, and biomarker analyses revealed adverse biological effects, including induction of biotransformation enzymes, oxidative stress, altered fatty acid composition, and genotoxicity. Genotoxicity was reflected by a hepatic DNA adduct pattern typical for exposure to a mixture of PAHs. Control material was collected from a North Sea area without oil production and from remote Icelandic waters. The difference between the two control areas indicates significant background pollution in the North Sea. Conclusion: It is most remarkable to obtain biomarker responses in natural fish populations in the open sea that are similar to the biomarker responses in fish from highly polluted areas close to a point source. Risk assessment of various threats to the marine fish populations in the North Sea, such as overfishing, global warming, and eutrophication, should also take into account the ecologically relevant impact of offshore oil production.

  • A Comprehensive Evaluation of the Relationship Between Different IgG and IgA Anti-Modified Protein Autoantibodies in Rheumatoid Arthritis

    • Seropositive rheumatoid arthritis (RA) is characterized by the presence of rheumatoid factor (RF) and anti-citrullinated protein autoantibodies (ACPA) with different fine-specificities. Yet, other serum anti-modified protein autoantibodies (AMPA), e.g. anti-carbamylated (Carb), -acetylated (KAc), and malondialdehyde acetaldehyde (MAA) modified protein antibodies, have been described. In this comprehensive study, we analyze 30 different IgG and IgA AMPA reactivities to Cit, Carb, KAc, and MAA antigens detected by ELISA and autoantigen arrays in N=1985 newly diagnosed RA patients. Association with patient characteristics such as smoking and disease activity were explored. Carb and KAc reactivities by different assays were primarily seen in patients also positive for anti-citrulline reactivity. Modified vimentin (mod-Vim) peptides were used for direct comparison of different AMPA reactivities, revealing that IgA AMPA recognizing mod-Vim was mainly detected in subsets of patients with high IgG anti-Cit-Vim levels and a history of smoking. IgG reactivity to acetylation was mainly detected in a subset of patients with Cit and Carb reactivity. Anti-acetylated histone reactivity was RA-specific and associated with high anti-CCP2 IgG levels, multiple ACPA fine-specificities, and smoking status. This reactivity was also found to be present in CCP2+ RA-risk individuals without arthritis. Our data further demonstrate that IgG autoreactivity to MAA was increased in RA compared to controls with highest levels in CCP2+ RA, but was not RA-specific, and showed low correlation with other AMPA. Anti-MAA was instead associated with disease activity and was not significantly increased in CCP2+ individuals at risk of RA. Notably, RA patients could be subdivided into four different subsets based on their AMPA IgG and IgA reactivity profiles. Our serology results were complemented by screening of monoclonal antibodies derived from single B cells from RA patients for the same antigens as the RA cohort. Certain CCP2+ clones had Carb or Carb+KAc+ multireactivity, while such reactivities were not found in CCP2- clones. We conclude that autoantibodies exhibiting different patterns of ACPA fine-specificities as well as Carb and KAc reactivity are present in RA and may be derived from multireactive B-cell clones. Carb and KAc could be considered reactivities within the "Cit-umbrella" similar to ACPA fine-specificities, while MAA reactivity is distinctly different.

    • I denna avhandling studeras en ny teknik för att hantera växthuseffekten. Den nya tekniken heter koldioxidavskiljning och lagring (CCS) och granskades av FN:s klimatpanel 2005 och tillskrevs då möjligheterna att stå för 15-55% av alla CO2-reducering fram till 2100 och detta till en 30% lägre kostnad än vad som annars vore möjligt. EU är en framträdande pådrivare av CCS och för fram att växthuseffekten inte kan hanteras utan att CCS implementeras skyndsamt. CCS beskrivs i dessa sammanhang som en hållbar teknik. CCS är emellertid förbunden med långtidslagring, en betydande teknisk komplexitet och tillämpas främst på kolkraftverk. Storskaliga satsningar på CCS kan medföra att kolanvändningen ökar.Syftet med avhandlingen är att analysera de vetenskapliga och politiska ansträngningarna att visa att CCS är en eftersträvansvärd teknik för att hantera växthuseffekten. Utifrån perspektivet ekologisk modernisering och genom granskning av studier av vetenskapliga rapporter, artiklar i massmedia, politiska dokument och intervjuer genomförs studien. Scenerier och prognoser har en central funktion för att kunna påvisa att CCS är en eftersträvansvärd teknik. I flera av dessa scenarier framställs en närmast linjär teknikutveckling och flera betydelsefulla problem och hinder bortses från. CCS framstår som en teknik med stor teknisk och ekonomisk potential och i massmedia beskrivs CCS ofta som oumbärlig.En mer nyanserad bild framträder vid intervjuer med CCS-experter då fler osäkerheter och hinder lyfts fram. Förståelsen för varför denna teknik för stöd av många starka aktörer blir även tydligare. Den dominerande beskrivningen av CCS egenskaper och inverkan på energisystemen ligger i linje med det som är utmärkande för den ekologiska modernisering och således även för det dominerande sättet att bedriva energi- och klimatpolitik idag.

    • STUDY DESIGN: In vivo porcine study of intervertebral kinematics. OBJECTIVES: This study investigated the effect of transversus abdominis and diaphragm activity, and increased intra-abdominal pressure on intervertebral kinematics in porcine lumbar spines. BACKGROUND: Studies of trunk muscle recruitment in humans suggest that diaphragm and transversus abdominis activity, and the associated intra-abdominal pressure contribute to the control of intervertebral motion. However, this has not been tested in vivo. METHODS: Relative intervertebral motion of the L3 and L4 vertebrae and the stiffness at L4 were measured in response to displacements of the L4 vertebra imposed via a device fixed to the L4 vertebral body. In separate trials, diaphragm and transversus abdominis activity was evoked by stimulation of the phrenic nerves and via electrodes threaded through the abdominal wall. RESULTS: When intra-abdominal pressure was increased by diaphragm or transversus abdominis stimulation, the relative intervertebral displacement of the L3 and L4 vertebrae was reduced and the stiffness of L4 was increased for caudal displacements. There was no change in either parameter for rostral displacements. In separate trials, the diaphragm crurae and the fascial attachments of transversus abdominis were cut, but intra-abdominal pressure was increased. In these trials, the reduction in intervertebral motion was similar to trials with intact attachments for caudal motion, but was increased for rostral trials. CONCLUSIONS: The results of these studies indicate that elevated intra-abdominal pressure, and contraction of diaphragm and transversus abdominis provide a mechanical contribution to the control of spinal intervertebral stiffness. Furthermore, the effect is modified by the muscular attachments to the spine.

  • Potential links between Baltic Sea submarine terraces and groundwater seeping

    • Submarine groundwater discharge (SGD) influences ocean chemistry, circulation, and the spreading of nutrients and pollutants; it also shapes sea floor morphology. In the Baltic Sea, SGD was linked to the development of terraces and semicircular depressions mapped in an area of the southern Stockholm archipelago, Sweden, in the 1990s. We mapped additional parts of the Stockholm archipelago, areas in Blekinge, southern Sweden, and southern Finland using high-resolution multibeam sonars and sub-bottom profilers to investigate if the sea floor morphological features discovered in the 1990s are widespread and to further address the hypothesis linking their formation to SGD. Sediment coring and sea floor photography conducted with a remotely operated vehicle (ROV) and divers add additional information to the geophysical mapping results. We find that terraces, with general bathymetric expressions of about 1 m and lateral extents of sometimes > 100 m, are widespread in the surveyed areas of the Baltic Sea and are consistently formed in glacial clay. Semicircular depressions, however, are only found in a limited part of a surveyed area east of the island of Asko, southern Stockholm archipelago. While submarine terraces can be produced by several processes, we interpret our results to be in support of the basic hypothesis of terrace formation initially proposed in the 1990s; i.e. groundwater flows through siltier, more permeable layers in glacial clay to discharge at the sea floor, leading to the formation of a sharp terrace when the clay layers above seepage zones are undermined enough to collapse. By linking the terraces to a specific geologic setting, our study further refines the formation hypothesis and thereby forms the foundation for a future assessment of SGD in the Baltic Sea that may use marine geological mapping as a starting point. We propose that SGD through the submarine sea floor terraces is plausible and could be intermittent and linked to periods of higher groundwater levels, implying that to quantify the contribution of freshwater to the Baltic Sea through this potential mechanism, more complex hydrogeological studies are required.

    • Cultivated oat (Avena sativa L.) is an allohexaploid (AACCDD, 2n = 6x = 42) thought to have been domesticated more than 3,000 years ago while growing as a weed in wheat, emmer and barley fields in Anatolia1,2. Oat has a low carbon footprint, substantial health benefits and the potential to replace animal-based food products. However, the lack of a fully annotated reference genome has hampered efforts to deconvolute its complex evolutionary history and functional gene dynamics. Here we present a high-quality reference genome of A. sativa and close relatives of its diploid (Avena longiglumis, AA, 2n = 14) and tetraploid (Avena insularis, CCDD, 2n = 4x = 28) progenitors. We reveal the mosaic structure of the oat genome, trace large-scale genomic reorganizations in the polyploidization history of oat and illustrate a breeding barrier associated with the genome architecture of oat. We showcase detailed analyses of gene families implicated in human health and nutrition, which adds to the evidence supporting oat safety in gluten-free diets, and we perform mapping-by-sequencing of an agronomic trait related to water-use efficiency. This resource for the Avena genus will help to leverage knowledge from other cereal genomes, improve understanding of basic oat biology and accelerate genomics-assisted breeding and reanalysis of quantitative trait studies.
