  • Variations in monsoonal rainfall over the last 21 kyr inferred from sedimentary organic matter in Tung-Yuan Pond, southern Taiwan

    • Changes in paleorainfall intensity linked to the strength of the East Asian (EA) summer monsoon since 21 cal kyr BP are inferred from the organic matter contents of a 15-m sediment core from Tung-Yuan Pond in southern Taiwan. High total organic carbon/total nitrogen (TOC/TN) values in association with increased TOC content suggest that more soil-derived material containing terrestrial organic matter (OM) was delivered to the lake during periods of increased runoff associated with extensive precipitation that resulted from intensified summer monsoons, whereas low values indicate OM possessing a dominant algal origin during weakened summer monsoons. Rainfall intensity in terms of the proportion of terrestrial OM was high in four periods: the last deglaciation (similar to 17.2 to similar to 12.2 ka), the early Holocene (similar to 10.6 to similar to 8.6 ka), the middle Holocene Thermal Optimum (similar to 7.7 to similar to 5 ka) and the late Holocene (similar to 4.2 to similar to 2 ka), whereas it was low in the intervening time periods. High TOC/TN values coincide with peak values of summer insolation, and thus the strongest EA summer monsoon during the early and middle Holocene: small drops in these ratios correspond to increasing and decreasing solar radiation in the deglacial period and the late Holocene, respectively. The four intervals with low TOC/TN ratios, as well as episodic drops of the ratios during the deglaciation and the early and late Holocene are concordant with the late last glacial (similar to 21 to similar to 17.2 ka), the Oldest (similar to 14.8 ka), the Older (similar to 13.3 ka) and the Younger Dryas (similar to 13 to similar to 11.5 ka), the 8.2 cold event (similar to 8.6-7.7 ka) and a late Holocene cold event (similar to 5-4.2 ka), and suggest a weakened EA summer monsoon during these times. Moreover, high frequency hydrological variability occurred during the early Holocene, heavy rainfall persisted during the middle Holocene, and precipitation intensity generally diminished after similar to 5 ka. The Tung-Yuan Pond sediment record indicates that the TOC/TN ratio can be used as a paleorainfall intensity proxy to trace variations in the EA summer monsoon strength in other small lakes.     • The Wilms' tumor suppressor gene (WT1) has been identified as an oncogene in many malignant diseases such as leukaemia, breast cancer, mesothelioma and lung cancer. However, the role of WT1 in non-small-cell lung cancer (NSCLC) carcinogenesis remains unclear. In this study, we compared WT1 mRNA levels in NSCLC tissues with paired corresponding adjacent tissues and identified significantly higher expression in NSCLC specimens. Cell proliferation of three NSCLC cell lines positively correlated with WT1 expression; moreover, these associations were identified in both cell lines and a xenograft mouse model. Furthermore, we demonstrated that up-regulation of Cyclin D1 and the phosphorylated retinoblastoma protein (p-pRb) was mechanistically related to WT1 accelerating cells to S-phase. In conclusion, our findings demonstrated that WT1 is an oncogene and promotes NSCLC cell proliferation by up-regulating Cyclin D1 and p-pRb expression.

  • Photothermally tunable silicon-microring-based optical add-drop filter through integrated light absorber

    • An optically pumped thermo-optic (TO) silicon ring add-drop filter with fast thermal response is experimentally demonstrated. We propose that metal-insulator-metal (MIM) light absorber can be integrated into silicon TO devices, acting as a localized heat source which can be activated remotely by a pump beam. The MIM absorber design introduces less thermal capacity to the device, compared to conventional electrically-driven approaches. Experimentally, the absorber-integrated add-drop filter shows an optical response time of 13.7 mu s following the 10%-90% rule (equivalent to a exponential time constant of 5 mu s) and a wavelength shift over pump power of 60 pm/mW. The photothermally tunable add-drop filter may provide new perspectives for all-optical routing and switching in integrated Si photonic circuits.

    • To identify susceptibility genes for amyotrophic lateral sclerosis (ALS), we conducted a genome-wide association study (GWAS) in 506 individuals with sporadic ALS and 1,859 controls of Han Chinese ancestry. Ninety top SNPs suggested by the current GWAS and 6 SNPs identified by previous GWAS were analyzed in an independent cohort of 706 individuals with ALS and 1,777 controls of Han Chinese ancestry. We discovered two new susceptibility loci for ALS at 1q32 (CAMK1G, rs6703183, P-combined = 2.92 x 10(-8), odds ratio (OR) = 1.31) and 22p11 (CABIN1 and SUSD2, rs8141797, P-combined = 2.35 x 10(-9), OR = 1.52). These two loci explain 12.48% of the overall variance in disease risk in the Han Chinese population. We found no association evidence for the previously reported loci in the Han Chinese population, suggesting genetic heterogeneity of disease susceptibility for ALS between ancestry groups. Our study identifies two new susceptibility loci and suggests new pathogenic mechanisms of ALS.

    • Efficient narrowband light absorption by a metal-insulator-metal (MIM) structure can lead to high-speed light-to-heat conversion at a micro-or nanoscale. Such a MIM structure can serve as a heater for achieving all-optical light control based on the thermo-optical (TO) effect. Here we experimentally fabricated and characterized a novel all-optical switch based on a silicon microdisk integrated with a MIM light absorber. Direct integration of the absorber on top of the microdisk reduces the thermal capacity of the whole device, leading to high-speed TO switching of the microdisk resonance. The measurement result exhibits a rise time of 2.0 mu s and a fall time of 2.6 mu s with switching power as low as 0.5 mW; the product of switching power and response time is only about 1.3 mW.mu s. Since no auxiliary elements are required for the heater, the switch is structurally compact, and its fabrication is rather easy. The device potentially can be deployed for new kinds of all-optical applications.

    • For economic reasons and to save time there is a need to shorten the drying operation associated with the production of chitosan materials. Hence it is of interest to extrude chitosan at as high a solids content as possible. This is, to our knowledge, the first systematic study of the extrusion of chitosan at high solids content (60 wt%). An orthogonal experimental design was used to evaluate the effect of processing conditions and material factors on the extrudability of chitosan. This, together with the examination of the evenness and surface finish of the extrudate, made it possible to determine the best conditions for obtaining a readily extrudable high quality material. It was observed that a 1/1 ratio of chitosans with molar masses of 12 and 133 kDa, a process liquid containing 30 wt% acetic acid and 70 wt% water, and extrusion at 50 rpm and 50°C were the optimal material and processing conditions. Materials processed under these conditions were evaluated mechanically at different times after extrusion (stored at 50% RH) in order to see when the properties stabilized. Most mass loss occurred within the first three days after extrusion and this governed the mechanical properties (stiffness and extensibility), which also exhibited the largest changes within these three days (an increase in modulus from 18 to 830 MPa and a decrease in elongation at break from 17 to 3%).

  • Wheat gluten/chitosan blends : A new biobased material

    • Wheat gluten and chitosan are renewable materials that suffer from some poor properties that limit their use as a potential replacement of petroleum-based polymers. However, polymer blends based on wheat gluten and chitosan surprisingly reduced these shortcomings. Films were cast from acidic aqueous or water/ethanol solutions of wheat gluten and chitosan. Wheat gluten was the discontinuous phase in the 30-70 wt.% wheat gluten interval investigated. The most homogeneous films were obtained when reducing agents were used (alone or together with urea or glycerol). They consisted mainly of 1-2 mu m wheat gluten particles uniformly distributed in the continuous chitosan phase. Slightly smaller particles were also observed in the water/ethanol solvent system, but together with significantly larger particles (as large as 200 mu m). Both small and large particles were observed, albeit in different sizes and contents, when surfactants (both with and without a reducing agent) or urea (without a reducing agent) were used. The particles were often elongated, and preferably along the film, the most extreme case being observed when the glyoxal crosslinker was used together with sodium sulfite (reducing agent), showing particles with an average thickness of 0.6 mu m and an aspect ratio of 4.2. This film showed the highest transparency of all the blend films studied. For one of the most promising systems (with sodium sulfite), having good film homogeneity and small particles, the mechanical and moisture solubility/diffusivity properties were studied as a function of chitosan content. The extensibility, toughness and moisture solubility increased with increasing chitosan content, and the moisture diffusivity was highest for the pristine chitosan material. It is noteworthy that the addition of 30 wt.% wheat gluten to chitosan reduced the moisture uptake, while the extensibility/toughness remained unchanged.
