  • miRNA-378 Is Downregulated by XBP1 and Inhibits Growth and Migration of Luminal Breast Cancer Cells

    • X-box binding protein 1 (XBP1) is a transcription factor that plays a crucial role in the unfolded protein response (UPR), a cellular stress response pathway involved in maintaining protein homeostasis in the endoplasmic reticulum (EnR). While the role of XBP1 in UPR is well-characterised, emerging evidence suggests its involvement in endocrine resistance in breast cancer. The transcriptional activity of spliced XBP1 (XBP1s) is a major component of its biological effects, but the targets of XBP1s in estrogen receptor (ER)-positive breast cancer are not well understood. Here, we show that the expression of miR-378 and PPARGC1B (host gene of miR-378) is downregulated during UPR. Using chemical and genetic methods, we show that XBP1s is necessary and sufficient for the downregulation of miR-378 and PPARGC1B. Our results show that overexpression of miR-378 significantly suppressed cell growth, colony formation, and migration of ER-positive breast cancer cells. Further, we found that expression of miR-378 sensitised the cells to UPR-induced cell death and anti-estrogens. The expression of miR-378 and PPARGC1B was downregulated in breast cancer, and higher expression of miR-378 is associated with better outcomes in ER-positive breast cancer. We found that miR-378 upregulates the expression of several genes that regulate type I interferon signalling. Analysis of separate cohorts of breast cancer patients showed that a gene signature derived from miR-378 upregulated genes showed a strong association with improved overall and recurrence-free survival in breast cancer. Our results suggest a growth-suppressive role for miR-378 in ER-positive breast cancer where downregulation of miR-378 by XBP1 contributes to endocrine resistance in ER-positive breast cancer.

  • UPR-Induced miR-616 Inhibits Human Breast Cancer Cell Growth and Migration by Targeting c-MYC

    • C/EBP homologous protein (CHOP), also known as growth arrest and DNA damage-inducible protein 153 (GADD153), belongs to the CCAAT/enhancer-binding protein (C/EBP) family. CHOP expression is induced by unfolded protein response (UPR), and sustained CHOP activation acts as a pivotal trigger for ER stress-induced apoptosis. MicroRNA-616 is located within an intron of the CHOP gene. However, the regulation of miR-616 expression during UPR and its function in breast cancer is not clearly understood. Here we show that the expression of miR-616 and CHOP (host gene of miR-616) is downregulated in human breast cancer. Both miR-5p/-3p arms of miR-616 are expressed with levels of the 5p arm higher than the 3p arm. During conditions of ER stress, the expression of miR-616-5p and miR-616-3p arms was concordantly increased primarily through the PERK pathway. Our results show that ectopic expression of miR-616 significantly suppressed cell proliferation and colony formation, whereas knockout of miR-616 increased it. We found that miR-616 represses c-MYC expression via binding sites located in its protein coding region. Furthermore, we show that miR-616 exerted growth inhibitory effects on cells by suppressing c-MYC expression. Our results establish a new role for the CHOP locus by providing evidence that miR-616 can inhibit cell proliferation by targeting c-MYC. In summary, our results suggest a dual function for the CHOP locus, where CHOP protein and miR-616 can cooperate to inhibit cancer progression.

  • Metallic one-dimensional heterostructure for gas molecule sensing

    • We have investigated a new metallic core-shell nanowire (NW) geometry of that could be obtained experimentally, that is silicon (Si) and germanium (Ge) NWs with cores constituted by group-10 elements palladium (Pd) and platinum (Pt). These NWs are optimized with two different diameters of 1.5 angstrom and 2.5 angstrom. The nanowires having diameter of 1.5 angstrom show semi-metallic nature with GGA-PBE calculation and metallic nature while spin orbit interaction (SOC) is included. The quantum conductance of the NWs increases with the diameter of the nanowire. We have investigated current-voltage (IV) characteristics for the considered NWs. It has been found that current values in accordance with applied voltage show strong dependence on the diameter of the NWs. The optical study of the NWs shows that absorption co-efficient peak moves to lower energies; due to quantum confinement effect. Furthermore, we have extensively studied optical response of Pd and Pt based core-shell NWs in O-2 and CO2 environment. Our study on Si and Ge based metallic core/shell NW show a comprehensive picture as possible electron connector in future nano-electronic devices as well as nano gas detector for detecting O-2 gas.

    • In the last few decades, the miniaturization of devices has been taking place and therefore the quest for new nanowires has become more significant. In the present study, we have investigated the geometry of new ultrathin nanowires (NWs) of PdP2 and PdAs2 that could be obtained experimentally. We have optimized the pentagonal structures of both the NWs and studied their dynamical stability using the phonon dispersion curve. The electronic band structure shows semiconducting behaviour of PdP2 NWs with a band gap of 380 meV and PdAs2 NWs with a band gap of 294 meV, with higher charge carrier mobility than that of their 2D counterparts. The NWs show a band gap of 840 meV and 740 meV for PdP2 and PdAs2, respectively, through hybrid potential calculations. The PdX2 structure shows a transition from semiconducting to semi-metallic behaviour at a compressive strain of 8% within a sustainable pressure of 0.2–0.3 GPa. A negative differential conductance (NDC) effect is observed in the current–voltage graph for both the NWs. The semi-metallic behaviour with an asymmetric density of states near the Fermi energy boosts the Seebeck co-efficient value and therefore the ZTe value is enhanced for both the nanowires. The strained PdP2 and PdAs2 NWs show ZTe values of 4.75 and 5.49, respectively. Our study stimulates the feasibility of both nanowires and thermoelectric applications for the conversion of waste heat into electricity.

    • Accurate prediction of reference evapotranspiration (ETo) is crucial for many water-related fields, including crop modelling, hydrologic simulations, irrigation scheduling and sustainable water management. This study compares the performance of different soft computing models such as artificial neural network (ANN), wavelet-coupled ANN (WANN), adaptive neuro-fuzzy inference systems (ANFIS) and multiple nonlinear regression (MNLR) for predicting ETo. The Gamma test technique was adopted to select the suitable input combination of meteorological variables. The performance of the models was quantitatively and qualitatively evaluated using several statistical criteria. The study showed that the ANN-10 model performed superior to the ANFIS-06, WANN-11 and MNLR models. The proposed ANN-10 model was more appropriate and efficient than the ANFIS-06, WANN-11 and MNLR models for predicting daily ETo. Solar radiation was found to be the most sensitive input variable. In contrast, actual vapour pressure was the least sensitive parameter based on sensitivity analysis.

  • Mineralogy, Morphology, and Emplacement History of the Maaz Formation on the Jezero Crater Floor From Orbital and Rover Observations

    • The first samples collected by the Perseverance rover on the Mars 2020 mission were from the Maaz formation, a lava plain that covers most of the floor of Jezero crater. Laboratory analysis of these samples back on Earth would provide important constraints on the petrologic history, aqueous processes, and timing of key events in Jezero crater. However, interpreting these samples requires a detailed understanding of the emplacement and modification history of the Maaz formation. Here we synthesize rover and orbital remote sensing data to link outcrop-scale interpretations to the broader history of the crater, including Mastcam-Z mosaics and multispectral images, SuperCam chemistry and reflectance point spectra, Radar Imager for Mars' subsurface eXperiment ground penetrating radar, and orbital hyperspectral reflectance and high-resolution images. We show that the Maaz formation is composed of a series of distinct members corresponding to basaltic to basaltic-andesite lava flows. The members exhibit variable spectral signatures dominated by high-Ca pyroxene, Fe-bearing feldspar, and hematite, which can be tied directly to igneous grains and altered matrix in abrasion patches. Spectral variations correlate with morphological variations, from recessive layers that produce a regolith lag in lower Maaz, to weathered polygonally fractured paleosurfaces and crater-retaining massive blocky hummocks in upper Maaz. The Maaz members were likely separated by one or more extended periods of time, and were subjected to variable erosion, burial, exhumation, weathering, and tectonic modification. The two unique samples from the Maaz formation are representative of this diversity, and together will provide an important geochronological framework for the history of Jezero crater.

  • Strain-mediated ferromagnetism and low-field magnetic reversal in Co doped monolayer WS2

    • Strain-mediated magnetism in 2D materials and dilute magnetic semiconductors hold multifunctional applications for future nano-electronics. Herein, First principles calculations are employed to study the influence of biaxial strain on the magnetic properties of Co-doped monolayer WS2. The non-magnetic WS2 shows ferromagnetic signature upon Co doping due to spin polarization, which is further improved at low compressive (-2 %) and tensile (+2 %) strains. From the PDOS and spin density analysis, the opposite magnetic ordering is found to be favourable under the application of compressive and tensile strains. The double exchange interaction and p-d hybridization mechanisms make Co-doped WS2 a potential host for magnetism. More importantly, the competition between exchange and crystal field splittings, i.e. (Delta(ex) > Delta(cfs)), of the Co-atom play pivotal roles in deciding the values of the magnetic moments under applied strain. Micromagnetic simulation reveals, the ferromagnetic behavior calculated from DFT exhibits low-field magnetic reversal (190 Oe). Moreover, the spins of Co-doped WS2 are slightly tilted from the easy axis orientations showing slanted ferromagnetic hysteresis loop. The ferromagnetic nature of Co-doped WS2 suppresses beyond +/- 2 strain, which is reflected in terms of decrease in the coercivity in the micromagnetic simulation. The understanding of low-field magnetic reversal and spin orientations in Co-doped WS2 may pave the way for next-generation spintronics and straintronics applications.
