  • Development and psychometric properties of the Swedish ALPS-Neo pain and stress assessment scale for newborn infants

    • AIM: To validate and evaluate the psychometric properties of the ALPS-Neo, a new pain assessment scale created for the continuous evaluation of pain and stress in preterm and sick term infants.METHODS: A unidimensional scale for continuous pain, Astrid Lindgren Children's Hospital Pain Scale (ALPS 1), was developed further to assess continuous pain and stress in infants treated in the neonatal intensive care unit (NICU). The pain scale includes observations of five behaviours. A manual was created, clarifying the scoring criteria. An internal and an external panel assessed face validity. Psychometric properties were evaluated in three different steps. Inter-rater reliability was estimated from video-based assessments (n = 625) using weighted kappa statistics (test I). Inter-rater reliability was further evaluated in test II (n = 125) and test III (n = 96) by real-time assessments using the intraclass correlation coefficient (ICC) and Cronbach's alpha.RESULTS: The final inter-rater reliability (test III) was assessed as good with ICC 0.91 for the total score and 0.62-0.81 for the five items. Cronbach's alpha showed 0.95 for the total score.CONCLUSION: ALPS-Neo is a new assessment tool for optimising the management of pain and stress in newborn infants in the NICU. It has proved easy to implement and user-friendly, permitting fast, reliable observations with high inter-rater reliability.

  • A field experiment on the impact of sounds from a jet-and-basin fountain on soundscape quality in an urban park

    • A field experiment was conducted to explore whether water sounds from a fountain had a positive impact on soundscape quality in a downtown park. In total, 405 visitors were recruited to answer a questionnaire on how they perceived the park, including its acoustic environment. Meanwhile the fountain was turned on or off, at irregular hours. Water sounds from the fountain were not directly associated with ratings of soundscape quality. Rather, the predictors of soundscape quality were the variables Road-traffic noise and Other natural sounds. The former had a negative and the latter a positive impact. However, water sounds may have had an indirect impact on soundscape quality by affecting the audibility of road-traffic and natural sounds. The present results, obtained in situ, agree with previous results in soundscape research that the sounds perceived particularly roadtraffic and natural sounds explain soundscape quality. They also agree with the results from laboratory studies that water sounds may mask roadtraffic sounds, but that this is not simple and straight forward. Thus sound should be brought into the design scheme when introducing water features in urban open spaces, and their environmental impact must be thoroughly assessed empirically.

  • The Sound Labyrinth Project: Catalyst for Creative Activity

    • Two large sound installations, developed by transdisciplinary sound art and research group Urban Sound Institute, created a meeting place for one year of artistic and pedagogic activity in a Swedish regional museum. The project involved a historic sound archive, a string quartet, a local radio station, pedagogic workshops, several schools, children’s groups and musical education programs. The installations created a complexity of interior spaces, and acted as huge musical instruments to be ‘played’ by professional musicians, dancers and visitors. Through advanced computer programming and careful composition, modeling and distribution of sounds as words, narratives, music, space, bodily experiences and memory, the Sound Labyrinth allowed for great variations, durability over time, and different forms of interaction. The article describes the project, and discusses the exhibition as a platform for collective, multiple interaction; as an expanded musical-architectonic composition; and as a contribution to artistic research methodologies relevant for sound spaces within so-called sound-making disciplines.

  • Comparative metabolomics of muscle interstitium fluid in human trapezius myalgia: an in vivo microdialysis study

    • The mechanisms behind trapezius myalgia are unclear. Many hypotheses have been presented suggesting an altered metabolism in the muscle. Here, muscle microdialysate from healthy and myalgic muscle is analysed using metabolomics. Metabolomics analyse a vast number of metabolites, enabling a comprehensive explorative screening of the cellular processes in the muscle. less thanbrgreater than less thanbrgreater thanMicrodialysate samples were obtained from the shoulder muscle of healthy and myalgic subjects that performed a work and stress test. Samples from the baseline period and from the recovery period were analysed using gas chromatography-mass spectrometry (GC-MS) together with multivariate analysis to detect differences in extracellular content of metabolites between groups. Systematic differences in metabolites between groups were identified using multivariate analysis and orthogonal partial least square discriminate analysis (OPLS-DA). A complementary Mann-Whitney U test of group difference in individual metabolites was also performed. less thanbrgreater than less thanbrgreater thanA large number of metabolites were detected and identified in this screening study. At baseline, no systematic differences between groups were observed according to the OPLS-DA. However, two metabolites, l-leucine and pyroglutamic acid, were significantly more abundant in the myalgic muscle compared to the healthy muscle. In the recovery period, systematic difference in metabolites between the groups was observed according to the OPLS-DA. The groups differed in amino acids, fatty acids and carbohydrates. Myristic acid and putrescine were significantly more abundant and beta-d-glucopyranose was significantly less abundant in the myalgic muscle. less thanbrgreater than less thanbrgreater thanThis study provides important information regarding the metabolite content, thereby presenting new clues regarding the pathophysiology of the myalgic muscle.

  • Multivariate Modeling of Proteins Related to Trapezius Myalgia, a Comparative Study of Female Cleaners with or without Pain

    • The prevalence of chronic trapezius myalgia is high in women with high exposure to awkward working positions, repetitive movements and movements with high precision demands. The mechanisms behind chronic trapezius myalgia are not fully understood. The purpose of this study was to explore the differences in protein content between healthy and myalgic trapezius muscle using proteomics. Muscle biopsies from 12 female cleaners with work-related trapezius myalgia and 12 pain free female cleaners were obtained from the descending part of the trapezius. Proteins were separated with two-dimensional differential gel electrophoresis (2D-DIGE) and selected proteins were identified with mass spectrometry. In order to discriminate the two groups, quantified proteins were fitted to a multivariate analysis: partial least square discriminate analysis. The model separated 28 unique proteins which were related to glycolysis, the tricaboxylic acid cycle, to the contractile apparatus, the cytoskeleton and to acute response proteins. The results suggest altered metabolism, a higher abundance of proteins related to inflammation in myalgic cleaners compared to healthy, and a possible alteration of the contractile apparatus. This explorative proteomic screening of proteins related to chronic pain in the trapezius muscle provides new important aspects of the pathophysiology behind chronic trapezius myalgia.

    • Syftet med denna rapport är att illustrera hur ett praktiskt inriktat ramverk, tekno- logiska innovationssystem (TIS), kan användas av analytiker och beslutsfattare vid departement och myndigheter för att analysera strategiskt viktiga teknikområden ?????????????????????????????????????????????????????????????????????????????I rapporten analyseras fem TIS centrerade kring havsbaserad vindkraft, marin energi, ????????????????????????????????????????????????????????????????????????????????????? systemsvagheter som bromsar områdets vidare utveckling, vilka som kan åtgärdas av systemets aktörer och vilka som motiverar särskilda politiska åtaganden. Rapporten utgör därmed ett underlag för att formulera åtgärder för att åstadkomma ökad innova- tion, teknikspridning och industrialisering inom ovan nämnda teknikområden.Studien har även möjliggjort en jämförande analys av likheter och skillnader ???????????????????????????????????????????????????????????????????????????????? ?????????????????????????????????????????????????????????????????????????????????????? mellan områdena – de är starka respektive svaga av olika orsaker. Detta visar att ???????????????????????????????????????????????????????????????????????Samtidigt har områdena gemensamma drag.   • Inactivation of Pmel Alters Melanosome Shape But Has Only a Subtle Effect on Visible Pigmentation

    • PMEL is an amyloidogenic protein that appears to be exclusively expressed in pigment cells and forms intralumenal fibrils within early stage melanosomes upon which eumelanins deposit in later stages. PMEL is well conserved among vertebrates, and allelic variants in several species are associated with reduced levels of eumelanin in epidermal tissues. However, in most of these cases it is not clear whether the allelic variants reflect gain-of-function or loss-of-function, and no complete PMEL loss-of-function has been reported in a mammal. Here, we have created a mouse line in which the Pmel gene has been inactivated (Pmel(-/-)). These mice are fully viable, fertile, and display no obvious developmental defects. Melanosomes within Pmel(-/-) melanocytes are spherical in contrast to the oblong shape present in wild-type animals. This feature was documented in primary cultures of skin-derived melanocytes as well as in retinal pigment epithelium cells and in uveal melanocytes. Inactivation of Pmel has only a mild effect on the coat color phenotype in four different genetic backgrounds, with the clearest effect in mice also carrying the brown/Tyrp1 mutation. This phenotype, which is similar to that observed with the spontaneous silver mutation in mice, strongly suggests that other previously described alleles in vertebrates with more striking effects on pigmentation are dominant-negative mutations. Despite a mild effect on visible pigmentation, inactivation of Pmel led to a substantial reduction in eumelanin content in hair, which demonstrates that PMEL has a critical role for maintaining efficient epidermal pigmentation.

    • Noise is steadily on the rise in urban settings, creating a potential health hazard as well as being a nuisance. In major European cities, noise levels are so high that the majority of urban parks can no longer truly serve as recreational environments, a problem the WHO and the EU are attempting to address. This study explores various strategies that promotethe sustainable development of urban soundscapes at locations meant for rest, recreation, and social interaction. How are people affected by the combined effects of traffic and nature sounds in urban parks? To this end, we adopted a new track – the use of interdisciplinarymethodology – bringing together architectural analysis and artistic experiments, along with psychoacoustic methodology to evaluate aesthetic, emotional, perceptual, and spatial effects. A large-scale case study was conducted at a city park to explore if and how subjects are affected by purposely distributed sounds. The working hypothesis was that it is possible to cancel out traffic noise by affecting aural perceptions using a process known as informational masking. Our long-term objective is to create a scientific foundation for action plans, both pre-emptive and trouble-shooting, targeting parks and other similar public spaces that provide a relaxing environment.
