    • Comparative analysis of multiple genomes in a phylogenetic framework dramatically improves the precision and sensitivity of evolutionary inference, producing more robust results than single-genome analyses can provide. The genomes of 12 Drosophila species, ten of which are presented here for the first time (sechellia, simulans, yakuba, erecta, ananassae, persimilis, willistoni, mojavensis, virilis and grimshawi), illustrate how rates and patterns of sequence divergence across taxa can illuminate evolutionary processes on a genomic scale. These genome sequences augment the formidable genetic tools that have made Drosophila melanogaster a pre-eminent model for animal genetics, and will further catalyse fundamental research on mechanisms of development, cell biology, genetics, disease, neurobiology, behaviour, physiology and evolution. Despite remarkable similarities among these Drosophila species, we identified many putatively non-neutral changes in protein-coding genes, non-coding RNA genes, and cis-regulatory regions. These may prove to underlie differences in the ecology and behaviour of these diverse species.

  • Length-scale effects on damage development in tensile loading of glass-sphere filled epoxy

    • Particle-reinforced polymers are widely used in load-carrying applications. The effect of particle size on damage development in the polymer is still relatively unexplored. In this study, the effect of glass-sphere size on the damage development in tensile loaded epoxy has been investigated. The diameter of the glass spheres ranged from approximately 0.5-50 mu m. The first type of damage observed was debonding at the sphere poles, which subsequently grew along the interface between the glass spheres and epoxy matrix. These cracks were observed to kink out into the matrix in the radial direction perpendicular to the applied load. The debonding stresses increased with decreasing sphere diameter, whereas the length to diameter ratio of the resulting matrix cracks increased with increasing sphere diameter. These effects could not be explained by elastic stress analysis and linear-elastic fracture mechanics. Possible explanations are that a thin interphase shell may form in the epoxy close to the glass spheres, and that there is a length-scale effect in the yield process which depends on the strain gradients. Cohesive fracture processes can contribute to the influence of sphere size on matrix-crack length. Better knowledge on these underlying size-dependent mechanisms that control damage development in polymers and polymer composites is useful in development of stronger materials. From a methodology point of view, the glass-sphere composite test can be used as an alternative technique (although still in a qualitative way) to hardness vs. indentation depth to quantify length-scale effects in inelastic deformation of polymers.

    • Manufacturing competitiveness highly depends on companies' ability to rapidly reconfigure their assembly systems. This paper introduces the concept of assembly system proactivity based on interrelated levels of human involvement in a planed way will contribute to increased system ability to proactively address predicted and unpredicted events. Correct involvement of human operators will utilize the full combined potential of human and technical capabilities, also providing cost-efficient assembly system solutions. The ProAct (project presented) will develop proactive assembly system models, evaluating proactive, feature-based solutions. Focus is on realizing the potential of semi-automated system with relevant human involvement, i.e. operators with high skills adding e.g. flexible capability and functionality.

    • Phytophthora infestans is the most destructive pathogen of potato and a model organism for the oomycetes, a distinct lineage of fungus-like eukaryotes that are related to organisms such as brown algae and diatoms. As the agent of the Irish potato famine in the mid-nineteenth century, P. infestans has had a tremendous effect on human history, resulting in famine and population displacement(1). To this day, it affects world agriculture by causing the most destructive disease of potato, the fourth largest food crop and a critical alternative to the major cereal crops for feeding the world's population(1). Current annual worldwide potato crop losses due to late blight are conservatively estimated at $6.7 billion(2). Management of this devastating pathogen is challenged by its remarkable speed of adaptation to control strategies such as genetically resistant cultivars(3,4). Here we report the sequence of the P. infestans genome, which at similar to 240 megabases (Mb) is by far the largest and most complex genome sequenced so far in the chromalveolates. Its expansion results from a proliferation of repetitive DNA accounting for similar to 74% of the genome. Comparison with two other Phytophthora genomes showed rapid turnover and extensive expansion of specific families of secreted disease effector proteins, including many genes that are induced during infection or are predicted to have activities that alter host physiology. These fast-evolving effector genes are localized to highly dynamic and expanded regions of the P. infestans genome. This probably plays a crucial part in the rapid adaptability of the pathogen to host plants and underpins its evolutionary potential.

  • Ligand specificity of the anaphylatoxin C5L2 receptor and its regulation on myeloid and epithelial cell lines

    • During complement activation the pro-inflammatory anaphylatoxins C3a and C5a are generated, which interact with the C3a receptor and C5a receptor (CD88), respectively. C5a and its degradation product C5a-des-Arg(74) also bind to the C5a receptor-like 2 (C5L2). C3a and C3a-des-Arg(77), also called acylation-stimulating protein, augment triglyceride synthesis and glucose uptake in adipocytes and skin fibroblasts. Based on data obtained using transfected HEK293 and RBL cells, C5L2 is additionally proposed as a functional receptor for C3a and C3a-des-Arg(77). Here we use (125)I-ligand binding assays and flow cytometry with fluorescently labeled ligands to demonstrate that neither C3a nor C3a-des-Arg(77) binds to C5L2. C5L2 expression and its regulation are investigated on various cell lines by a novel C5L2-restricted binding assay and quantitative real time PCR. Dibutyryl cAMP and interferon-gamma induce up-regulation of this receptor on myeloblastic cell lines (U937 and HL-60), whereas tumor necrosis factor-alpha (TNF-alpha) has no effect. In contrast, epithelial HeLa cells are found to constitutively express C5L2 but not the C5a receptor. In HeLa cells, interferon-gamma and TNF-alpha drastically reduce C5L2 expression. No C5a-dependent Ca(2+) signaling is observed even in these cells endogenously expressing C5L2. Taken together, C5L2 is not a receptor for C3a or C3a-des-Arg(77). Thus, this receptor is unlikely to be directly involved in lipid metabolism. Instead, the identification of stimuli modifying C5L2 expression indicates that C5L2 is a highly regulated scavenger receptor for C5a and C5a-des-Arg(74).

  • Detritivory: stoichiometry of a neglected trophic level.

    • Previous syntheses have identified the key roles that phylogeny, body size, and trophic level play in determining arthropod stoichiometry. To date, however, detritivores have been largely omitted from such syntheses, despite their importance in nutrient cycling, biodiversity, and food web interactions. Here, we report on a compiled database of the allometry and nutritional stoichiometry (N and P) of detritivorous arthropods. Overall, both N and P content for detritivores varied among major phylogenetic lineages. Detritivore N content was similar to the N content of herbivores, but below that of predators. By contrast, detritivore P content was independent of trophic level. Contrary to previous reports, neither nutrient varied with body size. This analysis places detritivores in the context of related herbivores and predators, and as such, sets the stage for future investigations into the causes and consequences of elemental (mis)matches between detritivores and their detrital resources.
