    • Chlorophyll biosynthesis is initiated by magnesium chelatase, an enzyme composed of three proteins, which catalyzes the insertion of Mg2+ into protoporphyrin IX to produce Mg-protoporphyrin IX. In barley (Hordeum vulgare L.) the three proteins are encoded by Xantha-f, Xantha-g and Xantha-h. Two of the gene products, XanH and XanG, belong to the structurally conserved family of AAA+ proteins (ATPases associated with various cellular activities) and form a complex involving six subunits of each protein. The complex functions as an ATP-fueled motor of the magnesium chelatase that uses XanF as substrate, which is the catalytic subunit responsible for the insertion of Mg2+ into protoporphyrin IX. Previous studies have shown that semi-dominant Xantha-h mutations result in non-functional XanH subunits that participate in the formation of inactive AAA complexes. In the present study, we identify severe mutations in the barley mutants xantha-h.38, -h.56 and -h.57. A truncated form of the protein is seen in xantha-h.38, whereas no XanH is detected in xantha-h.56 and -h.57. Heterozygous mutants show a reduction in chlorophyll content by 14-18% suggesting a slight semi-dominance of xantha-h.38, -h.56 and -h.57, which otherwise have been regarded as recessive mutations

  • Association Between TAS2R38 Gene Polymorphisms and Colorectal Cancer Risk: A Case-Control Study in Two Independent Populations of Caucasian Origin

    • Molecular sensing in the lingual mucosa and in the gastro-intestinal tract play a role in the detection of ingested harmful drugs and toxins. Therefore, genetic polymorphisms affecting the capability of initiating these responses may be critical for the subsequent efficiency of avoiding and/or eliminating possible threats to the organism. By using a tagging approach in the region of Taste Receptor 2R38 (TAS2R38) gene, we investigated all the common genetic variation of this gene region in relation to colorectal cancer risk with a case-control study in a German population (709 controls and 602 cases) and in a Czech population (623 controls and 601 cases). We found that there were no significant associations between individual SNPs of the TAS2R38 gene and colorectal cancer in the Czech or in the German population, nor in the joint analysis. However, when we analyzed the diplotypes and the phenotypes we found that the non-taster group had an increased risk of colorectal cancer in comparison to the taster group. This association was borderline significant in the Czech population, (OR = 1.28, 95% CI 0.99-1.67; P-value = 0.058) and statistically significant in the German population (OR = 1.36, 95% CI 1.06-1.75; P-value = 0.016) and in the joint analysis (OR = 1.34, 95% CI 1.12-1.61; P-value = 0.001). In conclusion, we found a suggestive association between the human bitter tasting phenotype and the risk of CRC in two different populations of Caucasian origin.

  • Challenging claims in the study of migratory birds and climate change.

    • Recent shifts in phenology in response to climate change are well established but often poorly understood. Many animals integrate climate change across a spatially and temporally dispersed annual life cycle, and effects are modulated by ecological interactions, evolutionary change and endogenous control mechanisms. Here we assess and discuss key statements emerging from the rapidly developing study of changing spring phenology in migratory birds. These well-studied organisms have been instrumental for understanding climate-change effects, but research is developing rapidly and there is a need to attack the big issues rather than risking affirmative science. Although we agree poorly on the support for most claims, agreement regarding the knowledge basis enables consensus regarding broad patterns and likely causes. Empirical data needed for disentangling mechanisms are still scarce, and consequences at a population level and on community composition remain unclear. With increasing knowledge, the overall support ('consensus view') for a claim increased and between-researcher variability in support ('expert opinions') decreased, indicating the importance of assessing and communicating the knowledge basis. A proper integration across biological disciplines seems essential for the field's transition from affirming patterns to understanding mechanisms and making robust predictions regarding future consequences of shifting phenologies.

  • Macroalgal community dynamics on coral reefs : Implications for management

    • Although rather inconspicuous on healthy coral reefs, macroalgae form the basis of coral food webs. Today, macroalgae are generally increasing and many reefs undergo transitions from coral to macroalgal dominance resulting from e.g. enhanced nutrient loading or increased fishing.This thesis aims to investigate the relative importance of top-down and bottom-up regulation, and different herbivore types, on macroalgal distribution, fecundity and community composition on coral reefs. Papers I and II indicate that macroalgal abundance in a coral reef system is largely governed by top-down regulation through grazing by herbivores, while bottom-up regulation through enhanced nutrient availability rather influence algal species composition. Paper II also shows that these regulating effects are not as evident in an area with relatively strong water motion, suggesting that impacts of anthropogenic disturbance may be site-specific. Paper III shows that herbivory is an important factor influencing macroalgal growth and subsequent reproduction. Furthermore, Paper IV and V conclude that efficiency in removing macroalgal biomass is dependent on the type of dominant herbivore, where sea urchins seem to be more effective than fish. Paper IV indicates a seasonal variation in macroalgal biomass and distribution in a small geographic scale but with relatively high temporal resolution. Paper V on the other hand shows these same effects, but with a focus on geographic variation, including a large part of the East African region, as well as between year temporal variations in Kenya. Together, results from the two latter studies indicate that herbivory by fish may not be able to prevent a macroalgal bloom in a degraded system where substrate availability for algal colonization is high, but that it may still facilitate coral recovery over time. Thus, a large algal biomass may not necessarily indicate a reef beyond the possibility of recovery.
